Tomorrow, Friday, Oct. 18, we will be hosting three of our young BME alums as our inaugural 38X38 College of Engineering Honorees for a panel session in MJIS 1001 at 10:30 am. What are the 38X38 Awards? Neil Armstrong was 38 years old when he became the first person to set foot on the moon. Inspired by his legacy, the inaugural #PurdueEngineering #38by38 award celebrates 38 alumni 38 years old and younger who have swiftly risen to the top of their fields. This group of BME alumni is impressive, embodying the #Boilermaker spirit through hard work, determination, innovation, leadership and impact. Each of them has achieved career milestones in record time, from starting their own successful businesses to holding high-ranking positions. This is a great opportunity for students and faculty to engage with our amazing alumni. Students will delve into the composition and function of the gut microbiome, understanding how diet influences microbial diversity and activity. Key topics include the role of the gut microbiome in nutrient digestion and absorption, metabolic processes, and its involvement in various diseases in which microbial imbalance has an etiological link. Students will also become familiar with current analytical and bioinformatic methods for studying the gut microbiome and the potential for prebiotic and probiotic interventions to prevent or manage diseases. This new health-related initiative is launching with support from the Life and Health Sciences Summit that was supported by the Office of Research and the Provost’s Office, and includes researchers from HHS, Engineering, and Science. Our new center will provide hearing-health education and outreach to the Indianapolis community and beyond, while recruiting subjects with diverse racial, socioeconomic, and hearing-health backgrounds with the goal of using the data to improve precision audiology diagnostics through ML and data analytics. Hi Boilermakers, We wanted to reach out about an exciting professional development opportunity. Tolmar, a fully integrated pharmaceutical company in northern Colorado, will be on site October 29th. We are recruiting for our Scientific Leadership Development Program (SLDP). This is a unique rotational program that helps recent PhD and PharmD graduates develop their leadership skills and broaden their understanding of pharmaceutical development. Please join us for a presentation followed by lunch in the Weldon School MJIS 2001 from 12:30-1:20 pm on October 29th to learn more. All years are welcome to come and learn more. Meyers Abstract: Cardiovascular diagnostics are crucial for effective treatment and management of critical cardiac conditions, yet current methods often lack the accuracy and clinical relevance needed to make timely and informed decisions. Despite advancements in computational techniques, gaps remain in utilizing multimodal data from electrocardiograms (ECGs), echocardiograms, and magnetic resonance imaging (MRI) for improving diagnostics. This study aims to address this gap by developing novel methodologies that integrate machine learning, signal and image processing, and biomechanical analysis. Our research focuses on four key areas. First, we aim to improve the detection of Atrial Fibrillation (AFib), a common heart rhythm disorder, by developing an interpretable and efficient feature extraction method from ECG data (Aim 1). By applying advanced algorithms like XGBoost (eXtreme Gradient Boosting), we expect to enhance AFib detection in real-time, even in shorter ECG recordings. Second, we are developing a hybrid framework to automate the segmentation of cardiac structures, such as the left ventricle (LV), from echocardiograms (Aim 2). This approach combines traditional image processing with deep learning, improving the accuracy and consistency of clinical measurements like ejection fraction. Third, we propose a novel fusion method that integrates Color Doppler echocardiography with 4D Flow MRI (Aim 3). This technique will enhance the accuracy of blood flow velocity measurements, providing a more complete understanding of cardiovascular flow dynamics—especially important in complex heart conditions. Lastly, we will investigate the biomechanical alterations in the Systemic Right Ventricle (SRV) during the critical transition from fetal to neonatal stages (Aim 4). By introducing advanced hemodynamic markers such as energy loss and vortex strength, this study seeks to offer new insights into congenital heart defects, particularly Hypoplastic Left Heart Syndrome (HLHS). Combinedly, these aims will significantly advance cardiovascular diagnostics by developing clinically interpretable, automated tools for early and accurate detection of cardiac abnormalities. Roudebush VA Medical Center Abstract: Maxillofacial and oral defects originate from congenital conditions such as cleft palate, diseases such as osteosarcoma, and injuries from blasts or vehicular accidents. These defects lead to complications for the patient, including difficulty speaking, infections, as well as damaging psychological effects owing to the patient’s physical appearance. The current standard of care uses particulates of freeze-dried auto- or allograft bone covered by a titanium mesh secured in place by screws. This approach is limited by: 1) graft variability; 2) duration of the surgery; and 3) overpacking of the bone particulates, often leading to diminished bone regeneration as blood vessels may fail to form. Prior research has demonstrated the benefits of using 3D printed titanium covers that protect the particulate core. The patient specific design reduces surgical times and has more reproducible protective features than meshes fitted during surgery. However, these covers lead to an increased degree of stress shielding, since 3D printed titanium covers are thicker than the current titanium meshes on the market. Additionally, this does not address the issues that can occur with an overpacked core. To address these fallbacks, we designed a 3D printable, biodegradable, and implantable device with patient-specific shape and a porous core-cover structure. We hypothesized that a 3D printed porous cover-core bone graft, with controlled macro- and micro-porosities, would enhance infiltration and osteointegration. Finite element analysis and in vitro modeling were used to guide the design of both the cover and core components. Our results demonstrated that a polycaprolactone (PCL) cover with 1 mm pores, secured with buccal screws minimized stress shielding while providing stresses within a range that would promote osteogenesis. Additionally, we developed a hybrid core composed of methacrylated alginate, methacrylated gelatin (AlgGelMa), and tricalcium phosphate (TCP), which provided elastic properties within the range of the FEA model, promoted cell infiltration, and supported growth factor sequestration. UCLA Abstract: The human hippocampus is necessary for episodic memory, i.e. memory of specific events experienced, the standard clinical test of Alzheimer's etc. On the other hand, the rodent hippocampus is believed to be an abstract cognitive map of space supported by place cells. Hence, spatial memory, e.g. water-maze escape, is the classic test of hippocampal function in mice. However, place cells are rare in primates. Instead, primate hippocampal neurons encode specific visual cues, e.g. faces. How can these major differences across species be reconciled? Further, while many drugs have cured hippocampal disorders, e.g. ADRD, in mice, all of them have failed in humans. We hypothesize that these two puzzles are related, and they arise from fundamental differences in the multisensory experiences of mice and primates. Indeed, when rodents are asked to rely only on vision, like primates, using a noninvasive virtual reality, most of the rodent-primate hippocampal differences vanish. This approach also provides early insights about how visual stimuli are transformed from V1 to CA1 to episodic memory. The results would help understand the functioning of neural deep-network works, and for reliable translation of memory therapies from mice to humans. Bio: Mayank Mehta is a professor in the department of Physics & Astronomy, Neurology, and Electrical and Communications engineering (ECE) at UCLA. He is the director of Keck Center for Neurophysics and of the Center for Physics of Life. During his PhD he investigated the effects of quantum fluctuations on the structure space-time. His subsequent research has focused on understanding how the brain creates abstract ideas such as abstract space-time and events. To test these theories, his laboratory has developed novel experimental tools such as an immersive VR for rodents, and a novel electrode called Dendrode that can measure the membrane potential of dendrites in freely behaving animals. Using these, their lab measures the activities of many individual neurons simultaneously while the subjects are learning and during the subsequent sleep. Using these they have uncovered novel mechanisms of learning and abstraction. Signal processing and robustness of the BMP network during zebrafish embryogenesis Nissa Larson (David Umulis, advisor) [cid:image003.jpg@01DB1E1C.B59AA4D0] Abstract: In developing tissues, signal transduction from morphogen gradients conveys positional information to cells, resulting in cell specification and differentiation. One such morphogen is bone morphogenetic protein (BMP), of the TGF-β superfamily, whose network is highly conserved across many species. In zebrafish species Danio rerio, this signaling pathway directs dorsoventral axis formation during early embryogenesis. Many of the molecules that play a role in this network are known; however, the mechanisms through which they achieve noise attenuation and gradient robustness have not been defined. Specifically, the heterodimer heterotetramer complex has been shown to be required for signal transduction, but deterministic modeling of the BMP membrane receptors at this stage has not given any insight into evolutionary drivers of the requirement. Building and developing a stochastic, multiscale model of this process will allow us to mechanistically assess zebrafish phenotype variability related to the distributions of noise and stochasticity. We can also analyze time-dependent signaling and frequency metrics that are not available in traditional, deterministic modeling. Fast Fourier Transform and cumulative energy spectral density visualization show that the heterodimer-heterotetramer complex may function as a low-pass filter in the dorsal-ventral axis formation process, specifically tuned to the noise of the system. To further probe this system, we are working to develop an experimental optogenetics protocol to genetically manipulate dorsoventral signal transduction in vivo and collect and analyze fluorescence intensity of the downstream pSmad gradient. Through these multiscale modeling efforts and experimental coupling, we hope to further understand the noise origins and signal processing of this network. As the BMP signaling pathway is highly conserved and has been implicated in human bone growth and wound healing, its study in simpler systems such as zebrafish stands to accelerate our comprehension of BMP network structure and molecular mechanisms with application in regenerative medical studies. Learning and integration of artificial and natural sensory stimuli Samuel Senneka (Maria Dadarlat Makin/Ed Bartlett, advisors) [cid:image005.jpg@01DB1E1C.B59AA4D0] Abstract: Our senses are the primary channels through which we collect information about the world around us, and our brains constantly assess similarities and differences in sensory signals through a host of functions collectively known as multisensory processing. Losing one or multiple sensory modalities has a drastic impact on our ability to interact with our environment and how information from remaining sensory modalities is processed. Devices aimed at restoring lost sensation are already being developed, however, these devices have a wide variability in patient outcomes and are not effective therapies for all forms of sensory loss pathologies. Recent methods using intracortical microstimulation (ICMS) have shown that many animal models can use ICMS as a form of sensory feedback but whether this feedback unifies with existing perception is unclear and the most efficacious methods for learning novel sensory modalities have yet to be identified. To further study ICMS as a form of sensory feedback, mice were trained to navigate to an arbitrary region of their training cage indicated by a visual target and closed-loop ICMS feedback. Probe trials were introduced to assess subjects' understanding of the ICMS signal and compare their performance against natural sensation. Subjects were able to learn this task to a high degree of accuracy in a very short time and performance with ICMS rivaled performance with a dim visual target. Metrics including success rate, average speed, time taken, and path efficiency also showed a significantly higher performance on combined visuo-ICMS trials than unimodal ICMS or visual trials indicating integration of the two signals. In future work, this behavior will allow the testing of a wide variety of ICMS parameters and interactions with natural sensory modalities to develop a stronger understanding of the brain's ability to learn novel sensory modalities and how new sensations interact with existing ones.
