The cell cortex, consisting of actin and microtubule cytoskeletons, plays a critical role in the spatiotemporal regulation of cell morphogenesis. Remodeling of the cytoskeleton is responsible for a large fraction of morphogenetic processes. However, due to experimental limitations, the underlying mechanism of the remodeling of the cortical cytoskeleton remains elusive. These limitations can be overcome by computational models. We computationally investigated two systems: the actin cytoskeleton in animal cells and cortical microtubule arrays in plant cells. For the actin cytoskeleton, we implemented a well-established agent-based model consisting of inter-connected cylindrical segments representing actin filaments, actin crosslinking proteins, and molecular motors. We found that the active remodeling of actin networks is regulated by network connectivity as well as the dynamic behaviors of crosslinking proteins and motors. In addition, we demonstrated that the fragmentation (i.e., severing) and turnover of actin filaments mediate various clustering behaviors due to a balance between force generation and force relaxation. For microtubule arrays, we developed a coarse-grained model of microtubules based on the Gillespie algorithm. Using the model, we studied the self-organization of the cortical microtubule arrays in response to tensile stress. By implementing a cellular stress pattern and a constitutive relationship between stress and plus-end dynamics of microtubules, we were able to identify the most sensitive parameters that constitute dynamic instability and recapitulate a distinct morphological transverse band. We further validated the importance of branched nucleation and severing during bundle formation and network evolution. Overall, our results regarding the two cytoskeleton systems provide insights into understanding the precise control of cytoskeletal remodeling at subcellular scales as well as integrative molecular mechanisms underlying cell morphogenesis. Without this important function, blood glucose becomes elevated, giving rise to the metabolic disorder diabetes. Therefore, it is unsurprising that β-cells are the focus of significant basic and translational research, including studies evaluating their potential as a therapeutic cell population. Unfortunately, measurement of β-cell function in the laboratory involves multi-step glucose-stimulated insulin secretion (GSIS) assays, followed by separate quantification of insulin by time-consuming and costly enzyme-linked immunosorbent assays (ELISAs). This often precludes researchers from performing dynamic GSIS measurements, which better represent β-cell physiology but are even more labor-intensive. Experimental microfluidic devices aim to reduce the burden of these dynamic measurements, however ELISAs are still required to process the many samples generated. Thus, there is a critical need for an easy-to-use, rapid, and low-cost alternative method of measuring insulin secretion in real-time. Additionally, most GSIS platforms keep β-cells in suspension, despite the importance of extracellular matrix mechanosignaling for their survival and function. To address this need, we propose to develop and validate an advanced microphysiological system that supports rapid functional assessment of β-cells. The proposed device will restore β-cells to a more physiological microenvironment through encapsulation in a fibrillar collagen scaffold and will integrate a custom electrochemical sensor to directly quantify insulin secretion. [Bmeroundtable-list] Upcoming Town Hall: community and climate

Dear Weldon School of Biomedical Engineering Community, During the spring semester of the previous academic year (2020-2021), we conducted a climate survey in the school. Diversity, Equity, Inclusion, and Access are critical for our program and evaluation of the survey results is one step in this process that informs where we are as a school and, importantly, identifies areas where we need to grow and do better as a school. First, in this email, please find attached a summary of the survey for review. Dr. Jackie Linnes, Dr. Hyowon Lee, and Dr. Deva Chan have prepared a summary of the data from the survey and also make recommendations. Over 200 members of the Weldon School provided their reflections, suggestions, and concerns in the anonymous 2020 Climate Survey. I’ve gone over all the data and responses as well and find that this summary is representative of the main themes and content of the survey. The summary is attached to this email. TOWN HALL DISCUSSION On Oct 29th from 1-2:30pm the Weldon School will be holding a Town Hall in MIIS 2001 and virtually via zoom link <https://purdue-edu.zoom.us/j/94084070089?pwd=cmRRWnRpSU9jUnNieEhaUitOcUZTQT…>to discuss the results of the 2021 Climate Survey and our current and future plans in the school. I strongly encourage you to participate and also acknowledge that entering into self reflection can be intimidating for many. During my interview for the Headship, one of my primary goals that I had the privilege to present and also discuss with faculty, students, and staff was to work on the School’s diversity, equity, and inclusion mission and climate. I believe the community is ready to hold this Town Hall conversation and to work together with empathy, grace, and intentionality so that we can each grow in our community and ensure everyone in the Weldon School feels a sense of belonging and wellbeing. The town hall is a big step in this direction. Participants will have an opportunity to ask questions about the climate survey results and engage in discussion about Weldon School DEI efforts and priorities both in person and virtually during the Town Hall. History of DEIA activities and survey in the Weldon School For those wondering about how we arrived at this point, below are some details from recent BME DEIA work: Purdue BME has had a Diversity and Inclusion Working Group (DIWG) led by Andrew Brightman for 4+ years now and the group has done a lot of work to uncover underlying needs at the Weldon School. DIWG held an open forum in July 2020 and found that more proactive School-wide DEI efforts were needed. In August 2020, the BME DEI Steering Committee was initiated with Jackie Linnes directing the efforts. The DEI Steering Committee commenced a number of initial actions including meetings with grad and undergrad students about their specific needs, beginning a Graduate and Postdoc Peer Mentoring Program, an Undergraduate Resource Center, holding Faculty and Staff and Grad DEI Journal Clubs, and hosting a High School Internship Program for minoritized students, as well as contracting with the Higher Education Data Sharing consortium to complete an anonymous Diversity and Equity Climate Survey (analysis attached). The survey goal was to obtain a baseline for our School and to determine where we need to grow. One thing in the climate survey was a need for more communication and transparency around current DEIA support, education, resources, and efforts. The Oct 29th Town Hall will enable discussions of the results of the climate survey, existing resources, and guide future actions and priorities by giving voice to the Weldon School community to provide advice and insights into the needs and implementation. Dear BME community, It’s hard to believe we are already into mid October and the semester. We will have a number of updates and announcements over the coming weeks, so please stay tuned! First, I want to let everyone know, that starting Jan. 1 2022, we will be adding a 5th research area to BME. Neuroengineering and Neurotechnology. There’s a lot of work to do in the meantime, and we will have a web presence for recruiting later this fall (BEFORE the Jan start date) to get the word out and it is expected that Jan. 1 is when we will have leadership in the area assigned and that will be the functional start date. I want to thank Ed Bartlett and members of the working group for soliciting input from internal and external stakeholders, identifying strengths and weaknesses for adding this research area, and formally making recommendations as the outcome of the working group. I’m excited to see this new area emerge that represents a strong cohort of faculty in the Weldon School! Neuroengineering and Neurotechnology Working to 1) decipher how the nervous system functions in normal and pathological conditions through the collaborative development of tools, experimental techniques, and analysis/modeling techniques at multiple levels of biological organization, spanning subcellular to network to behavioral scales and 2) apply understanding of the nervous system towards the development of devices, diagnostics, and treatments to improve human health. Grill, Ph.D. Edmund T. Pratt, Jr. School Distinguished Professor of Biomedical Engineering Duke University Abstract: Many neurological disorders, for example pain and spasticity, are characterized by undesirable increases in sensory, motor, or autonomic nerve activity. Local application of kilohertz frequency (KHF) alternating currents can block conduction of undesired hyperactivity through peripheral nerves and could be a therapeutic approach for alleviating disease symptoms. In contrast to electrical stimulation, our understanding of how KHF nerve block depends on waveform parameters is quite limited, and this lack of knowledge precludes optimization and application of this promising therapeutic approach. I will describe our recent work, combining computational modeling, in vivo experimental measurements, and engineering optimization, to quantify the effects of KHF waveform parameters in nerve conduction block. The outcomes provide fundamental insights into the interaction of waveform shape and block, the potential for selective block of specific nerve fiber types (diameters), and an approach to eliminate the undesirable initial burst of action potentials that occurs prior to achieving block, known as onset response. Collectively, the results advance the therapeutic utility of KHF nerve block. Bio: Warren M. Grill is the Edmund T. Pratt, Jr. School Distinguished Professor of Biomedical Engineering at Duke University. He received the B.S. in biomedical engineering in 1989 from Boston University and the Ph.D. in biomedical engineering in 1995 from Case Western Reserve University. Professor Grill teaches courses on circuits and instrumentation, bioelectricity, and the fundamentals and applications of electrical stimulation. He received the Capers & Marion McDonald Award for Excellence in Teaching and Research at Duke University on 2008 and again in 2018, in 2013 was awarded Outstanding Postdoc Mentor at Duke University, and in 2014 received the University Scholar/Teacher of the Year Award. His research interests are in neural engineering and neuromodulation and include design and testing of electrodes and stimulation techniques, the electrical properties of tissues and cells, and computational neuroscience with applications to restoration of bladder function, treatment of movement disorders with deep brain stimulation, electrical stimulation for treatment of pain, and vagus nerve stimulation for regulation of organ function. He has published over 220 peer reviewed journal articles and has been awarded 59 US patents. He is Co-Founder, Director, and Chief Scientific Officer of NDI Medical, a medical device incubator, Co-Founder, Director, and Chief Scientific Officer of DBI, which is commercializing a novel approach to brain stimulation for neurological disorders, and Chief Scientific Advisor at SPR Therapeutics, which has developed a novel therapy for treating pain. Dr. Grill serves as a Consultant to the Neurological Devices Panel of the FDA Medical Devices Advisory Committee, on the editorial boards of Brain Stimulation and Current Opinion in Biomedical Engineering, and is Deputy Editor for the Journal of Neural Engineering.
