Stretching the Limits: Unveiling Key Matrix Factors and Survival Mechanisms in the Metastatic Tumor Microenvironment Madison Howard (Luis Solorio, advisor) [cid:image005.jpg@01DB1BC2.3B1F6440] Abstract: Breast cancer (BC) often metastasizes to organs experiencing high mechanical stress, including the lungs. Despite this knowledge, the effect that dynamic forces native to the lungs have on early disseminated tumor cells is a currently under explored area of the metastatic cascade. Recent in vitro findings suggest BC cells enter a dormant state in response to tensile stress, yet the mechanisms by which these cells adapt to the dynamic conditions at metastatic sites remain unclear. In vitro cell stretching experiments and ex vivo tissue characterization could help uncover the mechanisms behind this mechanical adaptation, but the capabilities of existing testing platforms are limited. Here, we developed a simple and cost-effective in-plane actuation platform with high-resolution force sensing and simultaneous imaging capabilities. We will demonstrate the device utility for ex vivo tissue characterization by analyzing transient mechanical changes in murine metastatic lung tissue. Given that the extracellular matrix (ECM) is a main driver of tissue stiffness, this study will be paired with a proteomic analysis to identify transient changes in ECM composition throughout disease progression. To further investigate BC dormancy at the cellular level, an existing magnetic actuation platform will also be used to investigate how varying levels of strain energy affect the proliferation and mechanotransduction signaling of BC cells. Together, these aims will address gaps in current testing systems and provide insight into the mechanical adaptation mechanisms driving metastatic BC at both the tissue and cellular levels. >From Injury to Innervation: Investigating the spatiotemporal dynamics of CGRP in OA Rahaf Salim (Deva Chan/Matthew Ward, advisors) [A person wearing a head scarf Description automatically generated] Abstract: Calcitonin gene-related peptide (CGRP) is a neuropeptide expressed in sensory nerve fibers that innervate joint tissues, where it plays a significant role in mediating pain and inflammation. While CGRP's involvement in osteoarthritis (OA) pain is increasingly recognized, there remain significant gaps in our understanding of how CGRP-expressing nerve fibers change over time and space within the joint during disease progression. Most studies to date have relied on traditional histological sectioning, which restrict the ability to examine the complete architecture of joints and the intact distribution of neural networks that innervate them in a spatially accurate manner. This knowledge gap is particularly important in the context of post-traumatic osteoarthritis (PTOA), where joint degeneration and pain often progress rapidly. In this study, we utilize a non-invasive ACL rupture (ACLR) mouse model of PTOA to investigate how CGRP-expressing nerve fibers respond to joint injury and how their distribution and morphology evolve as the disease progresses. This research leverages advanced optical clearing techniques to render joint tissues transparent, allowing for the visualization and in-depth evaluation of CGRP-expressing nerve fibers in intact joint structures-an approach that contrasts with traditional histological sectioning. Furthermore, this study integrates assessments of bone remodeling and pain behavior to establish functional links between sensory nerve fiber dynamics and structural changes in the joint. Through this work, we aim to uncover critical insights about the interplay between CGRP expression and joint pathology, with the potential to guide the development of spatially targeted interventions. Dr. Longtin holds a dual appointment as an associate professor in the departments of Obstetrics and Gynecology and Communication Studies. Her research focuses on communication education and faculty development in the sciences and health professions. Dr. Longtin serves as the co-editor for the Public Library of Science's Science Communication blog and leads a graduate minor in Communicating Science at Indiana University Indianapolis. She has served as a consultant on communication and education projects for national and international organizations including the Indiana Department of Health and the European Science Media Hub. In 2016, Dr. Longtin was highlighted by the Indianapolis Business Journal as one of their Forty Under 40 young professionals to watch for her work using improvisational theatre to teach physicians and scientists to communicate more effectively. Prior to earning her graduate degrees, Dr. Longtin worked for Indiana Public Radio and The Weather Channel. She keeps her broadcaster voice alive by serving as the co-host of Intersections, a weekly public radio show where art and science meet. Dr. Longtin is not a native Hoosier, but she did the tour of Indiana schools, earning a bachelor's degree from Ball State University, a master's from Purdue University, and a PhD from Indiana University. On Friday, Oct. 18 we will be hosting three of our young BME alums as our inaugural 38X38 College of Engineering Honorees for a panel session in MJIS 1001 at 10:30 am. What are the 38X38 Awards? Neil Armstrong was 38 years old when he became the first person to set foot on the moon. Inspired by his legacy, the inaugural #PurdueEngineering #38by38 award celebrates 38 alumni 38 years old and younger who have swiftly risen to the top of their fields. This group of BME alumni is impressive, embodying the #Boilermaker spirit through hard work, determination, innovation, leadership and impact. [Bmeroundtable-list] REMINDER: Fall MJIS air handling shutdown

Our semi-annual air handling shutdown is scheduled for Tuesday, October 8th. As a reminder: the entire building's air handling system in MJIS will be turned off. ALL chemical hood activity, biosafety cabinet activity, autoclave work and activities in wet labs will need to STOP by 5 PM on Monday, October 7th. Any chemicals stored in the hoods will have to be sealed at that time. Signs will be posted on all hoods to remind users not to use the hoods Monday evening, and removed when the maintenance work is completed and normal operations can resume (some time on Tuesday). Please plan ahead - DO NOT use chemicals in the lab areas that you would normally use in the hoods. A model-guided approach to designing a biological controller for macrophage paralysis in sepsis Derrick Dankwa (Leo Green, advisor) [cid:image002.jpg@01DB1319.272507D0] Abstract: Sepsis, a life-threatening condition characterized by the body's extreme response to infection, affects millions globally and is a leading cause of death in intensive care units. A critical aspect of sepsis pathophysiology is macrophage paralysis, where these vital immune cells become dysfunctional, severely compromising the body's ability to fight infection. Despite advances in sepsis management, effective therapies targeting macrophage paralysis remain elusive. Therapeutic modulating macrophage populations has been proposed as a novel strategy to dampen inflammation in septic conditions. To inform the design of a novel class of immunotherapies that control the critical divergence of the immune response during septicemia, we must understand the intricate differences in immune cell dynamics and coordinated molecular signals between healthy and septic states. Here, we deployed an ordinary differential equation (ODE)‑based model to capture the hyper and hypo-inflammatory phases associated with sepsis. Our findings suggest that disrupted macrophage polarization results in excess monocytes, M1 and M2 macrophage phenotypes, leading to immune paralysis. Using our model-based analysis, we designed a proof-of-concept design of a biological controller that could effectively regulate macrophage dysregulation observed in sepsis. Our model describes a systems biology approach to predicting and exploring critical parameters as potential therapeutic targets that promise to transition sepsis-induced inflammation toward an improved healing state. AI Surveillance and On-Farm Diagnostics for Improved Livestock Farming Bibek Raut (Mohit Verma, advisor) [cid:image003.png@01DB1319.272507D0] Abstract: The rising global demand for animal-based products has increased pressure on animal farms to increase their productivity while maintaining high animal welfare standards. Concurrently, a shrinking pool of available labor and the specialized training required for effective in-person surveillance have led to less frequent animal health monitoring. Thus, there is a critical need for practical and deployable digital health and productivity technologies capable of providing real-time observations and analysis, to allow data-driven decision-making. To fill this gap, our study combines artificial intelligence (AI)-assisted surveillance with portable, on-farm nucleic acid-based diagnostics. The AI surveillance component is an intelligent sensor system that combines various video types with environmental sensors and provides insights into animal behaviors such as time spent feeding and drinking, number of aggressive incidents, as well as tracking body weight gains. This data-driven approach enables the detection of anomalies in behavior and environment, allowing farmers to focus on specific groups of animals that may require attention, thereby optimizing resource allocation and improving overall productivity. On the other hand, our portable nucleic acid-based tests employ loop-mediated isothermal amplification (LAMP) on microfluidic paper sensors, enabling rapid and cost-effective on-farm diagnostics and reducing the dependency on centralized facilities. Thus, by integrating continuous behavioral monitoring with advanced on-site diagnostics, the proposed system enhances decision-making processes, supports farm staff in their roles without replacing human expertise, and helps improve animal welfare and increase farm productivity.
