Hello everyone, Recently there have been several incidents involving tampering, theft, and violation of Weldon School security policies. I'm writing here to raise everyone's level of awareness as well as clearly indicate responsibility. * Within the past month there were research equipment thefts from laboratories. * Also, within the past month, there was clear tampering with core equipment that was most likely intentional and potentially damaging. * This weekend there were multiple instances of propping doors open after hours (please see picture below). This is a direct and egregious violation of building and university policy. [1000027122.jpg] The safety and security of our Weldon team members and the associated facilities and property is tantamount to our operation, mission, and goals. Therefore, there can be little or no tolerance for practices that jeopardize these. * Please recognize that involvement with the Weldon programs is a privilege and not a right. I encourage all who see something to bring it to my direct attention. * We have been given many amazing gifts that we must shepherd and protect. Further, we have too many valuable activities that support our collective mission to be inhibited by those not following policies. A model-guided approach to designing a biological controller for macrophage paralysis in sepsis Derrick Dankwa (Leo Green, advisor) [cid:image002.jpg@01DB1319.272507D0] Abstract: Sepsis, a life-threatening condition characterized by the body's extreme response to infection, affects millions globally and is a leading cause of death in intensive care units. A critical aspect of sepsis pathophysiology is macrophage paralysis, where these vital immune cells become dysfunctional, severely compromising the body's ability to fight infection. Despite advances in sepsis management, effective therapies targeting macrophage paralysis remain elusive. Therapeutic modulating macrophage populations has been proposed as a novel strategy to dampen inflammation in septic conditions. To inform the design of a novel class of immunotherapies that control the critical divergence of the immune response during septicemia, we must understand the intricate differences in immune cell dynamics and coordinated molecular signals between healthy and septic states. Here, we deployed an ordinary differential equation (ODE)‑based model to capture the hyper and hypo-inflammatory phases associated with sepsis. Our findings suggest that disrupted macrophage polarization results in excess monocytes, M1 and M2 macrophage phenotypes, leading to immune paralysis. Using our model-based analysis, we designed a proof-of-concept design of a biological controller that could effectively regulate macrophage dysregulation observed in sepsis. Our model describes a systems biology approach to predicting and exploring critical parameters as potential therapeutic targets that promise to transition sepsis-induced inflammation toward an improved healing state. AI Surveillance and On-Farm Diagnostics for Improved Livestock Farming Bibek Raut (Mohit Verma, advisor) [cid:image003.png@01DB1319.272507D0] Abstract: The rising global demand for animal-based products has increased pressure on animal farms to increase their productivity while maintaining high animal welfare standards. Concurrently, a shrinking pool of available labor and the specialized training required for effective in-person surveillance have led to less frequent animal health monitoring. Thus, there is a critical need for practical and deployable digital health and productivity technologies capable of providing real-time observations and analysis, to allow data-driven decision-making. To fill this gap, our study combines artificial intelligence (AI)-assisted surveillance with portable, on-farm nucleic acid-based diagnostics. The AI surveillance component is an intelligent sensor system that combines various video types with environmental sensors and provides insights into animal behaviors such as time spent feeding and drinking, number of aggressive incidents, as well as tracking body weight gains. This data-driven approach enables the detection of anomalies in behavior and environment, allowing farmers to focus on specific groups of animals that may require attention, thereby optimizing resource allocation and improving overall productivity. On the other hand, our portable nucleic acid-based tests employ loop-mediated isothermal amplification (LAMP) on microfluidic paper sensors, enabling rapid and cost-effective on-farm diagnostics and reducing the dependency on centralized facilities. Thus, by integrating continuous behavioral monitoring with advanced on-site diagnostics, the proposed system enhances decision-making processes, supports farm staff in their roles without replacing human expertise, and helps improve animal welfare and increase farm productivity. A service worker found this plant propping open an exterior door this (9/27) morning at 6:30am in the East Vestibule. We have no idea how long the building was not secured. Propping doors (either interior or exterior) is strictly prohibited by university policy. You are not only violating university policy, but are also putting everyone in the building at risk by allowing someone without approved access to enter the building. The safety and security of every BME faculty, staff and student is our utmost top priority. Never prop a door open, or open a locked door for someone you don't know! Krohn Assistant Professor of Biomedical Engineering Smart Materials & Robotics Lab, Mechanical & Materials Engineering, University of Nebraska-Lincoln Abstract: Biological systems exhibit a unique combination of intrinsic functionalities that seamlessly integrate sensory perception, coordinated movement, energy management, and self-repair mechanisms. Engineering soft-matter counterparts that emulate the mechanical compliance and versatility of these systems requires new classes of soft materials and manufacturing techniques. This work aims to extend the range of physical properties and capabilities in multifunctional systems towards bridging the gap between traditional rigid systems and biological counterparts. In this talk, I will present several new techniques to create multifunctional soft materials that exhibit embodied sensing, actuation, and energy harvesting functionalities. This includes progress in the development of novel material architectures that contain room temperature gallium-based liquid metal alloys for passive thermal management, mechanically robust circuity, and the ability to steer cracks. Next, I will present recent work on direct ink writing 3D printing strategies to control liquid metal microdroplet microstructure in-situ to program mechanical, electrical, and thermal behavior to create robust soft multifunctional systems. Lastly, I will describe efforts to create integrated soft material systems with a bioinspired nervous system to detect and manipulate diverse underwater objects. Bio: Dr. Eric Markvicka is the Robert F. and Myrna L. Krohn Assistant Professor of Biomedical Engineering in the Department of Mechanical and Materials Engineering at the University of Nebraska-Lincoln (UNL). There, he also holds a courtesy appointment in the School of Computing and the Department of Electrical and Computer Engineering. At UNL Eric directs the Smart Materials and Robotics Laboratory, an interdisciplinary research lab that is creating multifunctional soft materials that exhibit a unique combination of mechanical, electrical, and thermal properties. These materials are critical components for the emerging fields of wearable computing, soft robotics, and robotic materials. Prof. Markvicka has received the 2024 NSF CAREER award, 2024 ASME Rising Star of Mechanical Engineering award, 2024 College of Engineering Edgerton Innovation Award, 2023 College of Engineering Excellence in Research Award, and 2021 NUtech Ventures Emerging Innovator of the Year award. Quantifying the Intercellular Mechanisms of Ebolavirus Transmission using Multi-scale Agent-based Modeling Himabindu Kovvali (Elsje Pienaar, advisor) [cid:image005.jpg@01DB1016.9A401EE0] Abstract: Ebola Virus Disease (EVD) is caused by an Ebolavirus (EBOV) infection and is associated with fatal hemorrhagic fever. It has caused more than 30 outbreaks, some with 100% mortality. There is a gap in our knowledge of the protein-protein and lipid-protein interactions that drive effective viral production, making it challenging to develop therapies. Matrix protein VP40, when expressed independently, can produce virus-like particles (VLPs) with similar size, shape, entry, and cell attachment properties as EBOV virions. The production of VLPs is enhanced when VP40 is co-expressed with NP, nucleoprotein, which forms NP inclusions encapsulated in the VP40 VLP matrix. The production of VLPs has been quantified in individual cells with a subcellular differential equation model. Here, we employ this existing data to develop a multi-scale Agent-based model to represent the VP40-NP system in a population of cells using a Java-based modeling system called Repast Simphony. The model has five agent types: Transfected Kidney cells, IB-free-VLP (no NP inclusions), IB-containing-VLP (with NP inclusions), Healthy Kidney cells, and VLP-entered cells. Kidney cells are assumed to be adherent. VLP's have movement via Brownian motion. VLP agents are budded from Transfected Kidney cells and enter Healthy Kidney cells through contact. We stochastically quantified the number and type of VLPs that enter Healthy Kidney cells in a population of cells. Results suggest that irrespective of the kind of VLP produced, their entry into a healthy cell is defined primarily by the number of VLPs in the environment. The findings provide valuable insights to elucidate how single-cell VP40 and NP interactions propagate across a population of cells to sustain robust viral proliferation. This study introduces a novel framework of multi-scale Agent-based modeling in the context of computational virology. Quantifying Hemodynamic Metrics and Assessing Their Uncertainty in Intracranial Aneurysms with Computational Fluid Dynamics and 4D Flow MRI Mohammadreza Balouchestani (Vitaliy Rayz, advisor) [cid:image006.jpg@01DB1016.9A401EE0] Abstract: Proper risk assessment of the stability of unruptured intracranial aneurysms (IA) is crucial for making treatment decisions. In addition to clinical and morphological factors, hemodynamic forces have been linked to both IA growth and rupture, though their exact role remains unclear. Our work is motivated by the relationship between adverse hemodynamic environments and the growth or rupture of IAs. We propose new metrics to assess the spatiotemporal heterogeneity of the wall shear stress (WSS) vector field, to enable comparison between pre- and post-growth stages of IAs. Spatial autocorrelation of WSS is used to evaluate the heterogeneity of the WSS vector field. Additionally, spatial variation of topological shear is introduced to capture sharp directional changes in the vector field. Post-processing analysis is applied to velocity fields derived from computational fluid dynamics (CFD) simulations. The boundary conditions for CFD models are informed by 4D flow MRI measurements and implemented using Windkessel models. An additional focus of this work is to evaluate the reliability of velocity-based metrics derived from 4D flow MRI, by developing an automated, physic-based method for estimating uncertainty. This method uses a standardized mean of patches (SMP) to estimate velocity error correlations by standardizing background patches, regardless of their signal magnitude. Conservation of mass is employed to infer local variance at each voxel, based on observed errors in the velocity divergence and the estimated correlation matrix. The model is capable of capturing various error sources, such as noise, partial volume effects, image resolution and phase wrapping. It also accounts for error correlations between velocity components, as well as spatial and temporal correlations. The statistical distance of the velocity measurement and the modeled velocity error distribution can be used to assess the reliability of both the velocity measurement and the respective velocity-based metrics. The current model has the potential for inferring the uncertainty and improving the accuracy of gradient-based hemodynamic metrics. Geddes Assistant Professor of Biomedical Engineering Abstract: Probing multiplexed basal dendritic computations is significant for understanding how neurons process information, which can directly inform advancements in computation and future neuro-inspired AI systems. Basal dendrites of layer 5 cortical pyramidal neurons exhibit Na+ and N-methyl-D-aspartate receptor (NMDAR) regenerative spikes and are uniquely poised to influence somatic output. Nevertheless, due to technical limitations, how multibranch basal dendritic integration shapes and enables multiplexed barcoding of synaptic streams remains poorly mapped. Here, I will describe my group's efforts in combining 3D two-photon holographic transmitter uncaging, whole-cell dynamic clamp, and biophysical modeling to reveal how synchronously activated synapses (distributed and clustered) across multiple basal dendritic branches are multiplexed under quiescent and in vivo-like conditions. I will show that while dendritic regenerative Na+ spikes promote millisecond somatic spike precision, distributed synaptic inputs and NMDAR spikes regulate gain. These concomitantly occurring dendritic nonlinearities enable multiplexed information transfer amid an ongoing noisy background, including under back-propagating voltage resets, by barcoding the axo-somatic spike structure. Our results unveil a multibranch dendritic integration framework in which dendritic nonlinearities are critical for multiplexing different spatial-temporal synaptic input patterns, enabling optimal feature binding. Drawing from these biological principles, we envision new algorithms that mimic the brain's efficiency and adaptability, focusing on principles such as parallel processing, feature binding, and functioning in noisy environments. These insights will deepen our understanding of neuronal integration and pave the way for more sophisticated AI architectures. Bio: Krishna Jayant is an assistant professor at Purdue University's Weldon School of Biomedical Engineering. His laboratory's research focuses on delivering biophysically based accounts of behaviorally relevant computations using novel electrical and optical neurotechnologies. Projects in the lab cover various topics, including the examination of synaptic and dendritic computations in individual neurons and network-wide circuit computations that underlie sensorimotor integration, including synucleinopathies. Dr. Jayant completed his graduate training in electrical and computer engineering at Cornell University, where he worked with Dr. Edwin Kan on bioelectronics. He subsequently completed postdoctoral training at Columbia University with Dr. Rafael Yuste, Dr. Ken Shepard, and Dr. Ozgur Sahin, researching cutting-edge neurotechnologies to probe synaptic and dendritic biophysics. In addition to the NIH Director's New Innovator Award, Dr. Jayant has been recognized as an NIH NIBIB Trailblazer awardee, a Human Frontiers Science Young Investigator grant awardee, a Ralph E. Seasonal flu shots, which support the physical health pillar of the Healthy Boiler Wellness Program<https://www.purdue.edu/hr/Benefits/healthyboiler/index.php>, are provided annually at no charge to all benefits-eligible Purdue faculty and staff, as well as spouses and dependents covered on a Purdue health plan, through a variety of means. Drive-through flu shots at the Center for Healthy Living on Purdue's West Lafayette campus, 1400 W. State St., Building B, Suite C, begin Sept. 25 from 7 to 9 a.m. Drive-through flu shots will be offered on the south side of the center, with both morning and evening time slots available for added convenience. Drive-through flu shots at the West Lafayette CHL will take place: * Sept. 25, 7 to 9 a.m. * Sept. 26, 4 to 6 p.m. * Oct. 2, 7 to 9 a.m. * Oct 3, 4 to 6 p.m. Additional West Lafayette flu shot availability In addition, the CHL on the West Lafayette campus has on-campus flu shot events planned in October and will offer flu vaccines at the center to benefits-eligible employees and dependents covered on a Purdue health plan on a walk-in basis beginning Oct. 1. Simply stop by to receive a flu shots; appointments are not required. Pediatric patients 6 months and older can also receive their flu shots at the CHL beginning Oct. 1. Another option for flu shots on the West Lafayette campus is the Purdue University Pharmacy (PUP), 575 Stadium Mall Drive. PUP is offering flu shots to all faculty, staff and students as well as dependents (ages 11 and up) by appointment only.
