Good Morning, The BME PMP students will be putting together sack lunches on March 25th in MJIS for the LUM Winter Warming Station as their community service event for this spring. We are reaching out to see if anyone would be willing to donate any items that are needed. We will be providing 80 sack lunches for those in the area that are in need. Tendinopathies range from acute to chronic degeneration leading to micro-or macro-scale partial or complete tears. Most tears fail to heal naturally. Tear repair is currently done through surgical procedures, yet they lead to fibrosis due to extended inflammatory stages and limited matrix remodeling. For example, around 460,000 rotator cuff tears are currently repaired in the U.S. each year and retear rates can be up to 90%. Tissue engineering works towards eliminating these shortcomings. Tendon-engineered approaches supply scaffolds and functional cells to aid regeneration. Engineered novel approaches are evaluated through specific tendon-related outcomes (e.g., expression of tendon-related genes, development of highly aligned collagen). These outcomes evaluate only a small fraction of the tendon phenotype and therefore don't rule out dedifferentiation or an unhealthy phenotype. Hence, promising results can inadvertently lead to debilitating states in unevaluated areas (e.g., fibrosis, metabolic dysregulation). Implementation of Omics platforms to profile tendon biology could define better outcomes and increase our understanding of engineered tendon approaches. Omics characterize molecule types (i.e. genetic information, proteins, metabolites) in a sample without the bias and restrictions of current targeted methods used to quantify known markers. However, deep learning models require large datasets for training, which are often unavailable in medical contexts due to cost, privacy and the rarity of certain conditions. In this talk, Dr. Amir Amini will present strategies to overcome these challenges including innovative machine learning techniques developed at the University of Louisville's Medical Imaging Lab. His work focuses on improving AI capabilities in diagnostic imaging, reducing dependence on invasive procedures and extracting physiologic data from underutilized sources - all while managing the limitations of sparse data environments. Bio: Dr. Amir Amini is the Endowed Chair in Bioimaging and Professor of Electrical and Computer Engineering at the University of Louisville. Prior to his current role he held faculty appointments at Yale University and Washington University in St. Louis. A recognized leader in medical image analysis, he has served in editorial and leadership roles with IEEE Transactions on Medical Imaging and multiple international conferences including SPIE Medical Imaging and the IEEE International Symposium on Biomedical Imaging. His research, funded by NIH, NSF and private industry, spans MRI-based motion and flow analysis and deep learning for diagnostic imaging. Dr. Amini was named a Fellow of the IEEE, AIMBE, SPIE and the International Academy of Medical and Biological Engineering. In 2025, he was appointed Editor-in-Chief of IEEE Transactions on Biomedical Engineering. His laboratory focuses on developing AI-driven analysis tools across a variety of imaging modalities with applications in computer-aided diagnosis and radiation therapy planning. His work also addresses challenges in extracting clinically meaningful data from imaging sequences that were previously inaccessible, helping reduce the need for invasive biopsies and improving patient outcomes. When cancer cells develop resistance to a therapy, cancer can progress or recur at sites local or distant to the treated disease. In recent decades, antibody-drug conjugates have improved the ability to target cancer cells and deliver chemotherapeutic agents with higher specificity. However, recurrence and progression remain common with these drugs. In the present study, we describe a novel mechanism of resistance to the antibody-drug conjugate T-DM1 in HER2 overexpressing breast cancer cells. Although there are several known mechanisms of resistance to T-DM1, it is not fully clear why T-DM1 preforms much worse than the newer drug T-DXd, the same targeting antibody with different cytotoxic payload. Addressing this knowledge gap is vitally important to inform further research and development. Cells become resistant when exposed exclusively to the cytotoxic payload of T-DM1 in the absence of the associated antibody, confirming resistance arising from the microtubule inhibitor. Increasing density of cells and subsequently higher β1 integrin expression correlated with increasing survivability of escalating T-DM1 dose. Further, overexpression of the matrix stabilizing enzyme transglutaminase 2 (TG2) confers a survival advantage in chronic T-DM1 exposure. Inhibition of focal adhesion kinase and myosin II reverses this survival advantage. These facts indicate an a-actinin as a key mediator of resistance. We thus demonstrate the role of TG2 and β1 integrin in HER2 overexpressing breast cancer cells in gaining resistance to T-DM1 via an actin cytoskeletal adaptive mechanism. However, deep learning models require large datasets for training, which are often unavailable in medical contexts due to cost, privacy and the rarity of certain conditions. In this talk, Dr. Amir Amini will present strategies to overcome these challenges including innovative machine learning techniques developed at the University of Louisville's Medical Imaging Lab. His work focuses on improving AI capabilities in diagnostic imaging, reducing dependence on invasive procedures and extracting physiologic data from underutilized sources - all while managing the limitations of sparse data environments. Bio: Dr. Amir Amini is the Endowed Chair in Bioimaging and Professor of Electrical and Computer Engineering at the University of Louisville. Prior to his current role he held faculty appointments at Yale University and Washington University in St. Louis. A recognized leader in medical image analysis, he has served in editorial and leadership roles with IEEE Transactions on Medical Imaging and multiple international conferences including SPIE Medical Imaging and the IEEE International Symposium on Biomedical Imaging. His research, funded by NIH, NSF and private industry, spans MRI-based motion and flow analysis and deep learning for diagnostic imaging. Dr. Amini was named a Fellow of the IEEE, AIMBE, SPIE and the International Academy of Medical and Biological Engineering. In 2025, he was appointed Editor-in-Chief of IEEE Transactions on Biomedical Engineering. His laboratory focuses on developing AI-driven analysis tools across a variety of imaging modalities with applications in computer-aided diagnosis and radiation therapy planning. His work also addresses challenges in extracting clinically meaningful data from imaging sequences that were previously inaccessible, helping reduce the need for invasive biopsies and improving patient outcomes. Yet, despite their widespread use, most catheters remain passive conduits-lacking the ability to sense, actuate, or communicate. This fundamental limitation hampers real-time physiological monitoring, impedes precise therapy delivery, and precludes secure device traceability. In procedures such as thermal ablation and balloon angioplasty, reliance on external estimates rather than direct sensing increases the risk of tissue injury, incomplete treatment, or device malfunction. Additionally, the lack of embedded identifiers exacerbates concerns around counterfeiting and untraceable devices-an issue flagged by the World Health Organization. To address these challenges, we introduce a closed-loop rotary direct ink writing (DIW) platform capable of printing multifunctional electronics directly onto flexible, cylindrical catheter surfaces. The system integrates rotational and linear motion with real-time confocal height sensing and PID-controlled Z-axis adjustment, enabling high-resolution, conformal deposition of functional inks without adhesives or intermediate transfer steps. Using this platform, we fabricate and validate strain sensors for mechanical deformation monitoring, temperature sensors for localized thermal feedback, microheaters for controlled therapeutic heating, and chipless RFID tags for passive wireless identification via frequency-coded resonators. We also propose programmable magnetic actuation using printed magnetorheological elastomer composites for steerable catheter navigation. All components are rigorously tested under physiologically relevant conditions-bending, inflation, heating-demonstrating robust mechanical integrity and electrical performance. Machine learning algorithms decode RFID signatures, which are securely logged on a blockchain framework to enable tamper-proof device tracking.
