Impairments in CSF circulation, arising from sleep disruption, neurological injury, or disease, are associated with increased risk of neurodegeneration and cognitive decline. Although cardiac pulsation contributes to CSF movement, recent work demonstrates that slow, low-frequency oscillations (LFOs) in cerebral blood flow and autonomic activity strongly drive CSF transport. These observations suggest that CSF dynamics may be enhanced by intentionally modulating the physiologic oscillations that couple blood and CSF flow. Specifically, respiration alters heart rate variability (HRV), CO₂ dynamics, and vascular tone, all of which influence cerebral blood flow, and consequently, CSF. However, the mechanistic pathway linking low-frequency respiration to LFO enhancement and downstream CSF motion remains poorly understood. This project aims to determine whether controlled low-frequency respiration can amplify autonomic and vascular oscillations and thereby enhance CSF flow. Aim 1 will quantify how low-frequency breathing (<0.15 Hz) shapes HRV, blood pressure, and low-frequency cerebral blood flow oscillations using concurrent transcranial Doppler ultrasound, fNIRS, PPG, respiration monitoring, and blood pressure measurement. Aim 2 will test whether respiration-driven autonomic oscillations enhance CSF low-frequency power using fMRI with synchronized physiological recordings. We expect that slow breathing will entrain autonomic rhythms, increase LFO power in cerebral blood flow, and drive corresponding increases in CSF oscillations. Despite this progress, existing AI models often require substantial expert knowledge, depend heavily on large-scale labeled datasets, cannot integrate multimodal information effectively, and lack generalization across domains. These limitations highlight the need for label-efficient, multimodal deep learning methods capable of robust segmentation and disease characterization across diverse biomedical imaging modalities. We hypothesize that integrating domain adaptation, semi-supervised, few-shot, and multimodal learning can yield generalizable image representations that enable accurate segmentation and classification with substantially fewer labels. To address this research gap, we will first develop an AI framework that combines task-specific segmentation with radiomics feature extraction to characterize osteoporosis from high-resolution CT scans, validating the effectiveness of machine and deep learning methodologies in a clinically relevant setting. Next, to reduce the dependence on costly manual annotations and improve cross-modality performance, we will design a mixed-domain semi-supervised segmentation pipeline that leverages state-of-the-art domain adaptation strategies and foundation model priors. Finally, to further enhance the representation capacity of standard deep learning models for label-efficient medical image classification, we will explore few-shot learning approaches built on multimodal vision-language models. Title: Investigating Chronic Performance of Ultramicroelectrode Arrays for Intracortical Microstimulation Date: Dec 9th, 2025 Time: 2:00 PM Location: MJIS 2001 Committee: Kevin Otto (Chair), Mark Orazem (Member), Tamara Kinzer-Ursem (Member), Maria Dadarlat (Member) Abstract: Neural stimulation has shown great promise in treating neurological disorders and in developing advanced neuroprosthetic devices, but challenges with traditional electrode arrays persist. These larger microelectrode arrays (MEAs) can cause significant immune response which results in encapsulation of the device by immune cells. The glial sheath that forms can reduce device stimulation and recording efficacy and ultimately limit their long-term stability. Ultramicroelectrode arrays (UMEAs), with electrodes an order of magnitude smaller and a reduced surgical footprint, offer a potential solution by reducing inflammatory responses which in turn minimizes encapsulation of the device and extends the lifespan of the device. Preliminary studies have demonstrated that UMEAs can achieve effective neural stimulation at chronic timepoints, providing initial validation of their potential utility. However, there is still a lack of understanding about how the miniaturization of these electrodes affects their response to standard stimulation parameters over chronic time periods. The goal of this project is to address a key knowledge gap by investigating the impact of electrode size, spacing, and stimulation pulse frequency on the effectiveness of UMEAs. By addressing these critical gaps in knowledge, this research will not only improve fundamental understanding of UMEA performance but could also lead to better therapeutic outcomes for individuals with neurological disorders such as Parkinson's disease, epilepsy, and spinal cord injuries. By minimizing inflammation and improving the precision of neural activation, UMEAs could offer a significant advantage over traditional MEAs by potentially enhancing both the longevity and effectiveness of long-term neuromodulation therapies. This shortens antigen exposure to antigen-presenting cells, limits the duration of immune activation, and often necessitates multiple doses to achieve durable protection. Vaccine formulation development is further complicated by material-induced inflammation, batch-to-batch variability, antigen instability during fabrication, and reliance on iterative, resource-intensive animal testing. These limitations highlight the need for biocompatible vaccine delivery platforms that prolong antigen retention, improve the magnitude and longevity of immune responses without compromising safety or increasing adverse effects, and streamline formulation design while reducing reliance on in-vivo experiments. To address this need, we are integrating the evaluation of an established preclinical rodent model with an ordinary differential equation (ODE)-based computational model. Our preliminary work evaluated an in-situ self-assembling type I oligomeric collagen (Oligomer) material as a vaccine depot to localize Ovalbumin (OVA) and CpG, and assessed its effects on antigen retention, local tissue response, and systemic inflammation. We are developing a mechanistic ODE-based computational framework to quantify and predict vaccine-induced antigen availability and immune response dynamics to evaluate vaccine design parameters. We will integrate predictive modeling with in-vivo validation by using the computational model to predict antibody responses and assessing antibody responses and infertility outcomes in vivo following an Oligomer depot-based, single-dose contraceptive vaccine. These proteins are implicated in several neurological disorders and integral players in healthy inter-neuronal signaling resulting in learning and memory formation via a calcium signaling cascade. The structure of this NMDA Receptor Complex has never been fully elucidated despite decades of research addressing the individual proteins. Likewise, the regulation of the complex via post-translational phosphorylation is yet to be investigated. This study will determine whether all five proteins simultaneously incorporate into the complex via prediction of the complex structure with AlphaFold Multimer and verification of the structure through comparison of reported interaction sites in the literature. The regulation of the complex will be elucidated via all-atom molecular dynamics simulations of the complex with and without phosphorylated modifications.
