Lafayette/West Lafayette/Purdue University are currently under a tornado watch (until 4:00 p.m.)
Tornado Watch The National Weather Service or NOAA issues a "Tornado Watch" when local atmospheric conditions are favorable for the formation of tornadoes. During a tornado watch, stay informed via one or more of the weather warning signals. You do not need to shelter in place, but rather remain aware of rapidly changing weather conditions. Should the NWS upgrade the Watch to a Warning, that means that you should take immediate actions to insure your safety.
Purdue University is located in a region of the United States that is susceptible to dangerous weather events such as severe thunderstorms and tornadoes. Faculty, staff, and students must prepare themselves to respond to severe weather by reviewing building emergency plans to locate the building "shelter in place" location (in MJIS that is the basement corridors) and consider the following in any building:
Tornado Warning The National Weather Service or NOAA issues a "Tornado Warning" when atmospheric conditions make a tornado likely or a tornado has been sighted and you must seek shelter immediately. Monitor the expiration of the tornado warning, and remained sheltered until the tornado warning expires.
I. Preparation - Shelter in place location and weather warnings
a. Locate the accessible basement or lowest interior corridor in the building.
b. Identify a shelter in place location away from glass and exterior walls, if possible.
c. Stay informed through local media sources when severe weather is expected.
d. Obtain a National Oceanic and Atmospheric Administration (NOAA) weather application or radio.
e. Keep a working flashlight in your office or residential area.
II. Response - Go indoors and shelter in place
a. A "Tornado Warning" issued by the National Weather Service means atmospheric conditions make a tornado likely or a tornado has been sighted and you must seek shelter immediately. When the All-Hazards Outdoor Warning Sirens sound (typically for three minutes) or NOAA sounds a warning alarm tone on the weather application or radio, you must seek shelter immediately. Proceed to the shelter in place location or the lowest level of any building, away from glass and exterior walls, if possible.
b. A Tornado Warning can be considered "all clear" when NOAA states that the Tornado Warning has expired. NOAA weather applications and local radio and television stations can be used as a source for the expiration of the Tornado Warning time period.
c. Always remain in a place of shelter until you receive an all-clear message or the warning has expired.
d. Dial 911 (Purdue Public Safety Dispatch) to report any life threatening emergency conditions resulting from the storm.
e. Stay out of damaged buildings and avoid broken utility lines (i.e., gas, electric, water).

DEPUTY HEAD'S NOTE
* Dr. Taeyoon Kim graciously accepted the co-chair role for the Computational Biomedicine research area after I became the Deputy Head. Since becoming the new area co-chair, he and Dr. Elsje Pienaar have worked together to develop a ranked list of distinguished seminar speakers for the Spring 2024 session. Thanks go out to them. Now the area faculty are working on getting the invitations out. Getting distinguished and excellent speakers to our school and on our campus is something important and beneficial to our school facing outwardly.
* Four items were covered in last Friday's Engineering Leadership Team meeting. They are 1) launch of Purdue International Scholar Assistance (PISA); 2) CoE application and admission; 3) CoE COACH survey; and 4) raise of the graduate salaries.
* PISA is a new, more centralized service of ISS provided to non-US Purdue-sponsored international faculty, staff, post-docs, and visiting scholars. Beginning in July 2023, PISA will serve as administrative liaisons for our international faculty, staff, postdocs, and scholars, and their hosts and hiring units. Our CoE will be the test unit, which will start the adaption on March 1st, and anticipated to conclude on April 1. Beginning March 26, all J-1 visa requests will be directed to PISA; Permanent residence applications will still be handled by Cris Crozier, who will remain managing these until October; changes on H1-B requests (faculty) will apply to new faculty offers after March 26. For more information, please stay tuned for more information from the ISS over this semester. I am also happy to get you connected with the right people at ISS.
* The CoE target enrollment for the new cycle (2023 - 2024) is 2300. Up to now, the admissions have been down a bit comparing to this time last year. Applications are down among residents and international; and up among domestic non-residents; the number of women is up for both applicants and admits; and URM are up for both applications and admits; most of the merit-based awards are out by now.
* COACH survey: CoE 56% respondence, entire Purdue 45%. I am pleased to report to you that we are one of the three CoE units with above 50% response rate. Kudos to all of you. The analysis is completed by a task force and the analysis results should be out soon. Focus groups will be soon formed to strategize the improvements. I am happy to discuss with you about the survey results once you have a read-out.
* Drs. Tami Kinzer-Ursem and Mike Heinz have been actively working with the Associate Dean for Graduate Education on the matter of raising graduate salaries. Stay tuned for more information.
* Some of you have sent me requests for supporting/nominating/endorsing applications of yours and/or your postdocs and students for college-level, campus-wide, and external opportunities for career betterment. Thank you! For all, please do not hesitate to take such actions. I often take an approach of handling those requests upon their arrivals.

AAE Spring 2023 Colloquium Series, Tues., Feb. 28, 3:00 p.m. in ARMS 1103. Christian Franck, Bjorn Borgen Professor, Panther Program Director, and Director of the Center for Traumatic Brain Injury at the University of Wisconsin-Madison will present "Cellular NeuroMechanics-Concussions, Traumatic Brain Injury and the Mysterious Havana Syndrome."
Abstract: Current prediction, prevention and diagnosis strategies for mile traumatic brain injuries, including concussions, are still largely in their infancy due to a lack of detailed understanding and resolution of how physical forces give rise to tissue injury at a cellular level. In this talk I will present some recent work on our current understanding of the origin of concussions and traumatic brain injuries and how cells in the brain interpret and react to the physical forces of trauma. Specifically, I will show that the path to a better understanding of traumatic injuries involves addressing a variety of finite deformation, rate-dependent soft matter and cell mechanics problems along the way. Finally, I will provide an update on how our current understanding of the cellular neuromechanics cannot only help shed light on improving our prediction of TBI but also enable us to dissect the physical origin of emerging injuries such as the Havana Syndrome.
Biography: Christian Franck is a mechanical engineer specializing in cellular biomechanics and new experimental mechanics techniques at the micro and nanoscale. He received his B.S. in aerospace engineering from the University of Virginia in 2003, and his M.S. and Ph.D. from the California Institute of Technology in 2004 and 2008. Dr. Franck held a post-doctoral position at Harvard investigating brain and neural trauma. He was an assistant and associate professor in mechanics at Brown University from 2009 - 2018, and is now the Bjorn Borgen Professor in Mechanical Engineering at the University of Wisconsin-Madison. His lab at the University of Wisconsin-Madison has developed unique three-dimensional full-field imaging capabilities based on multiphoton microscopy and digital volume correlation. Current application areas of these three-dimensional microscopy techniques include understanding the 3D deformation behavior of neurons in the brain during traumatic brain injuries, and the role of non-linear material deformations in soft matter. He is the acting director of the Center for Traumatic Brain Injury at the University of Wisconsin-Madison and the ONR-funded Physics-based Neutralization of Threats to Human Tissues and Organs (PANTHER) program, which consists of over 24 PIs nationwide. Key objectives of the Panther program are in better detection, prediction, and prevention of traumatic brain injuries by providing accelerated translation from basic science discovery to civilian and warfighter protection solutions.

Joseph Wallace, PhD, Chair of the Department of Biomedical Engineering and Professor of Biomedical Engineering, Indiana University School of Medicine will present "Targeting Tissue Quality as an Interventional Approach to Improve Bone Material Properties and Fracture Resistance."
Abstract: The Bone Biology and Mechanics Laboratory (BBML) at IUPUI is committed to improving our understanding of how the body works through innovative preclinical and clinical research. The lab focuses on bone mechanics (ability to effectively bear loads) and mechanobiology (how cells respond to mechanical stimuli to maintain and/or increase bone mass) in the context of disease and treatment. We investigate bone from mechanical engineering and materials science perspectives by tying morphology and composition to mechanical function and fracture resistance at discrete length scales throughout bone's rich tissue hierarchy. Our long-term goal is to use combinatorial treatments targeting deficiencies in bone mass/quantity and bone tissue quality in complex disease states that have musculoskeletal complications leading to fragility and fracture. Our current focus is on mechanical and pharmaceutical manipulations in diseases including diabetes and chronic kidney disease.
Biography: Joseph Wallace is a Professor and Chair of the Department of Biomedical Engineering at Indiana University-Purdue University Indianapolis (IUPUI). He graduated with a B.S in Aerospace Engineering from Georgia Tech in 2002 before attending the University of Michigan for his PhD studies in Biomedical Engineering. After earning his PhD in 2007, he chose to remain at Michigan for a Postdoctoral Fellowship in the Department of Chemistry with funding from an NIH F32 individual postdoctoral award. He joined the BME faculty at IUPUI in September 2010 and has developed an active research program in the areas of bone mechanics and mechanobiology. He is currently funded as Principal Investigator by grants from NIH, NSF, and the Department of Veterans Affairs with several other co-Investigator grants from NIH and the VA. His lab is actively looking to grow so please reach out if interested in a funded PhD or postdoctoral position.

Title: Novel Microfluidic Device for In-Field Purification, Amplification, and Detection of DNA from Mycotoxin-Producing Fungi.

Abstract: Mycotoxins are toxic molecules produced by fungi and are harmful to human health when ingested. Mycotoxins produced by fungi that infect food crops can enter the food stream directly in manufactured produces (foods like bread) or indirectly through animal feed. Detection of mycotoxins is commonly performed after harvesting and processing of the crops because mycotoxins can increase in concentration during these steps. However, if farmers could detect the toxin-forming fungi prior to harvesting, growers can reduce cross-contamination, save wasted resources used in harvesting and processing of infected crops, and increase food safety and security. Fusarium graminearum is a fungal species that infects cereal crops and produces deoxynivalenol (DON), a harmful mycotoxin called vomitoxin. Because Fusarium graminearum has a large economic impact in the United States and globally on wheat and barley markets, it was selected as a proof-of-concept for the detection assay. For detection of fungus to be useful in determining infected areas of fields, the device must be inexpensive and portable. A low-cost, portable microfluidic device was designed to both extract the fungal DNA from wheat samples and perform a fluorescence-based loop-mediated isothermal amplification (LAMP) assay to detect the fungal DNA. Contaminated wheat samples were used to demonstrate the effectiveness of the device. The device was able to detect Fusarium graminearum in the wheat samples at concentrations relevant to DON concentration limits set forth by the U.S. Food and Drug Administration (FDA). Detection of multiple mycotoxin-producing fungal species is possible through the addition of more LAMP assays for multiplexing specific target genes.

Abstract: Diabetes, a chronic condition characterized by elevated blood glucose levels, arises when pancreatic β-cells lose capacity to produce a robust, dynamic glucose-stimulated insulin secretion (GSIS) response. Accurate measurement of β-cell health and function ex vivo is thus fundamental to diabetes research, including studies evaluating disease mechanisms, novel drug candidates, and replacement β-cell populations. However, present-day dynamic GSIS assays typically represent end-point measurements, involve expensive commercial perifusion machines, and require time-consuming enzyme-linked immunosorbent assays (ELISA) for insulin detection. Microfluidic devices developed as accessible, low-cost alternatives still rely on secondary ELISAs and suspend islets in liquid medium, limiting their survival in vitro. Here, we present a novel, 3D-printed microphysiological system (MPS) designed to recreate components of in-vivo microenvironments through encapsulation in fibrillar type I collagen and restoration of favorable molecular transport conditions. Following computational-informed design and rapid prototyping, the MPS platform sustained collagen-encapsulated mouse islet viability and cytoarchitecture for 5 days and supported in situ measurements of dynamic β-cell function. To rapidly detect insulin secretion from β-cells in the MPS, we then developed a highly sensitive electrochemical sensor for zinc (Zn2+), co-released with insulin, based on glassy carbon electrodes modified with bismuth and indium and coated with Nafion. Finally, we validated sensor detection of Zn2+ released from glucose-stimulated INS-1 β-cells and primary mouse islets, finding high correlation with insulin as measured by standard ELISA. Together, the 3D MPS and Zn2+ sensor developed in this dissertation represent novel platforms for evaluating dynamic β-cell function in a low-cost, user-friendly, and physiologically-relevant manner.

Ultrastructural and molecular biological studies have revealed the nanoscale structures, molecular components, and signaling cascades regulating the actin cytoskeleton step by step. However, it remains unclear how nanometer-sized molecules are self-organized into cell-scale structures and how they cooperate to drive cell functions. To understand the underlying physical principles that govern the cytoskeletal assembly and cell morphogenesis, we employed an in vitro reconstitution approach. We established a method to encapsulate purified proteins or the cytoplasmic extracts of living cells into cell-sized capsules. Using this minimal model of the cell, we are seeking biochemical and physical conditions under which structures and functions recapitulating living cells are self-organized. In this seminar, I will present our recent efforts in understanding the regulatory mechanisms of cell division machinery and motility.
Biography: Makito completed doctoral degree at Kyoto University. He did his postdoctoral work with Prof. Shin'ichi Ishiwata at Waseda University reconstituting active cytoskeleton from purified proteins. After working as Assistant Professor at Waseda University, he joined The Hakubi Center for Advanced Research at Kyoto University as a Principal Investigator in October 2017. He started his laboratory aiming to understand the self-organization mechanism of the cytoskeleton and biological functions, using a combination of single-molecule, in vitro reconstitution, and live cell experiments, and theories of physics. He was appointed as Program-Specific Associate Professor at Department of Physics, Kyoto University in October 2022.

Abstract: Positron emission tomography (PET) is a nuclear medicine imaging technique that supports the quantification of physiologic processes within the human body. PET radiotracers are developed with site-specific binding properties or as metabolic analogs, enabling specific tissue properties to be investigated with minimal disturbance to the underlying physiology. Clinically, the radiotracer standard uptake value is a semiquantitative metric that quantifies radiotracer accumulation over a single time interval. However, compartmental modeling of dynamic PET acquisitions provides additional insights into tissue perfusion, binding avidity, and intracellular tracer trapping. Dynamic PET image analysis techniques are first applied to 18F-fluoroethyltyrosine (FET) PET imaging of high-grade glioma and brain metastasis patients. Standard-of-care MRI is often equivocal for discriminating post-treatment imaging aberrations from disease progression. However, temporal 18F-FET efflux (tumor-to-brain ratio slope, intercept) was found to be a highly accurate (95%) metric for identifying cases of disease progression. In a second application, dynamic PET image analysis techniques are applied to 68Ga-PSMA-11 PET imaging for primary prostate cancer patients. In the presurgical planning for prostatectomy, nerve-sparing or nerve-excising surgery is elected based on the dual consideration of metastatic cancer risks and surgically-induced side effects. Even with advanced multiparametric MRI techniques, prostate cancer is often not effectively localized, resulting in over- or under-treatment. Dynamic 68Ga-PSMA-11 PET semiquantitative and kinetic parameters improve the localization of prostate cancer, reducing the prevalence of poor surgical outcomes. Overall, the application of dynamic PET imaging techniques supports improved clinical outcomes and enhanced clinician confidence for treatment modifications.

Frontiers in Biophysics Seminar Series, Wed., Apr. 5, 1:30 p.m. in MJIS 1001. Joe Howard, the Eugene Higgins Professor of Molecular Biophysics and Biochemistry, Department of Molecular Biophysics and Biochemistry, Yale University, will present "Mechanical Stability of Microtubules." Microtubules are rigid polymers of tubulin that give structure to cells and serve as tracks for molecular motors. Until recently, the growth and shrinkage of microtubules was thought to be confined to their ends. Over the last few years, however, evidence has emerged that exchange of tubulin in and out of microtubules can take place within the lattice and not just at the ends. I will present our recent work on the microtubule severing protein spastin on microtubule dynamics as well as measurements of the forces necessary to remove tubulin from the lattice. Special BME Seminar, Mon., March 27, 9:30 a.m., MJIS 2001. Joseph R. Leach, MD, PhD, Assistant Professor in Residence, Department of Radiology and Biomedical Engineering at the University of California, San Francisco, will present "Abdominal Aortic Aneurysm Assessment - Moving Beyond Measurement."
Abstract: Abdominal aortic aneurysms (AAA) have a pooled global prevalence of 4.8% and are most common in men 65 years or older. Small AAAs (<4 cm) are commonly asymptomatic and pose little danger, while larger AAAs have a significant risk of fatal rupture that increases with increasing aneurysm diameter. Despite decades of research elucidating the histologic and biochemical features of AAA evolution, clinicians have relatively little capacity to predict or modify AAA progression in practice - we are lacking an understanding of AAA progression on a patient-specific basis, informed by factors readily assessed during periodic imaging-based surveillance. In his talk, Dr Leach will review the current state-of-the-art in clinical AAA evaluation, and present ongoing research on a comprehensive AAA assessment in which aneurysm measurement is augmented by advanced imaging-based measures of aneurysm inflammation, biomechanics, and higher-order morphologic features.
Bio: Joseph R Leach, MD PhD is an Assistant Professor in Residence in the Department of Radiology and Biomedical Imaging at the University of California, San Francisco. Dr. Leach obtained his PhD in Bioengineering in 2009 from University of California -Berkeley, and completed medical school, residency, and a fellowship in abdominal imaging at UCSF prior to joining the faculty in 2018. Dr. Leach has broad clinical and research interests in vascular disease and applies advanced imaging techniques and imaging-based biomechanical modeling to better understand factors influencing disease initiation, progression, and patient outcomes. Outside of the hospital, Dr. Leach enjoys cooking for his wife, Joyce and their cats Jacks and Nigel, and is a lifelong musician and terrible dancer.

Course Description: The unique medical needs of pediatric patients in the context of current and emerging clinical technologies are presented in this novel course. Anatomical, physiological, and pathophysiological underpinnings of diseases in infants and children are studied. Unique aspects of pediatric device design, regulation, and translation, including clinical studies and ethical considerations, are explored. Numerous clinical sub specialties such as cardiology, surgery, nephrology, pulmonology, and gastroenterology are examined in terms of technology adoption and remaining clinical needs. Future directions in device development and emerging trends in patient care are considered throughout.

* I am pleased to report to you that we are ahead of curve in terms of the IUPUI BME faculty integration. The dates for submission of both tenure track and non-tenure track cases is March 1st. Three more seminars given by IUPUI BME faculty will be on 3/1, 4/12, 4/19, all hosted by Young Kim.
* Thank you for those who met with Dr. Fiona Kolbinger and provided your assessments. I will provide an update on her case in the next couple of weeks.
* On 2/23, Alexis Hoerter, a PhD student in Dr. Elsje Pienaar’s group, will talk in a RCHE Graduate Student seminar about her work on in-silico modeling of M. tuberculosis granuloma dynamics. I would like to take the liberty to highlight the upcoming presentations given by our BME Grad/UG students and Grad/UG students in your research labs in this Deputy Head’s Note block of the Roundtable every week. The session will be led by Chris Lukasik, provost’s fellow for Fulbright faculty awards, and will be followed by a Q&A. “Purdue continues to be a top-producing institution for the Fulbright Scholar Program,” Lukasik says. “Over half of all Purdue faculty applicants for the Fulbright Scholar award program end up winning.” Faculty interested in developing or expanding their international profile will learn more about the two types of Fulbright awards programs: 1) Fulbright Scholar Award and 2) Fulbright Specialist Award. Purdue professional development program within the Office of Undergraduate Research is now accepting applications from instructors of any discipline for the 2023-24 academic year. CURE – Purdue seeks to develop course-based undergraduate research experiences (CUREs) across the Purdue West Lafayette campus as a way to increase undergraduate students’ overall access to novel research. The program provides instructors with the time and framework for designing a CURE, either by transforming an existing course or developing a new one. CUREs have a number of features and positive outcomes, including increasing the overall capacity for undergraduate research by operating at a larger scale than the traditional apprenticeship model and increasing inclusion by allowing any eligible student to enroll. Research findings suggest additional benefits to instructors:
* Greater enjoyment when teaching CUREs over a traditional course.
* Ability to integrate their research and teaching.
* Advancement of their research agenda.
* Recruiting students to their personal research group.
* Potential to positively impact promotion and tenure.

Rita Patel, PhD, CCC-SLP, Associate Professor and Speech Language Pathologist, Department of Otolaryngology, Head and Neck Surgery, Indiana University School of Medicine, will present “An interdisciplinary scientific journey.”
Abstract: Voice pathologies occur across the life span, are disabling, interfere with gainful employment and compromise quality of life. According to the National Institute on Deafness and Other Communication 7.5 million people in the United States have voice disorders. It has been estimated that over 1 million or 6-9% children suffer from dysphonia nationwide. Estimates indicate that anywhere from 3-9% of the population has some type of voice dysfunction at any given point in time. Reported occurrence of hoarseness in the school-aged children ranges from 6%-23%. Dysphonia can be detrimental both psychologically and academically, hence early identification and restoration of optimal vocal health is critical. If untreated, dysphonia can substantially increase health care costs. Unfortunately, efficient diagnosis and effective management of voice disorders is challenged by difficulty accessing professionals with the necessary expertise and the availability of valid instrumentation to capture highly aperiodic voice qualities to diagnose and treat voice problems efficiently. To address these issues, technological advancements from a collaborative team of interdisciplinary experts in the fields of medicine, engineering, physics, and psychology show great promise for improving the efficiency and effectiveness of voice disorders assessment and treatment. This talk will focus on my current interdisciplinary work for advancing clinical assessment of voice disorders across the lifespan in the areas of laryngeal imaging, computational fluid dynamics, and mobile technology.
Biography: Dr. Rita Patel is an associate professor at the Department of Speech, Language and Hearing Sciences, Indiana University and an adjunct associate professor at the Department of Otolaryngology, Head and Neck Surgery Indiana University School of Medicine. Her research and clinical expertise focus on addressing voice and upper airway disorders in adult and pediatric population, with specialization in endoscopic laryngeal imaging. Dr. Patel has been awarded 8 extramural grants from National agencies including ASHA (American Speech Language Hearing Association) and NIH (National Institutes of Health); and 5 intramural grants. In 2022, Dr. Patel was awarded fellow by the American Speech-Language-Hearing Association (ASHA) for her outstanding contributions to research and excellent teaching in the area of voice and voice disorders. Currently, Dr. Patel serves as the editor for the American Journal of Speech-Language Pathology, American Speech-Language-Hearing Foundation’s Board of Trustees, and as an Advisory Board Member of Voice Foundation. She also serves as a reviewer for numerous journals in the fields of laryngology and speech language pathology and also serves as grant reviewer for the NIH study section, the Voice Foundation, and ASHA.

AAE Spring 2023 Colloquium Series, Tues., Feb. 28, 3:00 p.m. in ARMS 1103. Christian Franck, Bjorn Borgen Professor, Panther Program Director, and Director of the Center for Traumatic Brain Injury at the University of Wisconsin-Madison will present “Cellular NeuroMechanics-Concussions, Traumatic Brain Injury and the Mysterious Havana Syndrome.”
Abstract: Current prediction, prevention and diagnosis strategies for mile traumatic brain injuries, including concussions, are still largely in their infancy due to a lack of detailed understanding and resolution of how physical forces give rise to tissue injury at a cellular level. In this talk I will present some recent work on our current understanding of the origin of concussions and traumatic brain injuries and how cells in the brain interpret and react to the physical forces of trauma. Specifically, I will show that the path to a better understanding of traumatic injuries involves addressing a variety of finite deformation, rate-dependent soft matter and cell mechanics problems along the way. Finally, I will provide an update on how our current understanding of the cellular neuromechanics cannot only help shed light on improving our prediction of TBI but also enable us to dissect the physical origin of emerging injuries such as the Havana Syndrome.
Biography: Christian Franck is a mechanical engineer specializing in cellular biomechanics and new experimental mechanics techniques at the micro and nanoscale. He received his B.S. in aerospace engineering from the University of Virginia in 2003, and his M.S. and Ph.D. from the California Institute of Technology in 2004 and 2008. Dr. Franck held a post-doctoral position at Harvard investigating brain and neural trauma. He was an assistant and associate professor in mechanics at Brown University from 2009 - 2018, and is now the Bjorn Borgen Professor in Mechanical Engineering at the University of Wisconsin-Madison. His lab at the University of Wisconsin-Madison has developed unique three-dimensional full-field imaging capabilities based on multiphoton microscopy and digital volume correlation. Current application areas of these three-dimensional microscopy techniques include understanding the 3D deformation behavior of neurons in the brain during traumatic brain injuries, and the role of non-linear material deformations in soft matter. He is the acting director of the Center for Traumatic Brain Injury at the University of Wisconsin-Madison and the ONR-funded Physics-based Neutralization of Threats to Human Tissues and Organs (PANTHER) program, which consists of over 24 PIs nationwide. Key objectives of the Panther program are in better detection, prediction, and prevention of traumatic brain injuries by providing accelerated translation from basic science discovery to civilian and warfighter protection solutions.

Title: Novel Microfluidic Device for In-Field Purification, Amplification, and Detection of DNA from Mycotoxin-Producing Fungi.

Abstract: Mycotoxins are toxic molecules produced by fungi and are harmful to human health when ingested. Mycotoxins produced by fungi that infect food crops can enter the food stream directly in manufactured produces (foods like bread) or indirectly through animal feed. Detection of mycotoxins is commonly performed after harvesting and processing of the crops because mycotoxins can increase in concentration during these steps. However, if farmers could detect the toxin-forming fungi prior to harvesting, growers can reduce cross-contamination, save wasted resources used in harvesting and processing of infected crops, and increase food safety and security. Fusarium graminearum is a fungal species that infects cereal crops and produces deoxynivalenol (DON), a harmful mycotoxin called vomitoxin. Because Fusarium graminearum has a large economic impact in the United States and globally on wheat and barley markets, it was selected as a proof-of-concept for the detection assay. For detection of fungus to be useful in determining infected areas of fields, the device must be inexpensive and portable. A low-cost, portable microfluidic device was designed to both extract the fungal DNA from wheat samples and perform a fluorescence-based loop-mediated isothermal amplification (LAMP) assay to detect the fungal DNA. Contaminated wheat samples were used to demonstrate the effectiveness of the device. The device was able to detect Fusarium graminearum in the wheat samples at concentrations relevant to DON concentration limits set forth by the U.S. Food and Drug Administration (FDA). Detection of multiple mycotoxin-producing fungal species is possible through the addition of more LAMP assays for multiplexing specific target genes.

Abstract: Diabetes, a chronic condition characterized by elevated blood glucose levels, arises when pancreatic β-cells lose capacity to produce a robust, dynamic glucose-stimulated insulin secretion (GSIS) response. Accurate measurement of β-cell health and function ex vivo is thus fundamental to diabetes research, including studies evaluating disease mechanisms, novel drug candidates, and replacement β-cell populations. However, present-day dynamic GSIS assays typically represent end-point measurements, involve expensive commercial perifusion machines, and require time-consuming enzyme-linked immunosorbent assays (ELISA) for insulin detection. Microfluidic devices developed as accessible, low-cost alternatives still rely on secondary ELISAs and suspend islets in liquid medium, limiting their survival in vitro. Here, we present a novel, 3D-printed microphysiological system (MPS) designed to recreate components of in-vivo microenvironments through encapsulation in fibrillar type I collagen and restoration of favorable molecular transport conditions. Following computational-informed design and rapid prototyping, the MPS platform sustained collagen-encapsulated mouse islet viability and cytoarchitecture for 5 days and supported in situ measurements of dynamic β-cell function. To rapidly detect insulin secretion from β-cells in the MPS, we then developed a highly sensitive electrochemical sensor for zinc (Zn2+), co-released with insulin, based on glassy carbon electrodes modified with bismuth and indium and coated with Nafion. Finally, we validated sensor detection of Zn2+ released from glucose-stimulated INS-1 β-cells and primary mouse islets, finding high correlation with insulin as measured by standard ELISA. Together, the 3D MPS and Zn2+ sensor developed in this dissertation represent novel platforms for evaluating dynamic β-cell function in a low-cost, user-friendly, and physiologically-relevant manner.

Frontiers in Biophysics Seminar Series, Wed., Apr. 5, 1:30 p.m. in MJIS 1001. Joe Howard, the Eugene Higgins Professor of Molecular Biophysics and Biochemistry, Department of Molecular Biophysics and Biochemistry, Yale University, will present “Mechanical Stability of Microtubules.” Microtubules are rigid polymers of tubulin that give structure to cells and serve as tracks for molecular motors. Until recently, the growth and shrinkage of microtubules was thought to be confined to their ends. Over the last few years, however, evidence has emerged that exchange of tubulin in and out of microtubules can take place within the lattice and not just at the ends. I will present our recent work on the microtubule severing protein spastin on microtubule dynamics as well as measurements of the forces necessary to remove tubulin from the lattice. This seminar is sponsored by the Molecular Biophysics Training Program (MBTP), Structural and Computational Biology and Biophysics (SCBB) and the Weldon School of Biomedical Engineering.

Course Description: The unique medical needs of pediatric patients in the context of current and emerging clinical technologies are presented in this novel course. Anatomical, physiological, and pathophysiological underpinnings of diseases in infants and children are studied. Unique aspects of pediatric device design, regulation, and translation, including clinical studies and ethical considerations, are explored. Numerous clinical sub specialties such as cardiology, surgery, nephrology, pulmonology, and gastroenterology are examined in terms of technology adoption and remaining clinical needs. Future directions in device development and emerging trends in patient care are considered throughout.

Abstract:
Positron emission tomography (PET) is a nuclear medicine imaging technique that supports the quantification of physiologic processes within the human body. PET radiotracers are developed with site-specific binding properties or as metabolic analogs, enabling specific tissue properties to be investigated with minimal disturbance to the underlying physiology. Clinically, the radiotracer standard uptake value is a semiquantitative metric that quantifies radiotracer accumulation over a single time interval. However, compartmental modeling of dynamic PET acquisitions provides additional insights into tissue perfusion, binding avidity, and intracellular tracer trapping. Dynamic PET image analysis techniques are first applied to 18F-fluoroethyltyrosine (FET) PET imaging of high-grade glioma and brain metastasis patients. Standard-of-care MRI is often equivocal for discriminating post-treatment imaging aberrations from disease progression. However, temporal 18F-FET efflux (tumor-to-brain ratio slope, intercept) was found to be a highly accurate (95%) metric for identifying cases of disease progression. In a second application, dynamic PET image analysis techniques are applied to 68Ga-PSMA-11 PET imaging for primary prostate cancer patients. In the presurgical planning for prostatectomy, nerve-sparing or nerve-excising surgery is elected based on the dual consideration of metastatic cancer risks and surgically-induced side effects. Even with advanced multiparametric MRI techniques, prostate cancer is often not effectively localized, resulting in over- or under-treatment. Dynamic 68Ga-PSMA-11 PET semiquantitative and kinetic parameters improve the localization of prostate cancer, reducing the prevalence of poor surgical outcomes. Overall, the application of dynamic PET imaging techniques supports improved clinical outcomes and enhanced clinician confidence for treatment modifications.

Note that hybrid or all-virtual prelims are allowable if your advisor and committee members agree to this format.

The Purdue OWL is offering an in-person workshop for doctoral students in their 3rd-5th years. Academics need to produce a quantity of high-caliber writing on a regular basis. The Writing Tools for Mid-Stage Doctoral Students workshop series combines lessons related to productivity with independent writing time that will further attendees' productivity on specific projects they are currently writing. The workshop meets weekly for 5 weeks in March, with the week of Spring Break used for optional writing time. Participants must attend all 4 of the other weeks and are encouraged to attend Spring Break week. Topics of the workshop include Transitioning from Research to Writing, Maintaining Writing and Publishing Productivity, Clarity & Concision, and Peer Review. Coffee and bagels will be provided, compliments of the Purdue Graduate School.
