Academic Programs Update
We are writing to ask for your help in encouraging all instructors to make plans now for how they will approach providing final exam testing accommodations for those of their students who have this disability accommodation. Many instructors will plan to provide these accommodations themselves. That is great and often the best approach. However, some will want the DRC’s Accommodated Testing staff to administer their final for these students. In these cases, it is important that instructors are aware of three critical deadlines.
* November 11. This is the deadline for instructors to update their Test Information Form<https://www.purdue.edu/drc/testing/tif.php> (TIF) for finals week, which effectively gives permission for students to request a seat with DRC Testing.
* November 18 (11:55 p.m.). This is the deadline by which students must request a seat with DRC Testing. The DRC does everything in its power to encourage students to communicate with their instructors and do their part in meeting this and other deadlines.
* 48 hours before exam time. DRC Testing must have the exam in their possession to ensure that the student can access it at the scheduled time.
If these deadlines are met, DRC Testing will make every effort to administer the instructor’s exam.
One last consideration: While DRC Testing does everything it can to work with instructors, they cannot promise to administer the instructor’s exams at the same time that their class is taking it in all cases. Instructors who are able to be flexible with the date and time for their final exam increase the likelihood that DRC Testing will be able to support them.

Undergraduate Programs Update
The Provost’s annual Instructional Equipment Program will proceed in 2022-23, with the timeline noted below. This program provides funds to acquire new instructional equipment on the West Lafayette campus. The equipment should be directly tied to undergraduate courses and curricula. Funds will be awarded on a competitive basis with priority given to proposals that support innovative and high impact teaching models or student experiences. Each college may submit up to three proposals and all proposals must be ranked by the dean in priority order.

The NIH OITE is pleased to offer the RAISING A RESILIENT SCIENTIST<https://www.training.nih.gov/workshops_and_trainings_for_extramural_princip…> series for faculty, staff scientists, and administrators who mentor students and postdoctoral fellows in the biomedical, behavioral, and social sciences. A variety of topics will be covered with a specific focus on communication and shared problem-solving to help trainees deal with the inevitable setbacks experienced in high knowledge environments. The goal of the Raising a Resilient Scientist series is to promote the mental health and well-being of the academic research community by supporting faculty and administrators to develop self-management, relationship-management, and mentoring skills. There will be five units in the series and start on November 9th, 2022; each unit will consist of a 75-minute lecture followed by a 1-hour facilitated discussion with peers.
Previous participants found participation very valuable, and the tools presented practical and actionable. Here is what those who participated in series have to say:
1. “Informative workshop that I think all faculty should be required to attend. Making this a requirement for mentoring teams for fellowship and training grants would be a great approach! They need to be invested in mentoring and training, as much as training fellows.”
2. “Thank you! I've done a lot of mentoring workshops/training but feel like this one is already taking things to another level.”
3. “This course needs to be offered again and again, such an important resource in any academic curriculum.”

Special BME Seminar, Tues., Nov. 1, 11:00-12:00 p.m., MJIS 2001. Pierre Sicard, PhD, Research Engineer at Universite de Montpellier, INSERM, France will present “Cerebrovascular Hypoxia During Experimental Traumatic Brain Injury.”
Abstract
Background: Retrospective studies suggest that mild traumatic brain injury (mTBI) in pediatric patients may lead to an increased risk of cardiac events. However, the exact functional and temporal dynamics and the associations between heart-brain pathophysiological trajectories are not understood.
Methods: A single impact to the left somatosensory cortical area of the intact cranium was performed on juvenile mice 17 days postnatal. Cerebral 3D photoacoustic imaging was used to measure the oxygen saturation (sO2) in the impacted area 4 hours after mTBI followed by 2D and 4D echocardiography at days 7, 30, 90, and 190 post-impact. At 8 months, we performed a dobutamine stress test to evaluate cardiac function. Lastly, behavioral analyses were conducted one year after initial injury.
Results: We report a rapid and transient decrease in cerebrovascular sO2 and increased hemoglobin in the impacted left cortex. Cardiac analyses showed long-term diastolic dysfunction and a diminished systolic strain response under stress. We found linear correlations between brain oxygenation immediately post-mTBI and long-term cardiac strain after 8 months (r = -0.55, p = 0.03). We also found that initial cerebrovascular hypoxia chronic cardiac systolic dysfunction correlated with long-term behavioral changes, namely decreased time spent in the center of an open field and novel object recognition, hinting to anxiety-like and memory maladaptation.
Conclusion: Experimental juvenile mTBI induces time-dependent cardiac dysfunction that corresponds to the initial cerebrovascular sO2 dip and is associated with long-term behavior. These biomarkers of the heart-brain axis could be applied to improve clinical pediatric mTBI treatment.
Biography
Dr. Pierre Sicard is a research engineer at INSERM in Montpellier (France) where he leads the ultrasound/photoacoustic platform on campus (IPAM/INSERM/ Montpellier University). Dr Sicard obtained in 2007, from the University of Burgundy, a PhD in Cardiovascular Physiology. He studied the interplay between oxidative stress, hypertension and their consequences on the heart. In 2007, he joined the cardiovascular division at St Thomas’ Hospital (King’s College London, UK) as a postdoctoral fellow to investigate the importance of p38 MAPK isoforms during experimental myocardial infarction. He continued his research in the Institute of Metabolic and Cardiovascular Diseases in Toulouse (France) in order to develop a bench to bedside project about the potential contribution of Gadd45 proteins in post-myocardial infarction remodeling. His current research interests focused on the mechanism involved in multiple-organ dysfunction after traumatic injury.

Mentoring and the Dissertation Process for faculty and postdocs, Tues., Nov. 1, 2:00-3:30 p.m. Among the most consequential documents that writers produce in graduate school is the dissertation. Strong mentoring at specific critical junctures in the dissertation process can strengthen both the document itself and graduate students’ abilities to later support their own graduate students. Unfortunately, most faculty receive little to no training about the writing instruction central to the dissertation process, from invention to final deposit. Instead, they rely upon their own experiences producing a doctoral thesis, which might have been problematic and trauma producing or wonderful yet still mystifying. This workshop incorporates current research on graduate student writing and builds on insights about graduate student struggles that the Writing Lab gains through working with dissertation writers. Faculty will be offered actionable strategies for helping dissertation writers be more effective. The workshop also will offer faculty mentors guidance on when and how to direct their students to the Writing Lab for intensive and on-going support. This workshop is for faculty and postdocs only. BME Seminar Series, Wed., Nov. 2, 9:30 a.m. MJIS 1001 or via Zoom. Xiaoqian (Joy) Wang, Assistant Professor of ECE, Purdue, will present “Fair and Explainable Machine Learning.”
Abstract: Recent advances in machine learning have spawned innovation and prosperity in various fields. In machine learning models, nonlinearity facilitates more flexibility and ability to better fit the data. However, the improved model flexibility is often accompanied by challenges such as overfitting, less interpretability, and bias. Thus, my research has been focusing on designing new feasible nonlinear machine learning models to address the above different challenges, and bringing discoveries in both theory and applications. In this talk, I will introduce a new framework to improve the interpretability and fairness of deep learning models. From the model perspective, I will discuss our novel explainable and robust models, and provide rigorous theoretical guarantees on Pareto efficiency. From the data equity perspective, I will introduce our models to achieve fair predictions while being able to protect sensitive information. Last but not least, I will discuss the application of our framework in Alzheimer's disease study.
Bio: Xiaoqian Wang is an Assistant Professor of Electrical and Computer Engineering at Purdue University. She received her Ph.D. degree from the University of Pittsburgh in 2019, and the B.S. degree from Zhejiang University in 2013. She focuses on designing novel machine learning models to improve interpretability, fairness, and robustness. She also work on the intersection of machine learning and bioinformatics, healthcare. She has published over 30 papers in top conferences and journals including NeurIPS, ICML, ICLR, AAAI, IJCAI, KDD, CVPR, RECOMB, IPMI, MICCAI, Bioinformatics, etc. She received an NSF CAREER award in 2022.

Seminars in Hearing Research, Thurs., Nov. 3, 10:30 a.m. in LYLE 1150 and via Zoom. Sarthak Mangla will present "IndivHear: An Individualized Adaptive Deep Learning-based Hearing Aid."
Abstract: Prior to coming to Purdue, motivated by his grandmother’s experience with hearing loss and hearing aids, Sarthak developed an inexpensive smartphone-based individualized adaptive hearing aid using deep learning. For patients with mild hearing loss, hearing aids have limited user benefits due to the low accessibility of professional help, limitations in fitting and diagnostic procedures of hearing devices, and the tedious process of setting up hearing aids. For patients with moderate to profound hearing loss, hearing support is inadequate due to limitations in the individualized fitting and diagnostic procedures of hearing aids, inadequacies in the functional capabilities of existing hearing aids, and limited human-machine interfaces to steer the device based on the patient’s individual needs. In an attempt to solve these problems, Sarthak created an end-to-end solution that utilizes a novel data collection approach to train individualized deep learning networks as hearing aids. Additionally, he also developed fully automated and remote versions of traditional fitting and diagnostic procedures like Pure Tone Audiometry and Speech Audiometry.

Touchdown Ventures is launching a 12-week unpaid internship program that will provide venture capital classes and hands-on experience for those interested in exploring a career path in venture capital.
The program will start in January 2023 and last approximately 12 weeks. Touchdown will accept up to 10 undergraduate students into the program’s initial cohort.
This internship is a part-time, unpaid program. The program is designed so that students will be able to participate while still meeting the obligations of their current college or university as a student.

What roads, which skills lead to success? Is it enough to be a technical expert? What else do supervisors care about? Any special insights for international students? How to move from technical positions to management positions? What does advancement look like in industry? How can one learn on the job, and how can one be strategic about advancing in one's career? If you've wondered about any of these, come listen to our panelists!
While our panelists come from Engineering backgrounds, all STEM students (especially international) are most welcome to attend!

Advocacy and mentorship are critical soft skills, and not talked about enough! To be successful, one needs not only to possess skills like these but also to be actively looking for them in one's workplace. It's great if you and your organization have 'mission alignment' but you may still need to negotiate and advocate and deal with pushback and consequences. Our panelists have valuable insights to share about workplace cultures, advocating for oneself and/or for others, and seeking and/or offering mentorship.

InnovatED - Only Two Weeks Left to Submit
The Graduate School is seeking article submissions for the 2022-23 edition of its graduate research magazine, InnovatED! InnovatED is Purdue University's premier graduate research magazine. Its mission is to showcase the influential and inspirational graduate research conducted at our university. Each publication features ten students, selected by Graduate School leadership. There are only two weeks left to submit research for publication consideration!
What's Required?
* Submissions should be in the form of a popular press piece and should consist of 700 to 800 words. To get an idea of how submissions should be formatted, students can check out the 2021- 22 edition of InnovatED.
Why Submit?
* The student author or co-authors selected will receive an award of $250 per accepted manuscript, with the cover story receiving $500.
* Students selected for publication will receive one-on-one coaching focused on communicating complex information to a lay audience.
* Research published in InnovatED is shared widely, reaching readers in more than 45 countries.

The NIH OITE is pleased to offer the RAISING A RESILIENT SCIENTIST<https://www.training.nih.gov/workshops_and_trainings_for_extramural_princip…> series for faculty, staff scientists, and administrators who mentor students and postdoctoral fellows in the biomedical, behavioral, and social sciences. A variety of topics will be covered with a specific focus on communication and shared problem-solving to help trainees deal with the inevitable setbacks experienced in high knowledge environments. The goal of the Raising a Resilient Scientist series is to promote the mental health and well-being of the academic research community by supporting faculty and administrators to develop self-management, relationship-management, and mentoring skills. There will be five units in the series and start on November 9th, 2022; each unit will consist of a 75-minute lecture followed by a 1-hour facilitated discussion with peers.
Previous participants found participation very valuable, and the tools presented practical and actionable. Here is what those who participated in series have to say:
1. “Informative workshop that I think all faculty should be required to attend. Making this a requirement for mentoring teams for fellowship and training grants would be a great approach! They need to be invested in mentoring and training, as much as training fellows.”
2. “Thank you! I've done a lot of mentoring workshops/training but feel like this one is already taking things to another level.”
3. “This course needs to be offered again and again, such an important resource in any academic curriculum.”

Abstract: The invisibility cloak in Harry Potter and the dreams of invisibility as a superpower are no longer fiction. With the invention of metamaterials, they are theoretically and experimentally possible in real life. Metamaterials — materials that are engineered to have properties that are not found in naturally-occurring materials — allow us to overcome physical and biological limitations. Scientists around the world are researching metamaterials that can be used in diverse sectors, including healthcare, optical display, and military affairs. For example, metalenses, which can exceed the physical limitations of light, may facilitate leaps in biological, biomedical, and medical applications. The development of metamaterials has just begun, but their potential is limitless. In this talk, I will give a brief overview of metamaterials and metasurfaces: principles, applications and manufacturing methods towards their science-to-technology transition.
Bio: Prof. Rho is a Mu-Eun-Jae Endowed Chair Professor and Young Distinguished Professor at Pohang University of Science and Technology (POSTECH), Korea, with a double appointment in the Department of Chemical Engineering and the Department of Mechanical Engineering. He received his Ph.D. at the University of California, Berkeley (2013), M.S. at the University of Illinois, Urbana-Champaign (2008) and B.S. at Seoul National University, Korea (2007) all in Mechanical Engineering. Prior joining POSTECH, he conducted postdoctoral research in Materials Sciences Division & Molecular Foundry at Lawrence Berkeley National Laboratory, and also worked as a principal investigator (Ugo Fano Fellow) in Nanoscience and Technology Division & the Center for Nanoscale Materials at Argonne National Laboratory. Prof. Rho has authored and co-authored more than 250 high-impact journal papers including Science, Nature, Nature Nanotechnology, Nature Materials, Nature Photonics and Chemical Reviews. He serves 13 editorial positions including Light: Science and Applications (Springer-Nature), Microsystems and Nanoengineering (Springer-Nature) and Nanophotonics (De Gruyter).

Abstract: The progress of biomedical sciences depends on the availability of advanced instrumentation and imaging tools capable of attaining the state of biological systems in vivo without using exogenous markers. Mechanical forces and local elasticity play a central role in understanding physical interactions in all living systems. We demonstrate a novel way to image microscopic viscoelastic properties of biological systems using Brillouin microspectroscopy [1]. In my talk, I will discuss the ways how an old spectroscopic tool can be used for real time microscopic imaging [2-3] and provide possible solutions to long standing problems in Life Sciences and Medicine [4-6] while advancing instrumentation beyond classical limits [7].

Special BME Seminar, Tues., Nov. 1, 11:00-12:00 p.m., MJIS 2001. Pierre Sicard, PhD, Research Engineer at Universite de Montpellier, INSERM, France will present “Cerebrovascular Hypoxia During Experimental Traumatic Brain Injury.”
Abstract
Background: Retrospective studies suggest that mild traumatic brain injury (mTBI) in pediatric patients may lead to an increased risk of cardiac events. However, the exact functional and temporal dynamics and the associations between heart-brain pathophysiological trajectories are not understood.
Methods: A single impact to the left somatosensory cortical area of the intact cranium was performed on juvenile mice 17 days postnatal. Cerebral 3D photoacoustic imaging was used to measure the oxygen saturation (sO2) in the impacted area 4 hours after mTBI followed by 2D and 4D echocardiography at days 7, 30, 90, and 190 post-impact. At 8 months, we performed a dobutamine stress test to evaluate cardiac function. Lastly, behavioral analyses were conducted one year after initial injury.
Results: We report a rapid and transient decrease in cerebrovascular sO2 and increased hemoglobin in the impacted left cortex. Cardiac analyses showed long-term diastolic dysfunction and a diminished systolic strain response under stress. We found linear correlations between brain oxygenation immediately post-mTBI and long-term cardiac strain after 8 months (r = -0.55, p = 0.03). We also found that initial cerebrovascular hypoxia chronic cardiac systolic dysfunction correlated with long-term behavioral changes, namely decreased time spent in the center of an open field and novel object recognition, hinting to anxiety-like and memory maladaptation.
Conclusion: Experimental juvenile mTBI induces time-dependent cardiac dysfunction that corresponds to the initial cerebrovascular sO2 dip and is associated with long-term behavior. These biomarkers of the heart-brain axis could be applied to improve clinical pediatric mTBI treatment.
Biography
Dr. Pierre Sicard is a research engineer at INSERM in Montpellier (France) where he leads the ultrasound/photoacoustic platform on campus (IPAM/INSERM/ Montpellier University). Dr Sicard obtained in 2007, from the University of Burgundy, a PhD in Cardiovascular Physiology. He studied the interplay between oxidative stress, hypertension and their consequences on the heart. In 2007, he joined the cardiovascular division at St Thomas’ Hospital (King’s College London, UK) as a postdoctoral fellow to investigate the importance of p38 MAPK isoforms during experimental myocardial infarction. He continued his research in the Institute of Metabolic and Cardiovascular Diseases in Toulouse (France) in order to develop a bench to bedside project about the potential contribution of Gadd45 proteins in post-myocardial infarction remodeling. His current research interests focused on the mechanism involved in multiple-organ dysfunction after traumatic injury.

Mentoring and the Dissertation Process for faculty and postdocs, Tues., Nov. 1, 2:00-3:30 p.m. Among the most consequential documents that writers produce in graduate school is the dissertation. Strong mentoring at specific critical junctures in the dissertation process can strengthen both the document itself and graduate students’ abilities to later support their own graduate students. Unfortunately, most faculty receive little to no training about the writing instruction central to the dissertation process, from invention to final deposit. Instead, they rely upon their own experiences producing a doctoral thesis, which might have been problematic and trauma producing or wonderful yet still mystifying. This workshop incorporates current research on graduate student writing and builds on insights about graduate student struggles that the Writing Lab gains through working with dissertation writers. Faculty will be offered actionable strategies for helping dissertation writers be more effective. The workshop also will offer faculty mentors guidance on when and how to direct their students to the Writing Lab for intensive and on-going support. This workshop is for faculty and postdocs only. We have immediate openings for 2-3 Postdoctoral Research Associates to work in the lab of Prof. Dipanjan Pan at Pennsylvania State University on federally funded projects. These positions are primarily concerned with designing and synthesizing nanoparticles and DNA-based nanostructures for use in multiplexed imaging and sensing of biological processes.
The ideal candidates will have published peer-reviewed papers in nanomaterials and extensive experience in the synthesis and characterization of novel materials, and in vitro and in vivo studies. In order to be considered for the position, candidates must be self-motivated and capable of working in a fast-paced team environment. Priority will be given to candidates who demonstrate scientific curiosity, passion for research, and an understanding of the research literature. Candidates must hold a Ph.D. in chemistry, materials science, biomedical engineering, electrical engineering, or a closely related field. HEADS NOTE
Hello Weldon School of Biomedical Engineering,
Thank you to all the staff, faculty, and students who represented BME at the annual BMES meeting in San Antonio, TX. I thought that Purdue and the Weldon School were very well-represented and I heard many positive comments from other heads about the quality they are seeing at Purdue University. It was an exciting time for many of our students who went to BMES for the first time for talks, posters, and for design competitions. It was also great to work closely with our colleagues at IUPUI on strategic alignment as well as have an opportunity to work together representing the overall Purdue and Indianapolis ecosystem in biomedical engineering. This week- we have the opportunity to welcome an international speaker to the BME seminar series- where she will present on microfluidics research and applications in Biomedical Engineering. Megan Yi-Ping Ho represents the first of two speakers this fall from Hong Kong. The HK quarantine restrictions have been lifted just in time to allow for an in-person visit. With this visit, and the well-attended BMES meeting, it’s great to be returning to a greater sense of “normal.” Covid-19 is still a concern, and it has not gone away, but we have better tools to fight it and be responsive to changes as needed. The NIH OITE is pleased to offer the RAISING A RESILIENT SCIENTIST<https://www.training.nih.gov/workshops_and_trainings_for_extramural_princip…> series for faculty, staff scientists, and administrators who mentor students and postdoctoral fellows in the biomedical, behavioral, and social sciences. A variety of topics will be covered with a specific focus on communication and shared problem-solving to help trainees deal with the inevitable setbacks experienced in high knowledge environments. The goal of the Raising a Resilient Scientist series is to promote the mental health and well-being of the academic research community by supporting faculty and administrators to develop self-management, relationship-management, and mentoring skills. There will be five units in the series and start on November 9th, 2022; each unit will consist of a 75-minute lecture followed by a 1-hour facilitated discussion with peers.
Previous participants found participation very valuable, and the tools presented practical and actionable. Here is what those who participated in series have to say:
1. “Informative workshop that I think all faculty should be required to attend. Making this a requirement for mentoring teams for fellowship and training grants would be a great approach! They need to be invested in mentoring and training, as much as training fellows.”
2. “Thank you! I've done a lot of mentoring workshops/training but feel like this one is already taking things to another level.”

Peters VA Medical Center, presents “Physiologic and Fine Structural Mapping of the Peripheral Nervous System for Neuromodulation of Autonomic Function in Primates.”
Abstract: Dr. Havton will present recent developments in non-human primate studies of transcutaneous spinal cord stimulation (TSCS) for functional mapping of autonomic and motor functions in pelvic organs. He will also present a new anatomical tool for the correction of varied dispersion angles and myelination in light and electron microscopy as well as a new non-binary approach for the analysis of clustering of defined nerve fiber subtypes in the peripheral nervous system. Integration of detailed morphological and physiologic data will be discussed with implications for ongoing efforts to refine neuromodulation strategies to reverse conditions.
Bio: Dr. Leif A Havton serves as Professor of Neurology and Neuroscience at the Icahn School of Medicine at Mount Sinai in New York, NY. He also serves as Staff Physician in Neurology and Principal Investigator at the Spinal Cord Injury Research Center at the James J Peters VA Medical Center in the Bronx, NY. The Havton research laboratory studies neural repair and plasticity after injury to the spinal cord, proximal nerve roots, and peripheral nerves, and the lab has a special interest in the recovery and restoration of motor and autonomic functions. Dr. Havton and his research team develop translational research models in small and large animals for the development of new therapeutics and refinement of available treatment strategies, including neuromodulation and cell-based approaches. The laboratory includes an anatomical platform for the development of new analytical tools with expertise in ultrastructural studies.

Special BME Seminar, Wed., Oct. 19, 9:30 a.m. in MJIS 1001 and via Zoom. Megan Yi-Ping Ho, PhD, Associate Professor of Biomedical Engineering at The Chinese University of Hong Kong will present “Microfluidics for Single Cell Applications.”
Abstract: Microfluidics has gathered significant interest for biomedical applications, particularly the analysis of targeted biomolecules from minute amount of samples. This presentation will summarize our efforts on exploiting microfluidics for the manipulation and analysis of individual cells and subcellular organelles. In particular, we have explored the possibility of modulating hydrodynamic shear stress at microscale to selectively disrupt cellular membrane while maintaining the mitochondrial membrane intact. We have also investigated a microfluidics-based method to encapsulate protein payloads into red blood cells by controlling membrane deformation either transiently or extendedly in a microfluidic channel. This presentation will also highlight our recent development of a novel surfactant based on fluorinated plasmonic gold nanoparticles, rendering a previously unavailable feature of photo-responsiveness in droplet microfluidics. Looking forward, we plan to study the fundamentals of how individual cells alter their phenotypes in response to environmental and therapeutic stimuli. These efforts are expected to find clinical applications in diagnostics, prognostics and treatment evaluation of infectious and chronic diseases, as well as to expand our fundamental understanding towards disease development.
Bio: Megan Yi-Ping Ho is currently an Associate Professor in the Department of Biomedical Engineering at the Chinese University of Hong Kong. She received her B.S. and M.S. in Power Mechanical Engineering from National Tsing-Hua University, Taiwan. She received her Ph.D. in Mechanical Engineering from the Johns Hopkins University in USA. After her postdoctoral training with Duke University in USA, she received the Young Elite Researcher Award from the Danish Research Council for Independent Research and started her independent career in the Interdisciplinary Nanoscience Centre and the Department of Molecular Biology & Genetics at Aarhus University in Denmark. She is a co-founder of two start-up companies situated in Demark, Zymonostics and vPCiR, focusing on enzyme-based diagnostics. She has published 67 peer-reviewed journal articles, 6 book chapters, 77 conference papers, edited 1 book and holds 2 granted patents. The results that she presented have been recognized internationally by the American Society of Gene Therapy and Controlled Release Society. Her research team is focused on developing nanosensors and microfluidics as diagnostic tools to expand the capacity of disease detection and treatment evaluation.

Abstract: Metastases are responsible for approximately 90% of all cancer-related deaths, with metastatic breast cancer (BC) holding a 5-year survival rate of only 27%. Recent research has highlighted a complex dynamic between cancer cells and the tumor microenvironment as essential for the formation of macrometastases. Within this field, tissue stiffening through matrix accumulation and altered matrix organization at the primary tumor site were recently linked with sustained proliferation and increased migration of tumor cells. Separately, elevated levels of the glycoprotein, fibronectin, were correlated to poor patient survival in BC and were linked to enhanced seeding of disseminated tumor cells at metastatic sites. Through my thesis work, we have identified several mechanisms through which accumulated fibronectin impacts the metastatic potential of BC cells. First, we identified a transient increase in extracellular fibronectin in the lungs, which peaked before overt metastasis, coupled with a non-transient increase in total lung volume. To better recapitulate physiological conditions, we then developed a novel magnetically-actuating platform with the ability to apply tensile strain on cells at various amplitudes and frequencies in a high-throughput multi-well culture plate using suspended fibrillar fibronectin for 3D cell culture that is not reliant on a synthetic substrate. Using this as a biomimetic lung model, we found that cyclic mechanical force acted as a suppressor of cancer cell growth in a biomimetic lung model, implicating the accumulation and reorganization of extracellular matrix as an attempt by the cancer cells to alter the mechanical properties of the lung tissue and resist entering dormancy. However, our results showed that BC cells could not organize extracellular fibronectin independently. Instead, BC cells altered the accumulation and architecture of fibronectin by conditioning fibroblasts through soluble factors and extracellular vesicles. We observed that the fibronectin produced by conditioned fibroblasts varied as an effect of both the method of conditioning and the phenotype of the BC cell as the conditioning source. Taken together, these results have increased our knowledge of the relationship between disseminated breast cancer cells, fibroblasts, and fibronectin architecture in the early metastatic lung niche that paves the way for further investigation on targeting disseminated BC cells during early disease intervention in order to inhibit later overt metastatic outgrowth.

Abstract: Envelope or amplitude modulation (AM) cues in a signal are critically important for the perception of complex signals, e.g., speech. Neural coding of AM can be noninvasively probed using the envelope following response (EFR), which receives contributions from both cortical (slower fluctuations, <40 Hz) and subcortical (faster fluctuations, >100 Hz) generators. As subcortical versus cortical signatures of AM representations vary across hearing loss etiologies, the EFR has great diagnostic potential. AM representations are routinely evaluated by the temporal modulation transfer function (tMTF), which is the strength of the EFR as a function of AM frequency. Currently, tMTF is measured serially for discrete sinusoidally amplitude modulated (sAM) tones. This process is time-consuming and inefficient, impeding clinical translation. Here we present a dynamically varying AM tone (dAM) used to measure a tMTF. I will analyze the tMTF obtained with and without hearing pathologies (aging, inflammation). Methods: EEG responses were obtained using sub-dermal needle electrodes in rodent models. In rodents, carrier frequencies and amplitude modulation rates varied based on species-specific differences. Tones with dynamically varying AM, the frequency of which increased exponentially from 9 Hz to 1.5 kHz over one second, and identical carrier frequencies were also used to elicit EFRs and the tMTFs were compared. Fast Fourier transforms were used to calculate discrete EFR amplitudes. A spectrally specific frequency-demodulation-based analysis was used to calculate EFR amplitudes from dAM stimuli. Results: Robust tMTFs were obtained for discrete sAM stimuli from all species tested. Preliminary results show strong tracking of dAM envelopes in rodents, which can be used to estimate the tMTF at a fine frequency resolution. These tMTFs are comparable to those obtained using discrete sAM tones. In humans, preliminary results suggest that tracking of dAM envelopes was comparable to sAM stimuli, but only at lower modulation frequencies. Ongoing analysis is aimed at refining the dAM stimuli (trajectories, timescales) to optimize AM tracking while simultaneously reducing recording times in humans. Conclusions: These results suggest that dynamically varying AM tones can be used to efficiently estimate the tMTF. However, further optimization is needed to obtain robust tMTF estimates in humans at higher AM frequencies. When combined with spectrally specific analysis, the dAM tone can be used to substantially speed up the tMTF measuring time, paving the way for potential clinical translation.

Mentoring Graduate Writers Workshop for faculty and postdocs: Tues., Oct. 25, 2:00-3:30 p.m., A good deal of research indicates that mentoring graduate students on their writing is fraught with problems. This initial workshop will share a range of best practices around mentoring and writing in the disciplines, with the hope of making mentoring graduate writers more empowered, effective, and sustainable. Purdue Quantum Science and Engineering Institute Seminar, Fri., Oct. 28, 10:00 a.m., BRK 2001 and via Zoom. Dr. Vladislav V. Yakovlev, Texas A&M University will present “Seeing life in a new light: From simple classical physics to quantum-enhanced imaging.”
Abstract: The progress of biomedical sciences depends on the availability of advanced instrumentation and imaging tools capable of attaining the state of biological systems in vivo without using exogenous markers. Mechanical forces and local elasticity play a central role in understanding physical interactions in all living systems. We demonstrate a novel way to image microscopic viscoelastic properties of biological systems using Brillouin microspectroscopy [1]. In my talk, I will discuss the ways how an old spectroscopic tool can be used for real time microscopic imaging [2-3] and provide possible solutions to long standing problems in Life Sciences and Medicine [4-6] while advancing instrumentation beyond classical limits [7].

Mentoring and the Dissertation Process for faculty and postdocs, Tues., Nov. 1, 2:00-3:30 p.m. Among the most consequential documents that writers produce in graduate school is the dissertation. Strong mentoring at specific critical junctures in the dissertation process can strengthen both the document itself and graduate students’ abilities to later support their own graduate students. Unfortunately, most faculty receive little to no training about the writing instruction central to the dissertation process, from invention to final deposit. Instead, they rely upon their own experiences producing a doctoral thesis, which might have been problematic and trauma producing or wonderful yet still mystifying. This workshop incorporates current research on graduate student writing and builds on insights about graduate student struggles that the Writing Lab gains through working with dissertation writers. Faculty will be offered actionable strategies for helping dissertation writers be more effective. The workshop also will offer faculty mentors guidance on when and how to direct their students to the Writing Lab for intensive and on-going support. This workshop is for faculty and postdocs only.
