The inaugural Grant Spotlight Series event begins on Friday, December 1, 2023 at 1:00 - 2:30 pm CT, via Zoom. This event will feature three panelists who will focus on the NIH K99/R00 funding mechanism. Our first panelist is an MD Anderson professor and reviewer of NIH K awards. The other panelists are two former MD Anderson postdocs (and current Assistant Professors at other institutions) who are K99/R00 recipients and are currently in the R00 phase of their awards. At this event, learn what reviewers look for in a successful application and how our panelists structured their career development plans and responded to critiques. Abstract:
Decision making relies heavily on the ability to distinguish a highly valued stimulus from less valued objects in the environment, such as sensing food amongst rocks. Along the process of perceiving and interpreting the sensory information, a value is assigned to each perceived stimulus such that this newly learned information can assist future goal-directed tasks. How value-modulated sensory processing evolves across cortical and subcortical regions remains poorly understood. Moreover, it is unknown how the neural representations of stimulus value changes with engagement. To answer these questions, we investigate the mouse primary somatosensory cortex (S1) and the superior colliculus (SC) while the animal is performing an active spatial discrimination task. The mice are trained to actively touch and associate a whisker-dependent stimulus with reward (GO whisker) while ignoring tactile input to the adjacent whisker (NoGo whisker). Extracellular neuronal spikes were recorded with high density extracellular silicon probes, and whisker dynamics were recorded with a high-speed camera. Periodically, task engagement was modified by temporarily removing the reward lickport so that the animal lost motivation to respond. We hypothesize that sensory information in the S1 neocortex is encoded in the form of physical features such as location, which is then converted into a map of stimulus value in the SC. We found that both S1 and SC neurons accurately discriminated between adjacent whisker stimuli with the SC displaying a much stronger preference than S1 for the higher-valued stimulus. Unique to SC, many neurons displayed a bidirectional response profile, where spike rate increased during reward-whisker touch, yet it decreased during NoGo whisker touch. Moreover, pre-stimulus baseline activity of SC neurons was strongly modulated by task engagement and in some cases was able to predict upcoming trial performance, suggesting a direct role in initiating goal-directed action. On completion, this study will expand on the model of how value-modulated sensory information is encoded across cortical and subcortical depths, improving our understanding of goal-directed behavior.

ABSTRACT:
Animal behavior displays inherent bilateral coordination between the left and right sides of the body. However, the neural mechanisms governing the integration of information across hemispheres to generate a unified perceptual experience remains poorly understood. In this proposal, we address this question by recording activity from the cerebral cortices during goal-directed behavior. Due to lack of ipsilateral touch responses in the primary somatosensory cortex(S1), naïve models assume bilateral integration to happen primarily in higher cortical areas. However, since tactile behaviors necessitate high temporal resolution, we propose that S1, fortified by robust callosal projections from the contralateral hemisphere, likely plays a crucial role in bilateral integration and in tuning stimulus information in downstream regions. To tackle this question, we developed a novel Go/No-go bilateral discrimination task where mice discriminate between bilateral stimuli categories, compelling them to share tactile information across hemispheres. We observed that mice enhance the bilateral whisker movement symmetry during 'Go' stimulus presentation. Examination of neural activity revealed strong temporal coupling between the S1s along with enhanced ipsilateral facilitation modulated by behavioral context. Our findings challenge existing models and propose a novel framework suggesting that behavioral context governs bilateral integration in the sensory cortex. Additionally, neurons in the whisker motor cortex (wMC) were also found to accurately decode sensory and motor variables during the bilateral task. By innovative optogenetic axon silencing of specific feedforward projection neurons, we aim to dissect the circuits underlying this brain-wide computation. Completing this proposal will provide insights on the circuits and cell-types underlying bilateral sensation and movement.

Student Eligibility

* Nominees must be outstanding engineering PhD students.
* The following criteria will be used to rank the nominees:
* Merit. Includes publications, conference presentations, GPA etc.
* Funding need.
* Good citizen of the School and College. Includes service.
* Career promise. Includes leadership, innovation, readiness and potential to make an impact.
Application Package Should Include the following in one PDF:
* A letter (limit 2 pages) from the major professor nominating the student and confirming that the student's expected graduation date. This can be sent separately if the student is providing the rest of the information.
* The student's thesis abstract,
* A copy of the preliminary examination paperwork (to confirm the student is on track to graduate next year),
* The student's CV,
* A copy of the student's most recent transcript - graduate work only.
Salary & Usage:
* This fellowship includes $30,000 annual salary, tuition remission, medical insurance premium contribution, and coverage of most fees. Engineering differential fee is not covered.
* Bilsland Fellowships are administered as assistantships. It is expected that job duties will contribute to the completion of the student's dissertation.
* Bilsland's must be awarded in the student's last term, and the student is expected to graduate at the end of the fellowship term.
* This year's Bilsland allocation may be used Summer 2024, Fall 2024, Spring 2025, or Summer 2025.
* No additional work appointments may be added, but faculty may (and perhaps must; depending on our minimum salary requirement which may be set in BME) increase the salary or provide additional funds as scholarships.
* The fellowship is to be awarded for the full 12 months or until the student graduates, whichever comes first.
* Fellowship recipients who graduate before the completion of the fellowship or otherwise stop the fellowship before the full 12 month duration forfeit any remaining funds.
* If Bilsland Fellows do not graduate as expected this could hurt our funding allocation from the Graduate School, so please only nominate students you truly expect to graduate and are truly in their final year.

INTERIM HEAD'S NOTE
Welcome back. I hope you had a great Thanksgiving holiday. We have three more weeks in this semester, including the finals week. Coming down the stretch, everyone is busy. So please take care of yourself and your students both mentally and physically, especially as we finally start to get cold weather.
In this Friday's faculty meeting, we will talk about academic programs and teaching in general. The purpose is to share information and start these discussions critical to our department.
* Vitaliy will present an overview of our academic programs, UG curriculum, teaching load, and the use of lecturers and professors of engineering practice.
* To supplement, Tami Kinzer-Ursem and Milind Kulkarni will present an overview of graduate and professional programs and online programs at the college level, respectively. I feel these are the areas where we see room to improve for the department.

As of Mon., Nov. 27: The Club Room on the 1st floor of MJIS will be CLOSED indefinitely. A recent inspection by REM revealed numerous sanitation issues (food debris, uncleaned appliances, evidence of mice, etc.). As this is a recurring problem, they have shut the room down until it can be deep cleaned with an assurance that it will be maintained to university standards in the future. Stay tuned. Re: final exam accommodations. We have a number of students who may need accommodations for final exams and faculty need to be proactive in scheduling with the Testing Center if they plan to use that.
Below is some important information instructors should know if they choose to have Purdue Testing Services (PTS) administer their accommodated exams.
* The steps to submit a Testing Instruction Form (TIF) for finals week are the same as any other exam with one notable difference. In the "select the exam group/s for this test" field, be sure to select your course that starts with Accommodated FINAL Exam. This is required before students will be able to schedule during finals week.
*If you had already submitted a TIF for your final prior to receiving this email, PTS staff updated your TIF to the correct exam group
* Hours of operation:
* Monday - Friday | 8 a.m. - 10 p.m.
* Exam start times: 8 a.m., 10:30 a.m., 1 p.m., 3:30 p.m., 6 p.m. and 7 p.m.
* Saturday | 8 a.m. - 4:30 p.m.
* Exam start times: 8 a.m., 10:30 a.m., Noon and 1 p.m.
* Due to the volume of exams we administer, we will not start any exam outside of the start times mentioned above. Please take this into consideration if you are adding time restrictions to your TIF.
* The deadline for students to schedule their final exams with PTS is Dec. 1, 2023. Students who miss the deadline will be redirected to their instructors to discuss alternate arrangements to still receive their accommodations.
* There may be times when PTS reaches full capacity prior to the scheduling deadline. If this occurs, instructors will either be asked to select a different day/start time in which PTS has seats available or they will need to make alternate arrangements to provide the accommodations.

The Bilsland Dissertation Fellowship provides support for outstanding PhD candidates in their final year or semester of doctoral degree completion. The goal of the fellowship is to allow exceptional candidates to focus exclusively on finishing their dissertation. * Nominees must be outstanding engineering PhD students.
* The following criteria will be used to rank the nominees:
* Merit. Includes publications, conference presentations, GPA etc.
* Funding need.
* Good citizen of the School and College. Includes service.
* Career promise. Includes leadership, innovation, readiness and potential to make an impact.
Application Package Should Include the following in one PDF:
* A letter (limit 2 pages) from the major professor nominating the student and confirming that the student's expected graduation date. This can be sent separately if the student is providing the rest of the information.
* The student's thesis abstract,
* A copy of the preliminary examination paperwork (to confirm the student is on track to graduate next year),
* The student's CV,
* A copy of the student's most recent transcript - graduate work only.
Salary & Usage:
* This fellowship includes $30,000 annual salary, tuition remission, medical insurance premium contribution, and coverage of most fees. Engineering differential fee is not covered.
* Bilsland Fellowships are administered as assistantships. It is expected that job duties will contribute to the completion of the student's dissertation.
* Bilsland's must be awarded in the student's last term, and the student is expected to graduate at the end of the fellowship term.
* This year's Bilsland allocation may be used Summer 2024, Fall 2024, Spring 2025, or Summer 2025.
* No additional work appointments may be added, but faculty may (and perhaps must; depending on our minimum salary requirement which may be set in BME) increase the salary or provide additional funds as scholarships.
* The fellowship is to be awarded for the full 12 months or until the student graduates, whichever comes first.
* Fellowship recipients who graduate before the completion of the fellowship or otherwise stop the fellowship before the full 12 month duration forfeit any remaining funds.
* If Bilsland Fellows do not graduate as expected this could hurt our funding allocation from the Graduate School, so please only nominate students you truly expect to graduate and are truly in their final year.

Excellence in Teaching Awards Competition
Applications are now being accepted for the Midwestern Association of Graduate Schools (MAGS) 2024 Excellence in Teaching Awards Competition. The University is eligible to nominate one masters-level and one Ph.D.-level student for each award that includes a citation and a $750 honorarium that will be presented at the MAGS annual meeting on April 3-5, 2024. Interested students must submit their application materials to the Graduate School by Friday, December 1, 2023, at 5:00 p.m. for consideration.

Three Minute Thesis Competition
Nominate a graduate student to represent your academic unit!
The Graduate School's annual Three Minute Thesis competition is scheduled for April 9th! To increase academic representation in our 3MT® competition, we are asking that West Lafayette graduate faculty members nominate talented graduate student(s) to represent their academic units.
Nominees must be current Purdue graduate students in the final stages of their research. Each nominee will be contacted by Graduate School staff and encouraged to register for the competition. Nominations are not required for students to submit a draft video; they are simply intended to help the Graduate School identify and encouraged talented students to participate.

What is 3MT®?
Three Minute Thesis (3MT®) is a research communication competition that helps graduate students develop academic, presentation, and research communication skills so that they can effectively explain their research to a non-specialist audience. During each competition, graduate students will have three minutes to present a compelling discussion on their research topic, including its significance and relevance, to the public. This is a fast-paced competition where the top 10 finalists compete by summarizing their two to three-plus years of research in only three minutes with only one slide.
The cash awards for this year's competition are $5,000 for first place, $3,000 for second place, and $2,000 for the People's Choice Award winner. The first-place winner will also be invited to compete in the Midwestern Association of Graduate School's (MAGS) annual 3MT® competition.

Abstract: The development of wearable devices that monitor biometric signals in real-time to aid in maintaining health has played a crucial role in the field of biomedical engineering. In our rapidly evolving world, chronic diseases such as hypertension, diabetes, obesity, and heart diseases have imposed severe health and cost implications on public health. This has led to an increased demand for smart healthcare that can remotely and accurately measure and monitor long-term period. While wearable sensing technologies have made significant advancements, tracking specific diseases in real-time remains a challenging task. To address these challenges, we present a series of innovative wearable devices, each uniquely designed to tackle specific aspects of these health concerns. The first is a machine learning-enabled smart neckband designed to precisely monitor dietary intake for patients with diabetes and obesity. This device can effectively distinguish between various activities like body movements, speech, fluid intake, and food intake, showcasing a comfortable and ergonomically designed solution. Another contribution is a custom-designed wearable device focused on the real-time monitoring and classification of vocal cord dysfunction, efficiently differentiating between body movements and vocal cord closures. Extending beyond human health, we also investigate the healthcare of animals. Communication barriers often hinder the timely diagnosis of symptoms in animals. Addressing this, we introduce a wearable device for large animals, such as horses. This device is centered on monitoring respiration in real time, a critical tool for managing conditions like asthma.

Sarah Wiehe, MD, MPH, the Jean and Jerry Bepko Professor of Pediatrics and Associate Dean of Community and Translational Research at the Indiana University School of Medicine will present "Community-engaged research: A path to meaningful work as a translational scientist."
Abstract: In this talk, I will share my career path as a physician-scientist trained in epidemiology which took a turn for the better after facing recruitment challenges with my first funded project. I will present my research tracking adolescent girls using GPS-enabled cell phones (back when we had blackberry pearls) and discuss what I learned in terms of the benefits of stakeholder engagement and human-centered design research. I will also share how this led to additional interdisciplinary research and leadership opportunities as part of the Indiana CTSI.
Biography: Sarah Wiehe, MD, MPH is the Jean and Jerry Bepko Professor of Pediatrics and Associate Dean of Community and Translational Research at Indiana University School of Medicine. She is a Research Scientist at the Regenstrief Institute and Adjunct Professor of Epidemiology at Fairbanks School of Public Health at Indiana University. Dr. Wiehe co-directs the Indiana Clinical and Translational Sciences Institute (CTSI) and leads its community engagement program. The Indiana CTSI serves researchers and partners with patients and community members, community-based organizations, healthcare systems, and governmental agencies to translate scientific evidence into practice and policy. Dr. Wiehe's research focuses on addressing health equity outcomes among vulnerable populations, including individuals involved with the justice system, living in poverty, diagnosed with mental health and substance use disorder diagnoses, experiencing trauma, and living with HIV. She has expertise in the community engagement, patient-centered research methods, creative use of existing data, data-sharing partnerships, and health services research. Her work has been funded by the National Institute of Health, the Patient-Centered Outcomes Research Institute, and the Robert Wood Johnson Foundation. As a general pediatrician, Dr. Wiehe enjoys seeing patients at Eskenazi Health Center, the largest Federally Qualified Health Center network in Indianapolis, Indiana, and supervising and training pediatric residents. Dr. Wiehe earned her bachelor's degree in economics and biology and her medical degree from the University of Chicago. She completed her internship and residency in pediatrics and her master's in public health in epidemiology at the University of Washington, where she completed a postdoctoral fellowship with the Robert Wood Johnson Clinical Scholars Program.

Abstract: The wearable industry is experiencing a significant upward trajectory, marked by a diverse array of technological applications. These applications span a wide spectrum, encompassing real-time health monitoring, human-machine interfaces, and more. However, a prevailing sentiment persists - that being "wearable isn't always truly wearable." This sentiment emerges due to the prevailing issues within the current market offerings of wearable sensors, which tend to be bulky and rigid leading to uncomfortable user experiences, artifacts in motion tracking, and compromised data accuracy. As a response to this challenge, a global and vigorous pursuit of innovative materials and structures has been ignited. This preliminary report aims to offer strategies towards "truly wearables" in focusing on the pivotal role of material and structural innovation. This preliminary report presents a comprehensive set of material innovations and structural designs to develop mechanically "soft materials" tailored for wearable applications. In the first part of this preliminary report, emphasis is placed on the material design aspect, on the development of a "Rapid self-healing hydrogel with ultralow electrical hysteresis for wearable sensing." The second part introduces another material design, highlighting the creation of a "Tough conductive organohydrogel for wearable sensing in extreme environmental conditions." The third section delves into structural design considerations, focusing on the development of a "Spongy Ag foam for soft and stretchable strain gauges."

Abstract: Computational modeling has a broad range of applications in neuroscience, from discovering fundamental attributes of neuronal synaptic function to helping develop tools to fight neurodegenerative disorders. Here, we utilize computational modeling to investigate the dynamics of receptor phosphorylation that are important in regulating dynamic changes in synaptic strength. Within synapses, a thick network of proteins makes up a membrane-less structure called the post-synaptic density (PSD) in post-synaptic dendritic spines. The increased number and phosphorylation of glutamate receptors within the PSD is a hallmark of increased synaptic strength. A-kinase anchoring proteins (AKAPs) are important scaffolding proteins in the PSD that have been established to localize a number of regulatory enzymes such as protein kinase A, calcineurin, and phosphodiesterases near the cell membrane and glutamate receptor tails. However, it is still unclear what is their overall effect on glutamate receptor phosphorylation and trafficking. Here, a spatial stochastic computational model of AKAP and enzymatic binding partners in the PSD is developed and the dynamics of the system analyzed. Model results demonstrate the correlation between glutamate receptor tail phosphorylation and AKAP localization. By computational "knockdown" of AKAPs we are able to quantify their effect on glutamate receptor phosphorylation, investigate the role of spatial localization, and posit their potential impacts on LTP and synaptic plasticity.

Abstract: Groundbreaking research has been done on smart contact lenses, especially as a minimally invasive diagnostic platform and a drug delivery system. The eyes provide rich physiological and broad diagnostic information, making contact lens sensors a promising disease diagnostic tool. A smart contact lens, one such contact lens sensor, can quantify the concentrations of various biomolecules and electrolytes in ocular fluids and physical biomarkers of the eye, such as intraocular pressure (IOP). Continuous and non- or minimally invasive contact lens sensors can be directly utilized in a clinical or point-of-care setting. Here, we introduce the incorporation of contact lens sensors and their fabrication, sensitivity, power source, and readout mechanisms. The incorporation is to monitor glaucoma and conditions associated with eye health, such as dry eye syndrome and inflammation. The contact lens sensors fit seamlessly across different corneal curvatures and thicknesses in human eyes. The lens can also wirelessly capture the intraocular pressure by utilizing the principle of the mechanical deformation of the microfluidic channel under ambulatory conditions. Also, we will discuss the colorimetric ocular electrolyte tear fluid sensor to detect the color change of dry eye disease and inflammation. We assessed the in vitro biocompatibility, ex vivo functionality, and in vivo safety of the soft contact lens sensor.

Abstract: Cochlear implant (CI) speech-recognition outcomes are influenced by various combinations of bottom-up and top-down factors. As such, there is substantial individual variability in CI outcomes. The purpose of this project is to assess elliptical speech as a tool for evaluating bottom-up perceptual acuity in CI users. Noise-vocoded sentence stimuli were presented to 10 normal-hearing (NH) listeners. Stimuli consisted of IEEE sentences that either maintained correct consonant place-of-articulation cues (Intact Speech) or contained consonant "ellipses" that ambiguated spectral cues to consonant place (Elliptical Speech). The "ellipses" replaced each key-word consonant with a new consonant maintaining the same manner and voicing of the original, but with an altered place feature. A concurrent neuroimaging measurement was conducted to estimate cortical activity in bilateral auditory cortex and dorsolateral prefrontal cortex via functional near-infrared spectroscopy (fNIRS). This project tested our hypotheses that: 1) NH participants will successfully detect ellipses in more favorable listening conditions but will be unable to detect ellipses under more degraded listening conditions, and 2) listeners will demonstrate differences in cortical activity in response to Intact vs Elliptical speech depending on the severity of the signal degradation. Preliminary results demonstrate that the detection of ellipses in speech is impacted by the spectral resolution of the signal, and this result is also reflected in listeners' cortical activity. These results have important implications for CI users whereby elliptical speech is a potential tool for isolating the quality of the bottom-up input from top-down processing of speech.

Abstract: Nucleic acid testing has become a prominent method for rapid microbial detection. Unlike polymerase chain reaction (PCR), loop-mediated isothermal amplification (LAMP) is a simple method of nucleic acid amplification where the reaction can be performed at a constant temperature and the output provided in a colorimetric format. A transparent water bath heater is a desirable instrument to perform the heating and observe the visual results of nucleic acid amplification. However, existing methods of heating the water are not convenient for loading and unloading the nucleic acid samples. Here, we developed a field-deployable water bath heating device-an isothermal heater called IsoHeat for short-which is solely dedicated to performing LAMP reactions and can heat the water up to 85 °C (if needed). Using 3D-printing and LASER-cutting technology, we fabricated different parts of the device and mechanically assembled the parts to develop the entire device. Users can commence the heating by pressing the start button on the screen after entering the target temperature. Subsequently, the device heats up the water bath and maintains the target temperature through a PID algorithm-based control system. We demonstrate that IsoHeat can operate in environmental temperatures ranging from 5-33 °C and it can conduct LAMP reactions in a liquid format as well as in paper-based devices. IsoHeat is more efficient and user-friendly compared to a commercially available immersion-heating device, which is often used to perform LAMP reactions. This newly developed device would be helpful to detect pathogens conveniently in the field (e.g., at the point-of-care for human applications, on farms for plant and animal applications, and in production facilities for food safety applications).

Abstract: The vagus nerve (VN) is a central nervous system (CNS) that mediates most of the
parasympathetic functions of the autonomic nervous system. The axons of the human VN comprise a mix of unmyelinated and myelinated axons, where ~80% of the axons are unmyelinated C fibers (Havton et al., 2021). Understanding that the majority of VN axons are unmyelinated, there is a need to map the pathways of these axons to and from organs to understand their individual function(s) and whether C fiber morphology or signaling characteristics yield insights into their functions. The development of a machine learning model that detects and predicts the morphology of VN single fiber action potentials based on select fiber characteristics, e.g., diameter, myelination, and position within the VN, allows us to more readily categorize the nerve fibers with respect to their function(s). Additionally, the features of this machine learning model could help inform peripheral neuromodulation devices that aim to restore, replace, or augment one or more specific functions of the VN that have been lost due to injury, disease, or developmental abnormalities. We designed and trained four types of Multi-layer Perceptron Artificial Deep Neural Networks (MLP-ANN) with 10,000 rat abdominal vagal C-fibers simulated via the peripheral neural interface model ViNERS. We analyze the accuracy of each MLP-ANN's SFAP predictions by conducting normalized cross-correlation and morphology analyses with the ViNERS C-fiber SFAP counterparts. Our results showed that our best MLP predicted over 94% of the C-fiber SFAPs with strong normalized cross-correlation coefficients of 0.7 through 1 with the ViNERS SFAPs. Overall, this novel tool can use a C-fiber's biophysical characteristics (i.e. fiber diameter size, fiber position on the x/y axis, etc.) to predict C-fiber SFAP morphology.

Abstract: Cardiovascular disease remains the leading cause of death worldwide, highlighting the importance of improving cardiac imaging for early disease detection and treatment. While 3D and 4D imaging methods offer enhanced precision, their complexity necessitates accessible applications in research and clinical settings. This thesis introduces an advanced 4D kinematic analysis to evaluate Duchenne muscular dystrophy (DMD) cardiomyopathy, a progressive neuromuscular disorder with no cure, affecting approximately 1 in 5000 boys. Despite varied progression, the lack of standard imaging biomarkers impedes early detection or rapid-progressing DMD cardiomyopathy. Our novel 4D cardiac magnetic resonance imaging (CMR) analysis paradigm aims to comprehensively map left-ventricular kinematics, introducing innovative imaging biomarkers and computational methods for earlier and more accurate diagnosis of DMD. Our focus on myocardial strain biomarkers provides insights into early disease onset and progression, paving the way for multi-center studies to enhance treatment outcomes.

Abstract: Traumatic brain injury (TBI) is a worldwide health issue. Increasing prevalence of blast-induced TBI (bTBI), a predominantly combat-related injury, is an alarming trend necessitating a better understanding of the associated pathogenesis to develop treatments. Further, most TBI/bTBI injuries are mild and undiagnosed, permitting secondary biochemical injuries to propagate beyond possible intervention. While a link between TBI/bTBI and chronic neurodegenerative diseases, such as Alzheimer's disease (AD), has been established, the mechanisms are not yet understood. Animal model studies have identified key pathologies, however their spatial and temporal resolution limits detailed investigations into cellular and molecular mechanisms underlying these complex injuries. Additional investigative tools are urgently needed to elucidate the mechanisms behind this immediate and long-term damage. While most in vitro methodologies offer the resolution required, few can visualize bTBI-induced cellular damage during injury, and subsequent biochemical injury. Therefore, we introduce "bTBI-on-a-Chip," an in vitro blast injury model, capable of simultaneous morphological, biochemical, and bioelectrical assessments during and after injury, with a series of targeted experiments to address this unmet need. We hypothesize that primary blast injury and secondary biochemical injury can be recapitulated in vitro for mechanistic studies into trauma-induced neurodegeneration. Further, we hypothesize that synaptic loss following bTBI can be demonstrated in our system and mitigated by pharmacological intervention, further linking bTBI and AD, and providing key insights into possible prevention strategies. Our goal is to facilitate translational discoveries by studying the mechanisms of bTBI to not only establish early diagnosis criteria, but suggest possible treatments to curtail post-trauma neurodegeneration.

Abstract: Neuronal recording technologies have propelled neuroscience forward, facilitating the unraveling of neural functions across various spatiotemporal scales. Characterizing the membrane potential is crucial to understanding the brain's electrical activities and relationship with the structure and function of neural networks. However, scaling these recordings to measure across single neurons and nervous tissue simultaneously and integrating the measurements across various spatiotemporal scales are critical limitations. The recording limitations primarily result from the physical stress placed on delicate cell structures, preventing repeated membrane disturbance. Then, these limitations lead to the inherent incompatibility among various recording methods concerning their spatiotemporal scales. To address these challenges, we propose a novel application of DNA nanostructure as a versatile platform capable of simultaneously gathering and processing data from two representative recording techniques: patch-clamp electrophysiology and voltage-sensitive dye imaging. The engineered DNA nanodevice can be precisely inserted into live cell membranes using a patch pipette. Once inserted, the DNA nanopores create a synthetic ion channel for facilitating the acquisition of ground truth voltage measurements from individual neurons. Also, the incorporation of single Quantum Dots (QDs) within the DNA nanopore, allows for the recording of voltage signals from large populations of neurons. My research objectives are to (1) develop a reusable DNA nanopore to minimize membrane damage during successive recordings and (2) enhance signal detection by embedding QDs within the DNA nanopore, amplifying the Quantum-confined Stark Effect (QCSE). Achieving these aims will introduce an innovative method to harmonizing data acquisition from individual neurons through patch-clamp electrophysiology, which provides a reliable ground truth for voltage measurement in synthetic nanopores. Simultaneously, it will enable the concurrent recording of multiple neurons using QDs, ushering in an unprecedented era of multifaceted neuronal data acquisition. The expected outcomes of this work will enhance our comprehension of neuronal signaling processes, minimize disruptions to the cell membrane integrity for accurate interpretation of synaptic potential, and introduce an innovative approach to simultaneous and precise recording of neuronal signals across diverse spatiotemporal scales.

Abstract: Studying, treating, and mitigating the symptoms of Alzheimer's Disease and other Neurodegenerative Disorders is a big challenge due to the low Blood-Brain Barrier (BBB) permeability, which is highly selective on what can pass into the brain. In the proposed study, I aim to address the lack of a safe, efficient, and reliable brain drug delivery vehicle by optimizing the design of Tetrahedral DNA Nanomaterials (TDNs) to effectively deliver drugs through the BBB. Starting with two classical TDNs structures, I alter the design by changing the size, structure (one or two double-stranded DNA helices on the edges), and attached DNA aptamers for the receptor-medicated endocytosis pathway. I will study the influence of these factors on cellular uptake, stability of TDNs, toxicity, and potential delivery efficiency. I plan on testing the materials in vitro using primary mice cortical cells and human astrocytes Isogenic-induced Pluripotent Stem Cells, as well as in the human BBB in vitro model. To study the biodistribution and tissue toxicity, I will perform in vivo tests of the most promising TDNs designs in the C57BL/6 wild-type mouse model, harvest the tissues, and run histology analysis with Hematoxylin and Eosin Y stains. The TDNs concentration will be measured by using incorporated into the DNA strands cyanine 3 and 5 dyes fluorescence and the stability will be estimated by Fluorescence Resonance Energy Transfer measurements. After this work, I will have developed an effective, exceptionally biocompatible, and stable nanomaterial with great potential applications in drug delivery through BBB, brain imaging, and gene therapy.

Postdoctoral Fellow Opportunity - IUSM Emergency Medicine
The laboratory of Dr. Nathan J. Alves, PhD, within the Department of Emergency Medicine at the Indiana University School of Medicine (IUSM), is seeking a highly motivated and independent post-doctoral research fellow. Dr. Alves is a Chemical and Biomolecular Engineer, Associate Professor and the Director of Translational Research for the Department of Emergency Medicine in addition to having an affiliate faculty appointment at Purdue University in Biomedical Engineering. The Alves Lab explores highly interdisciplinary research interests applying engineering and biophysical principles and designs to create translational technologies and treatments in the area of blood coagulation and fibrinolysis. The primary focus of research in the laboratory is to develop targeted clot digesting therapeutic agents to more safely digest clinically relevant blood clots to treat pulmonary embolism (PE, blood clots in the lungs), deep vein thrombosis (DVT), and ischemic stroke. This interdisciplinary research utilizes diverse enzyme assays, chemical synthesis, blood clot formation and digestion assays under shear, and other novel testing/delivery platforms that include multivalent branched molecules and nanoparticle drug delivery systems. Projects Include: (1) Leveraging multivalency to modulate enzyme activity and improve inhibitor selectivity (nanoparticle drug delivery and targeting). (2) Ex-vivo flowing models of clot digestion under shear using in-vivo like fluorescently labeled blood clots from human whole blood. (3) Development of novel fibrinolytic diagnostics tools to assess coagulation state. (4) Mouse models of clot formation/targeting/digestion utilizing intravital microscopy. (5) Study the effects of environmental microplastic exposure on coagulation/fibrinolysis. Projects are funded both externally and internally.

Abstract:
Traumatic brain injury (TBI) is a worldwide health issue. Increasing prevalence of blast-induced TBI (bTBI), a predominantly combat-related injury, is an alarming trend necessitating a better understanding of the associated pathogenesis to develop treatments. Further, most TBI/bTBI injuries are mild and undiagnosed, permitting secondary biochemical injuries to propagate beyond possible intervention. While a link between TBI/bTBI and chronic neurodegenerative diseases, such as Alzheimer’s disease (AD), has been established, the mechanisms are not yet understood. Animal model studies have identified key pathologies, however their spatial and temporal resolution limits detailed investigations into cellular and molecular mechanisms underlying these complex injuries. Additional investigative tools are urgently needed to elucidate the mechanisms behind this immediate and long-term damage. While most in vitro methodologies offer the resolution required, few can visualize bTBI-induced cellular damage during injury, and subsequent biochemical injury. Therefore, we introduce “bTBI-on-a-Chip,” an in vitro blast injury model, capable of simultaneous morphological, biochemical, and bioelectrical assessments during and after injury, with a series of targeted experiments to address this unmet need.
We hypothesize that primary blast injury and secondary biochemical injury can be recapitulated in vitro for mechanistic studies into trauma-induced neurodegeneration. Further, we hypothesize that synaptic loss following bTBI can be demonstrated in our system and mitigated by pharmacological intervention, further linking bTBI and AD, and providing key insights into possible prevention strategies. Our goal is to facilitate translational discoveries by studying the mechanisms of bTBI to not only establish early diagnosis criteria, but suggest possible treatments to curtail post-trauma neurodegeneration.

Abstract: Computational modeling has a broad range of applications in neuroscience, from discovering fundamental attributes of neuronal synaptic function to helping develop tools to fight neurodegenerative disorders. Here, we utilize computational modeling to investigate the dynamics of receptor phosphorylation that are important in regulating dynamic changes in synaptic strength. Within synapses, a thick network of proteins makes up a membrane-less structure called the post-synaptic density (PSD) in post-synaptic dendritic spines. The increased number and phosphorylation of glutamate receptors within the PSD is a hallmark of increased synaptic strength. A-kinase anchoring proteins (AKAPs) are important scaffolding proteins in the PSD that have been established to localize a number of regulatory enzymes such as protein kinase A, calcineurin, and phosphodiesterases near the cell membrane and glutamate receptor tails. However, it is still unclear what is their overall effect on glutamate receptor phosphorylation and trafficking. Here, a spatial stochastic computational model of AKAP and enzymatic binding partners in the PSD is developed and the dynamics of the system analyzed. Model results demonstrate the correlation between glutamate receptor tail phosphorylation and AKAP localization. By computational "knockdown" of AKAPs we are able to quantify their effect on glutamate receptor phosphorylation, investigate the role of spatial localization, and posit their potential impacts on LTP and synaptic plasticity.
