CURE – Purdue seeks to develop course-based undergraduate research experiences (CUREs) across the Purdue West Lafayette campus as a way to increase undergraduate students’ overall access to novel research. The program provides instructors with the time and framework for designing a CURE, either by transforming an existing course or developing a new one. CUREs have a number of features and positive outcomes, including increasing the overall capacity for undergraduate research by operating at a larger scale than the traditional apprenticeship model and increasing inclusion by allowing any eligible student to enroll. Research findings suggest additional benefits to instructors: * Greater enjoyment when teaching CUREs over a traditional course. * Ability to integrate their research and teaching. * Advancement of their research agenda. * Recruiting students to their personal research group. * Potential to positively impact promotion and tenure. Rita Patel, PhD, CCC-SLP, Associate Professor and Speech Language Pathologist, Department of Otolaryngology, Head and Neck Surgery, Indiana University School of Medicine, will present “An interdisciplinary scientific journey.” Abstract: Voice pathologies occur across the life span, are disabling, interfere with gainful employment and compromise quality of life. According to the National Institute on Deafness and Other Communication 7.5 million people in the United States have voice disorders. It has been estimated that over 1 million or 6-9% children suffer from dysphonia nationwide. Estimates indicate that anywhere from 3-9% of the population has some type of voice dysfunction at any given point in time. Reported occurrence of hoarseness in the school-aged children ranges from 6%-23%. Dysphonia can be detrimental both psychologically and academically, hence early identification and restoration of optimal vocal health is critical. If untreated, dysphonia can substantially increase health care costs. Unfortunately, efficient diagnosis and effective management of voice disorders is challenged by difficulty accessing professionals with the necessary expertise and the availability of valid instrumentation to capture highly aperiodic voice qualities to diagnose and treat voice problems efficiently. To address these issues, technological advancements from a collaborative team of interdisciplinary experts in the fields of medicine, engineering, physics, and psychology show great promise for improving the efficiency and effectiveness of voice disorders assessment and treatment. This talk will focus on my current interdisciplinary work for advancing clinical assessment of voice disorders across the lifespan in the areas of laryngeal imaging, computational fluid dynamics, and mobile technology. Biography: Dr. Rita Patel is an associate professor at the Department of Speech, Language and Hearing Sciences, Indiana University and an adjunct associate professor at the Department of Otolaryngology, Head and Neck Surgery Indiana University School of Medicine. Her research and clinical expertise focus on addressing voice and upper airway disorders in adult and pediatric population, with specialization in endoscopic laryngeal imaging. Dr. Patel has been awarded 8 extramural grants from National agencies including ASHA (American Speech Language Hearing Association) and NIH (National Institutes of Health); and 5 intramural grants. In 2022, Dr. Patel was awarded fellow by the American Speech-Language-Hearing Association (ASHA) for her outstanding contributions to research and excellent teaching in the area of voice and voice disorders. Currently, Dr. Patel serves as the editor for the American Journal of Speech-Language Pathology, American Speech-Language-Hearing Foundation’s Board of Trustees, and as an Advisory Board Member of Voice Foundation. Christian Franck, Bjorn Borgen Professor, Panther Program Director, and Director of the Center for Traumatic Brain Injury at the University of Wisconsin-Madison will present “Cellular NeuroMechanics-Concussions, Traumatic Brain Injury and the Mysterious Havana Syndrome.” Abstract: Current prediction, prevention and diagnosis strategies for mile traumatic brain injuries, including concussions, are still largely in their infancy due to a lack of detailed understanding and resolution of how physical forces give rise to tissue injury at a cellular level. In this talk I will present some recent work on our current understanding of the origin of concussions and traumatic brain injuries and how cells in the brain interpret and react to the physical forces of trauma. Specifically, I will show that the path to a better understanding of traumatic injuries involves addressing a variety of finite deformation, rate-dependent soft matter and cell mechanics problems along the way. Finally, I will provide an update on how our current understanding of the cellular neuromechanics cannot only help shed light on improving our prediction of TBI but also enable us to dissect the physical origin of emerging injuries such as the Havana Syndrome. Biography: Christian Franck is a mechanical engineer specializing in cellular biomechanics and new experimental mechanics techniques at the micro and nanoscale. He received his B.S. in aerospace engineering from the University of Virginia in 2003, and his M.S. and Ph.D. from the California Institute of Technology in 2004 and 2008. Dr. Franck held a post-doctoral position at Harvard investigating brain and neural trauma. He was an assistant and associate professor in mechanics at Brown University from 2009 - 2018, and is now the Bjorn Borgen Professor in Mechanical Engineering at the University of Wisconsin-Madison. His lab at the University of Wisconsin-Madison has developed unique three-dimensional full-field imaging capabilities based on multiphoton microscopy and digital volume correlation. Current application areas of these three-dimensional microscopy techniques include understanding the 3D deformation behavior of neurons in the brain during traumatic brain injuries, and the role of non-linear material deformations in soft matter. He is the acting director of the Center for Traumatic Brain Injury at the University of Wisconsin-Madison and the ONR-funded Physics-based Neutralization of Threats to Human Tissues and Organs (PANTHER) program, which consists of over 24 PIs nationwide. Key objectives of the Panther program are in better detection, prediction, and prevention of traumatic brain injuries by providing accelerated translation from basic science discovery to civilian and warfighter protection solutions. Title: Novel Microfluidic Device for In-Field Purification, Amplification, and Detection of DNA from Mycotoxin-Producing Fungi. Advisory Committee: J. Paul Robinson, Chair; Euiwon Bae; Jacqueline Linnes Abstract: Mycotoxins are toxic molecules produced by fungi and are harmful to human health when ingested. Mycotoxins produced by fungi that infect food crops can enter the food stream directly in manufactured produces (foods like bread) or indirectly through animal feed. Detection of mycotoxins is commonly performed after harvesting and processing of the crops because mycotoxins can increase in concentration during these steps. However, if farmers could detect the toxin-forming fungi prior to harvesting, growers can reduce cross-contamination, save wasted resources used in harvesting and processing of infected crops, and increase food safety and security. Fusarium graminearum is a fungal species that infects cereal crops and produces deoxynivalenol (DON), a harmful mycotoxin called vomitoxin. Because Fusarium graminearum has a large economic impact in the United States and globally on wheat and barley markets, it was selected as a proof-of-concept for the detection assay. For detection of fungus to be useful in determining infected areas of fields, the device must be inexpensive and portable. A low-cost, portable microfluidic device was designed to both extract the fungal DNA from wheat samples and perform a fluorescence-based loop-mediated isothermal amplification (LAMP) assay to detect the fungal DNA. Contaminated wheat samples were used to demonstrate the effectiveness of the device. The device was able to detect Fusarium graminearum in the wheat samples at concentrations relevant to DON concentration limits set forth by the U.S. Food and Drug Administration (FDA). Detection of multiple mycotoxin-producing fungal species is possible through the addition of more LAMP assays for multiplexing specific target genes. Thesis Committee: Dr. Sherry Harbin (advisor), Dr. Carmella Evans-Molina (clinical co-advisor), Dr. Hyowon Lee, Dr. Adrian Buganza Tepole, Dr. Luis Solorio Abstract: Diabetes, a chronic condition characterized by elevated blood glucose levels, arises when pancreatic β-cells lose capacity to produce a robust, dynamic glucose-stimulated insulin secretion (GSIS) response. Accurate measurement of β-cell health and function ex vivo is thus fundamental to diabetes research, including studies evaluating disease mechanisms, novel drug candidates, and replacement β-cell populations. However, present-day dynamic GSIS assays typically represent end-point measurements, involve expensive commercial perifusion machines, and require time-consuming enzyme-linked immunosorbent assays (ELISA) for insulin detection. Microfluidic devices developed as accessible, low-cost alternatives still rely on secondary ELISAs and suspend islets in liquid medium, limiting their survival in vitro. Here, we present a novel, 3D-printed microphysiological system (MPS) designed to recreate components of in-vivo microenvironments through encapsulation in fibrillar type I collagen and restoration of favorable molecular transport conditions. Following computational-informed design and rapid prototyping, the MPS platform sustained collagen-encapsulated mouse islet viability and cytoarchitecture for 5 days and supported in situ measurements of dynamic β-cell function. To rapidly detect insulin secretion from β-cells in the MPS, we then developed a highly sensitive electrochemical sensor for zinc (Zn2+), co-released with insulin, based on glassy carbon electrodes modified with bismuth and indium and coated with Nafion. Finally, we validated sensor detection of Zn2+ released from glucose-stimulated INS-1 β-cells and primary mouse islets, finding high correlation with insulin as measured by standard ELISA. Together, the 3D MPS and Zn2+ sensor developed in this dissertation represent novel platforms for evaluating dynamic β-cell function in a low-cost, user-friendly, and physiologically-relevant manner. Until recently, the growth and shrinkage of microtubules was thought to be confined to their ends. Over the last few years, however, evidence has emerged that exchange of tubulin in and out of microtubules can take place within the lattice and not just at the ends. I will present our recent work on the microtubule severing protein spastin on microtubule dynamics as well as measurements of the forces necessary to remove tubulin from the lattice.
