Accurate measurement of β-cell health and function ex vivo is thus fundamental to diabetes research, including studies evaluating disease mechanisms, novel drug candidates, and replacement β-cell populations. However, present-day dynamic GSIS assays typically represent end-point measurements, involve expensive commercial perifusion machines, and require time-consuming enzyme-linked immunosorbent assays (ELISA) for insulin detection. Microfluidic devices developed as accessible, low-cost alternatives still rely on secondary ELISAs and suspend islets in liquid medium, limiting their survival in vitro. Here, we present a novel, 3D-printed microphysiological system (MPS) designed to recreate components of in-vivo microenvironments through encapsulation in fibrillar type I collagen and restoration of favorable molecular transport conditions. Following computational-informed design and rapid prototyping, the MPS platform sustained collagen-encapsulated mouse islet viability and cytoarchitecture for 5 days and supported in situ measurements of dynamic β-cell function. To rapidly detect insulin secretion from β-cells in the MPS, we then developed a highly sensitive electrochemical sensor for zinc (Zn2+), co-released with insulin, based on glassy carbon electrodes modified with bismuth and indium and coated with Nafion. Finally, we validated sensor detection of Zn2+ released from glucose-stimulated INS-1 β-cells and primary mouse islets, finding high correlation with insulin as measured by standard ELISA. Together, the 3D MPS and Zn2+ sensor developed in this dissertation represent novel platforms for evaluating dynamic β-cell function in a low-cost, user-friendly, and physiologically-relevant manner.

Paul Robinson, Chair; Euiwon Bae; Jacqueline Linnes Abstract: Mycotoxins are toxic molecules produced by fungi and are harmful to human health when ingested. Mycotoxins produced by fungi that infect food crops can enter the food stream directly in manufactured produces (foods like bread) or indirectly through animal feed. Detection of mycotoxins is commonly performed after harvesting and processing of the crops because mycotoxins can increase in concentration during these steps. However, if farmers could detect the toxin-forming fungi prior to harvesting, growers can reduce cross-contamination, save wasted resources used in harvesting and processing of infected crops, and increase food safety and security. Fusarium graminearum is a fungal species that infects cereal crops and produces deoxynivalenol (DON), a harmful mycotoxin called vomitoxin. Because Fusarium graminearum has a large economic impact in the United States and globally on wheat and barley markets, it was selected as a proof-of-concept for the detection assay. For detection of fungus to be useful in determining infected areas of fields, the device must be inexpensive and portable. A low-cost, portable microfluidic device was designed to both extract the fungal DNA from wheat samples and perform a fluorescence-based loop-mediated isothermal amplification (LAMP) assay to detect the fungal DNA. Contaminated wheat samples were used to demonstrate the effectiveness of the device. The device was able to detect Fusarium graminearum in the wheat samples at concentrations relevant to DON concentration limits set forth by the U.S. Food and Drug Administration (FDA). Detection of multiple mycotoxin-producing fungal species is possible through the addition of more LAMP assays for multiplexing specific target genes.

In the coming weeks, several of the IUPUI BME professors will give their research seminars in our seminar series. Dr. Young Kim has worked tirelessly on this. While technological innovations such as the adoption of minimally-invasive surgery and the introduction of surgical robots have transformed surgery since the 1970s, postoperative complications remain the third largest contributor to death worldwide. In this presentation, I will demonstrate how Artificial Intelligence-based image recognition and data analysis methods can contribute to the reduction of such complications. Combining routinely available clinical parameters with imaging data and video recordings of minimally-invasive pancreatic and colorectal surgeries, machine learning-based pre- and intraoperative complication risk stratification models were developed. These models could facilitate tailored surgical approaches considering individual cancer patients’ complication risk profiles. Biography Since 2019, Fiona Kolbinger is a surgical clinician scientist at the University Hospital, the National Center for Tumor Diseases and the Else Kroener Fresenius Center for Digital Health in Dresden, Germany. Her research focus is the intersection of medicine and computer science, as well as the translation of technological innovations, in particular Artificial Intelligence, into clinical practice. Based on automated analysis of medical imaging such as radiological, endoscopic, or microscopic images, she aims to contribute to improving clinical outcomes through more individualized interdisciplinary treatments and better risk and outcome stratification in patients with cancer. Fiona graduated from the University of Heidelberg medical school in 2019 and subsequently completed a research doctorate at the German Cancer Research Center Heidelberg, Germany. Dr. Kolbinger is a prospective research faculty member and will be hosted by Professor Young Kim. BME Distinguished Seminar Series, Wed., Feb. 15, 9:30 a.m., MJIS 1001 and via Zoom. Ken Yoshida, PhD, Professor of Biomedical Engineering, Indiana University-Purdue University Indianapolis will present “Neuromodulatory Electrical Stimulation Beyond the Pulse.” Abstract: The rectangular pulse waveform is arguably the dominant, standard, waveform used to stimulate bioelectrically active tissues. It has a broad range of applications that include functional electrical stimulation (FES), neuromuscular electrical stimulation (NMES), transcutaneous electrical nerve stimulation (TENS), cardiac pacing, sensory electrical stimulation (SES), etc. The mechanism has been well characterized in it's >300 year history. For example, the empirical Weiss-Lapicque strength duration relationship was described over a hundred years ago. Pulse stimulation has a biophyscial property that it preferentially activates large caliber myelinated nerve fibers. The consequence is that it leads to an order of nerve fiber utilization that does not align with the natural utilization of nerve fibers. As such, pulse stimulation is known to have an inverse nerve fiber recruitment property. Normalizing the order of recruitment through electric blocking techniques is particularly important in bioelectronic medicines which the target the small caliber myelinated/unmyelinated nerve fibers of the autonomic nervous system. In late 2016, our efforts in attempting to block nerve activity using kilohertz frequency alternating (kHFAC) block led to an accidental discovery and a new modality that efficiently blocked nerves using low frequency sinusoids that I named low frequency alternating current block (LFACb). In 2018, in another experiment aiming to maximize LFAC block we discovered that the waveform at higher amplitudes activated nerve fibers that led to strong, fatigue resistant muscle contractions. It is a phenomenon we named LFAC activation (LFACa). In-silico models and subsequent in-vivo experiments show that LFAC block and activation have interesting properties. It blocks without onset activation and activates nerve fibers in a size-wise order. LFAC presents itself as a promising new modality for subtractive and additive neuromodulation. Biography: I was born and raised in Los Angeles, CA. I received my BS in engineering (biocybernetics) from UCLA in 1989 after spending my first 2 undergraduate years at UCSD and transferring in my junior year. Following my undergraduate degree, I left for graduate work at the University of Utah where I received my PhD in 1994 based upon research developing an advanced peripheral nerve based closed loop functional electrical stimulation (FES) system. My postdoctoral work, December 1994 to April 1998, was at the Division of Neuroscience, University of Alberta, Edmonton, Canada with Richard B. Stein, the eminent Canadian motor control neurophysiologist, and inventor of the regeneration and cuff electrodes. In April 1998, I joined the Center for Sensory-Motor Interaction (SMI), Aalborg University, Denmark as a research assistant professor and worked with Thomas Sinkjær, the center head. In 2000, I was promoted to Assistant Professor and became a tenure-track faculty member at Aalborg University. Until recently, the growth and shrinkage of microtubules was thought to be confined to their ends. Over the last few years, however, evidence has emerged that exchange of tubulin in and out of microtubules can take place within the lattice and not just at the ends. I will present our recent work on the microtubule severing protein spastin on microtubule dynamics as well as measurements of the forces necessary to remove tubulin from the lattice. First, we have officially retired the G.S. Form 37, which was previously used to request post-facto edits to a deposited thesis or dissertation. Instead, students requesting this or any other exception to Graduate School policy should submit a letter justifying the request endorsed by major professor and the head/chair of the graduate program to the Graduate School office at your respective campus (see Section VII.I<https://catalog.purdue.edu/content.php?catoid=15&navoid=18605#theses> of the University Catalog). Please see attached Bottorff Fellowship application form for Summer 2023. The Bottorff Graduate Fellowship Program is designed to increase the number of Purdue engineering graduate students focused on medical technology innovation and clinical translation. Funds to support a summer clinical immersion experience are available to help students who are still in the early stage of their research gain clinical exposure at the Indiana University School of Medicine (IUSM). This full-time 8 to 10-week summer clinical immersion experience will be followed by a fall class focused on biomedical product translation. Finally, a spring Regulatory class will help fellows understand the workings of the US FDA and other regulatory agencies. CURE - Purdue seeks to develop course-based undergraduate research experiences (CUREs) across the Purdue West Lafayette campus as a way to increase undergraduate students' overall access to novel research. The program provides instructors with the time and framework for designing a CURE, either by transforming an existing course or developing a new one. Goergen; Michael Markl Abstract: 4D flow magnetic resonance imaging (MRI) has emerged as an imaging modality capable of assessing cardiovascular flow non-invasively. Despite interest in using these in vivo measurements for cardiovascular disease diagnostics and progression assessment, there are no adequate models of 4D flow MRI's measurement error. Error models are essential in evaluating accurate hemodynamic biomarkers and judging the quality of 4D flow MRI scans. Here, we present a comprehensive error model of 4D flow MRI velocity measurements using fluid dynamics and imaging physics principles. 4D flow MRI velocity error is described as a multivariate normal distribution which is a function of space and time. The error distribution's scale is assessed using the principle of conservation of mass, and error correlations are assessed using the novel standardized difference of means (SDM) velocity metric. We define the SDM velocity as the difference between the local mean velocity and the global background mean velocity relative to the standard error of the local mean velocity. We also demonstrate the SDM velocity's utility in segmenting vessels in 4D flow MRI measurements. This work also introduces a bias error model, which accounts for insufficient spatial resolution and partial volume effects. These bias error estimates serve as one means of validating our proposed 4D flow MRI error model. All algorithms presented are validated in 4D flow MRI measurements in scaled in vitro cerebral aneurysms and in vivo 4D flow measurements. This work ultimately supports the development of reliable biomarkers indicative of cardiovascular disease progression. Yiyang Gong, Assistant Professor of Biomedical Engineering, Duke University will present "Traditional and computational development of fluorescence imaging tools for neuroscience" Abstract: Modern basic research in neuroscience aims to dissect neural activity from genetically identifiable neuron populations within dense neural circuits. Such research has relied increasingly on tools that perform optical imaging and optical stimulation. In this area, my lab develops a diverse toolbox that employs multiple engineering disciplines simultaneously. In this talk, I will highlight our recent gains in protein engineering, optical microscopy, and data analysis. The protein tools will attempt to enable multi-channel optical experiments by increasing spectral separation between fluorescent protein sensors and actuators. The microscopy tools will attempt to support fast volumetric imaging experiments that accurately capture neural activity. The data analysis tools will attempt to find neurons and calculate their neural activity from the large datasets resulting from optical imaging experiments quickly and accurately. Lastly, this talk will touch on the future of the protein engineering pipeline seen through the lens of protein tools in neuroscience. Bio: Yiyang Gong obtained his B.S. in Electrical Engineering from the California Institute of Technology. He then obtained his M.S. and Ph.D. in Electrical Engineering from Stanford University, training with Prof. Jelena Vuckovic on plasmonic and photonic crystal optical devices. He trained with Prof. Mark Schnitzer at Stanford University as a postdoc in Biology, developing genetically encoded voltage sensors that are advancing the current generation of neuroscience experiments. While technological innovations such as the adoption of minimally-invasive surgery and the introduction of surgical robots have transformed surgery since the 1970s, postoperative complications remain the third largest contributor to death worldwide. In this presentation, I will demonstrate how Artificial Intelligence-based image recognition and data analysis methods can contribute to the reduction of such complications. Combining routinely available clinical parameters with imaging data and video recordings of minimally-invasive pancreatic and colorectal surgeries, machine learning-based pre- and intraoperative complication risk stratification models were developed. These models could facilitate tailored surgical approaches considering individual cancer patients' complication risk profiles. Biography Since 2019, Fiona Kolbinger is a surgical clinician scientist at the University Hospital, the National Center for Tumor Diseases and the Else Kroener Fresenius Center for Digital Health in Dresden, Germany. Her research focus is the intersection of medicine and computer science, as well as the translation of technological innovations, in particular Artificial Intelligence, into clinical practice. Based on automated analysis of medical imaging such as radiological, endoscopic, or microscopic images, she aims to contribute to improving clinical outcomes through more individualized interdisciplinary treatments and better risk and outcome stratification in patients with cancer. Fiona graduated from the University of Heidelberg medical school in 2019 and subsequently completed a research doctorate at the German Cancer Research Center Heidelberg, Germany. Until recently, the growth and shrinkage of microtubules was thought to be confined to their ends. Over the last few years, however, evidence has emerged that exchange of tubulin in and out of microtubules can take place within the lattice and not just at the ends. I will present our recent work on the microtubule severing protein spastin on microtubule dynamics as well as measurements of the forces necessary to remove tubulin from the lattice. And yet, it continues to be an ever-present part of people's identity, especially those working within the high stress environment of biomedical research. In this webinar we will define perfectionism and better understand the toll it takes on our lives, explore the contributing factors that perpetuate it, and identify strategies to cultivate a healthy striving mindset. The healthy striving mindset serves an alternative to the impossibility of perfectionism which is eroding our mental health and our resilience. Immediately following the 1-hour webinar will be a 45-minute small group discussion led by a trained facilitator. There will also be an hour-long supplemental session on February 16th focused on growth mindset, a set of skills necessary to work toward interrupting unhelpful patterned responses of perfectionism. The webinar will run 3:00-4:00pm EST and will be followed by small discussion groups facilitated by an OITE wellness advisor from 4:05-4:50pm EST. If you are missing your knit cap with a pom-pom on top, it was found on the stairs in MJIS (between the 1st and 2nd floor). Thought you might need it, with the current cold weather.
