Title: The Pathological Role of Acrolein Scavenging and ALDH2*2 in EAE Mice Date: July 22,, 2024 Time: 12:00pm Location: DLR 131 Committee: Dr. Riyi Shi (Major) Adam Kimbrough Elsje Pienaar Jessica Fortin Abstract: Multiple Sclerosis (MS) is a severely debilitating, demyelinating disease with no known cure. Acrolein, a reactive aldehyde, has recently been implicated in the pathology of neurodegenerative diseases, including MS, for its ability to directly degrade myelin. Therefore, major efforts have been expended to determine if acrolein could serve as a viable therapeutic target in MS, as current treatments only provide symptomatic relief without significantly altering the disease pathology. Our lab has demonstrated that several acrolein scavengers, such as hydralazine, dimercaprol, and phenelzine, are effective in alleviating MS symptoms in an animal model, experimental autoimmune encephalomyelitis (EAE) mice. In addition to exogenous applications, we tested the administration of Alda-1, a drug that activates an endogenous acrolein-scavenging enzyme, ALDH2. Alda-1 application also demonstrates a significant alleviation of EAE symptoms. To further understand the role of acrolein in EAE, we look at a clinically relevant genotype in mice, ALDH2*2, a human enzymatic deficiency suffered by millions. In this genotype, ALDH2 efficiency is dramatically reduced compared to healthy individuals. Thus, we hypothesize that ALDH2*2 EAE mice will suffer dramatically worse symptoms than wild-type EAE controls. As suspected, we have observed significant deficits in motor and behavioral symptoms in addition to an increase in several pathological hallmarks of MS, including inflammation and demyelination, in ALDH2*2 EAE mice compared to their wild-type counterparts. Taken together, our data provides additional support that acrolein plays a critical role in MS pathology and suggests that ALDH2 potentiation could serve as an effective treatment, particularly in individuals with ALDH2*2 deficiency.

Weldon family, Yesterday, July 1, marked my first official day, and it has been a truly invigorating start. I am filled with gratitude and enthusiasm for our Weldon team, who have welcomed me with incredible warmth and eagerness. The appreciation I feel is accompanied by a sense of profound awe. I feel fortunate to lead the Weldon School as we embark on our Good to Great journey. Last Thursday, I visited Indianapolis, where the anticipation for the future is palpable. In preparation for starting and over the last few days I've met and talked with many wonderful team members. I look forward to connecting with all of you soon. However, the specific molecular processes underlying this aggregation are not fully understood, mirroring the limitations seen in understanding the etiology of other prion-like neurodegenerative diseases. There are proposed mechanisms connecting alpha-synuclein aggregation to endocytic processes involving the escape and retention of fibrillar seeds. Additionally, intracellular protein-membrane interactions may also play a role. However, effective methods to probe the evolution of aggregation states with sufficient sensitivity in the context of these cellular processes are lacking. We used a custom-built fluorescence lifetime imaging microscope to monitor the evolution of seeded aggregation in primary neurons in the context of endocytic processes that have not been well explored. This was enabled by measuring self-quenching-induced fluorescence lifetime changes of alpha-synuclein-fluorophore fusion proteins, providing a sensitive aggregate detection method. Our results indicate that both escape and retention of fibrillar seeds from endocytic compartments are seeding pathways for aggregation. In addition, we developed a novel imaging scheme using fluorescence lifetime measurements of tethered Förster resonance energy transfer (FRET) reporters to probe membrane-induced alpha-synuclein aggregation. Employing this method in neurons enabled us to decipher which intracellular membrane surfaces likely play a role in alpha-synuclein aggregation. Fluorescence lifetime imaging enabled insights into the underlying mechanisms of alpha-synuclein aggregation, which has broader applications to other prion-like neurodegenerative diseases. Qazi Abstract: Pancreatic ductal adenocarcinoma (PDAC), the most common type of pancreatic cancer, is a highly metastatic cancer that has a low survival rate. It is histologically characterized by a thick desmoplastic stroma. Counterintuitively, pancreatic cancer cells can still manage to survive in such a restrictive environment and even metastasize to distant organs. Over the years, efforts have been made to find out the mechanisms underlying these perplexing behaviors. However, questions about the role of ECM accumulation and enhanced stiffness in pancreatic cancer cell dissemination remained unanswered. In this dissertation, we aim to advance the material design for tumor modeling, and propose an explanation for the malignant cell behavior in the PDAC tumor microenvironment (TME). This is achieved through the use of hydrogel-based tumor models that recapitulate the elevated stiffness of the tumor tissue. Specifically, hydrogel stiffness was tuned to mimic the PDAC TME to understand how pancreatic cancer cells and cancer-associated fibroblasts (CAFs) respond to various substrate stiffnesses temporally. Next, we employ bio-orthogonal click chemistries to create hydrogels with on-demand stiffening capabilities, as well as hyaluronic acid deposition in the hydrogel, to investigate the effect of dynamic change in matrix stiffness and composition on PDAC cells and CAFs. Lastly, by leveraging thiol-norbornene, aldehyde-hydrazide, and tetrazine-norbornene click chemistries, we created a microporous hydrogel system with a conformation that combines both the advantage of 3D cell culture and the non-restricting nature of 2D cell culture. Additionally, the system allows the application of modularized user-defined factors, including, but not limited to stiffness and hyaluronic acid (HA) deposition to the system. Stiff gel in 2D facilitated cell spreading of Pa03C in the presence of CAF. Despite being more restrictive on cell spreading, stiff gelatin gel in 3D induced cytokines that promote matrix remodeling and spreading cell morphology can be restored by stiffening with HA. Overall, this dissertation demonstrated that ECM component (i.e., HA), culture dimensionality, and cell-cell interaction play a huge role in cell behavior, and these factors may interact with each other and result in synergistic effects. Postdoctoral Fellow Opportunity - IUSM Emergency Medicine The laboratory of Dr. Nathan J. Alves, PhD, within the Department of Emergency Medicine at the Indiana University School of Medicine (IUSM), is seeking a highly motivated and independent post-doctoral research fellow. Dr. Alves is a Chemical and Biomolecular Engineer, Associate Professor and the Director of Translational Research for the Department of Emergency Medicine in addition to having an affiliate faculty appointment at Purdue University in Biomedical Engineering. The Alves Lab explores highly interdisciplinary research interests applying engineering and biophysical principles and designs to create translational technologies and treatments in the area of blood coagulation and fibrinolysis. The primary focus of research in the laboratory is to develop targeted clot digesting therapeutic agents to more safely digest clinically relevant blood clots to treat pulmonary embolism (PE, blood clots in the lungs), deep vein thrombosis (DVT), and ischemic stroke. This interdisciplinary research utilizes diverse enzyme assays, chemical synthesis, blood clot formation and digestion assays under shear, and other novel testing/delivery platforms that include multivalent branched molecules and nanoparticle drug delivery systems. Projects Include: (1) Leveraging multivalency to modulate enzyme activity and improve inhibitor selectivity (nanoparticle drug delivery and targeting). (2) Ex-vivo flowing models of clot digestion under shear using in-vivo like fluorescently labeled blood clots from human whole blood. (3) Development of novel fibrinolytic diagnostics tools to assess coagulation state. (4) Mouse models of clot formation/targeting/digestion utilizing intravital microscopy. (5) Study the effects of environmental microplastic exposure on coagulation/fibrinolysis. Projects are funded both externally and internally.
