Title: Understanding the Risk of Type 2 Diabetes on Alzheimer's Disease Date: Thursday, March 6th Time: 3:00pm Location: Hybrid via MJIS 2001 and Zoom (https://purdue-edu.zoom.us/j/5554725555<https://nam04.safelinks.protection.outlook.com/?url=https%3A%2F%2Fpurdue-ed…>) Committee Members: Dr. Doug Brubaker (Co-Chair), Dr. Deva Chan (Co-Chair), Dr. Tamara Kinzer-Ursem, Dr. Elizabeth Proctor, Dr. Brendan Lucey Abstract: Individuals with type 2 diabetes (T2D) are at major risk of developing Alzheimer's disease (AD) during their lifetime. Despite this connection, there is lack of understanding on the biological mechanisms involved in disease pathology. To improve our understanding, I conducted a series of studies with increasing complexity to interrogate the T2D-AD axis. These investigations included an in vitro neuronal culture model stimulated by AD- and T2D-associated metabolites, a computational mouse-to-human translational framework using brain tissue of different mouse disease models, and a cross-disease model that synthesized peripheral blood data of T2D and AD patients. There were overarching themes shared across these studies. I first identified immune signaling mechanisms shared across T2D and AD as demonstrated by differential cytokine responses of neuronal cells and significant inflammatory pathways from gene set enrichment analysis. I next demonstrated that T2D mouse models could predict human AD outcomes better than AD or ADxT2D mouse models, which identified a metabolic component to AD pathology. From cross-disease modeling, I identified blood-based biomarkers of people with T2D that could distinguish between AD and control groups. A panel of these biomarkers were also found to reflect alterations in multiple brain regions, opening the possibility of blood-based testing for AD. As a result of this work, I established a translational modeling platform that integrates human demographic features for data synthesis through cross-species or cross-disease modeling. The keynote speaker, Dr. Kristina Deligiannidis (MD), a psychiatrist who uses advanced MRI methods in her studies, has been the leader of a large clinical trial that got the first medication for postpartum depression FDA approved. Dr. Deligiannidis is a nationally recognized leader in the field of perinatal depression and in novel therapeutics research. She served as a PI on the clinical trials that led to the FDA approval of two rapid-acting antidepressants for postpartum depression. Her keynote will be followed by talks of several outstanding recipients of WGHI pilot grants, on reproductive health and policy (LaRoche), sexual health intervention (Brown), and breast cancer high-risk monitoring (Tao) and metastasis (Teegarden). "Bridging the Gap: Translational Research and Product Development for ECM-based Bioimplants" [A person smiling at the camera Description automatically generated] Beth Young Sports Medicine Product Manager at LifeNet Health Abstract: Extracellular matrix (ECM)-based biomaterials, especially xenograft scaffolds and allograft tissues, have gained significant traction in the healthcare marketplace for tissue repair and regeneration. However, translating novel biomaterials and manufacturing techniques into clinical practice faces substantial challenges and often lags behind advancements in academic research. To bridge this gap, intentional industry collaborations and a clear understanding of commercialization strategies can facilitate the translation of research. Key elements to consider include unmet needs, surgeon preferences, and patient outcomes. As Biomedical Engineers, we play a crucial role in translating this science through effective communication, educating regulatory bodies, and conducting relevant foundational research to promote technological adoption and prevent industry stagnation. In this seminar, Dr. Young will discuss emerging trends in ECM-derived bioimplants and share insights from her career path, which has spanned academia, start-up companies, and the commercialization of orthopedic and cardiovascular allograft-based products. She will demonstrate how consistent user input and a well-defined regulatory and market strategy from the ideation phase can set the technology up for success. Biography: Dr. Bethany Young is a Sports Medicine Product Manager at LifeNet Health in Virginia Beach, Virginia. LifeNet Health is a global leader in transplant solutions, providing a wide range of services, from organ transplantation and clinical biologics to human-based life science applications. Dr. Young's work in clinical biologic product development focuses on tissue engineering of allografts for orthopedic and cardiovascular applications, maximizing the gift of donation. Her career has evolved into a strategic role, where she guides product development from the bench, leveraging her expertise in directing product design by gathering user input and translating it into engineering requirements. Prior to joining LifeNet Health, Dr. Young served as a Senior Development Engineer at Tympanogen, a Richmond-based otologic medical device startup. There, she led the development and regulatory efforts for new technologies, including their flagship product, the Perf-Fix Otologic Gel Patch. Perf-Fix is the first nonsurgical gel patch designed for eardrum repair and will be the only product on the market for this procedure to receive approval for pediatric use. Dr. Young holds a B.S. in Biology from the University of Richmond. She also earned her doctorate and master's degrees in Biomedical Engineering from Virginia Commonwealth University, specializing in pulmonary tissue engineering and biomaterial design. Good Afternoon, All Purdue University faculty, staff, and postdoctoral scholars now have access to Proofig AI, which is a tool that can be used to identify potential image-related research integrity violations in research publications. The tool can be accessed by selecting "Sign Up" at https://www.proofig.com/<https://nam04.safelinks.protection.outlook.com/?url=https%3A%2F%2Fwww.proof…>, and choosing "Purdue" under Institutional Sign In. Proofig uses Purdue's single sign-on capability. Departments are strongly encouraged to use the Proofig AI software to check theses and dissertations that have life science images including microscopies (confocal, light, fluorescence, and electron), histology slides, pathology slides, Western blot bands, gel electrophoresis, flow cytometry (FC), fluorescence-activated cell sorting (FACS), cell culture, in-vitro, and in-vivo. in addition to the required iThenticate review.
