BME WEEKLY ROUNDTABLE

INTERIM HEAD'S NOTE
We are about to enter August. This signifies the first month of me being the Interim Head. I have sat in many meetings with our BME colleagues, college-wide colleagues, and staff at the Dean's Office. I have had a couple of onboarding meetings. So far, so good. I feel I have an understanding of most aspects of School affairs by now. Of course, experiences are critical to successfully managing the School operations. In the past few weeks, one priority has been to discuss the committee assignments. Last week, we announced that Dr. Andrew Brightman would assume the role of Director of Graduate Programs and Chair of the Graduate Committee in the Weldon School. Andrew's seasoned leadership and expertise will be crucial to maintain the excellence of our graduate programs and navigating any challenges that may arise during a period of transition within our School. As Director of the Graduate Programs, he will oversee the continued improvement of our graduate and professional programs, including the integration of the Indianapolis and West Lafayette sites.
I am hopeful the assignments for both Undergraduate and Graduate Committees will be completed by the first week of August. Thank you for those who have volunteered and/or helped strategize the personnel assignments.

In honor of this year's Watanabe Awardee, Dr. Zoghbi, our annual meeting theme will focus on translational neuroscience. Thus we will be showcasing the Indiana CTSI's cutting-edge research in neuroscience and related topics from across all our campuses. Fri., Sept. 8: Abstract Submission Deadline for The Trauma THOMPSON Challenge. The primary goal of The Trauma THOMPSON Challenge is to encourage research to build better algorithms for autonomous instruction systems in uncontrolled and austere environments, especially for life-saving procedures. The envisioned algorithms include action recognition, action anticipation, procedure recognition, and visual question answering (VQA). The challenge participation details are below.

Abstract: Single-molecule localization microscopy (SMLM) leverages the detection and localization of individual fluorescently tagged molecules to visualize cellular and tissue samples at a nanometer resolution. The reconstruction of a super-resolution image via SMLM, however, involves intricate algorithms that may require hours to achieve a single high-quality 3D image. Additionally, the image generation process calls for specialized expertise and resources, such as knowledge of image processing and massively parallel computing. To address these challenges, we are developing an integrated cloud-based SMLM suite named "cloud-SMLM". This suite combines key algorithms from each stage of single molecule analysis, integrating with high levels of parallelization and modularization. Deployable on cloud-based computational services like Amazon Elastic Compute Cloud (Amazon EC2), cloud-SMLM interacts with its users through a dedicated, easy-to-use drag-and-drop webpage. This design eliminates the prerequisite for hardware such as GPU and reduces the knowledge barrier for users of single molecule nanoscopy. Accessible from any web-browser equipped device, it enables users to initiate and modify data reconstruction. Furthermore, it provides visualization and data mining tools for users to explore the reconstructed three-dimensional ultra-high-resolution cellular and tissue volumes. We expect that this project will democratize existing and new single molecule super-resolution imaging capacities by integrating them into computational pipelines for a wide range of biological model systems. We believe that the success of cloud-SMLM will represent a significant step towards unlocking the full potential of ultra-high resolution optical imaging in biological and biomedical research.

Real-Time Dietary Monitoring Using Wearable Technology: An Approach for Managing Chronic Diseases such as Obesity and Diabetes
Abstract: The growing incidence of chronic diseases has drawn considerable societal attention toward devising effective strategies for their management. Among the various approaches, dietary control holds a key position, especially for diseases requiring strict management of food intake. The obesity epidemic has grown to affect a significant fraction of the U.S. population, including one-fifth of children and over a third of adults. The adverse health effects of uncontrolled obesity are far-reaching. It raises the risk of heart disease, type 2 diabetes, and certain types of cancer, thereby leading to substantial medical expenses. The need for careful dietary management in combating obesity cannot be overstated. Dietary habit management also plays a crucial role in controlling diabetes, a condition often linked with obesity. Effective diabetes management requires constant monitoring of blood glucose levels, which are directly impacted by the type and amount of food consumed. Given this background, there's an urgent need for innovative solutions to monitor and manage the dietary habits of patients with obesity and diabetes. Such a solution could aid in real-time monitoring of dietary habits, facilitate weight loss, manage blood glucose levels effectively, and ultimately, help reduce the societal costs associated with the treatment of obesity and diabetes. To address this requirement, a custom-designed wearable device has been developed. This device is designed to monitor dietary habits in real-time, offering an innovative and practical solution. It promises to play a vital role in obesity and diabetes management by enabling personalized diet modifications and providing valuable insights for effective disease control.

Auditory Deficits of Mild Blast-induced Traumatic Brain Injury (bTBI)
Abstract: Traumatic brain injury (TBI) is an increasing casualty among civilians and military personnel. TBIs are estimated to affect 69 million civilians annually, and approximately 18,500 service members were affected in 2022. These numbers are not expected to decrease in the coming years. TBI is a complex pathology comprised of an acute mechanical injury phase followed by a chronic phase characterized by metabolic and biochemical changes. Injuries are classified as mild, moderate, and severe based on neurological examination and neuroimaging; however, misdiagnosis is common since symptoms are hard to detect. Objective neuroimaging techniques only rule out life-threatening injuries (i.e., intracranial lesions or hemorrhage) in mild and moderate TBIs-failing to detect subtle, but important, structural abnormalities. Hearing loss is often associated with TBI since trauma tends to affect the areas responsible for auditory processing in the peripheral and central nervous system, especially in blast-related injuries given the intense sound levels experienced. Although the high susceptibility of the auditory system to mild and moderate brain injury can be exploited as an avenue to develop sensitive diagnostics of TBI, the underlying mechanisms of injury at the peripheral and central level remain unknown. Using a chinchilla animal model, this project will elucidate the effects of mild blast-induced TBI on the peripheral and early subcortical central auditory system in order to identify potential auditory evoked responses and blood biomarkers to diagnose TBI.

Abstract: The term neurovascular unit (NVU) indicates a brain-specific multi-cellular assembly controlling brain homeostasis for physiological neuronal transmission in networks. The blood-brain barrier (BBB) within the NVU is the microvascular interface that governs the exchanges between the peripheral blood and the brain parenchyma. It regulates or partakes in inflammatory modifications occurring during disease. Within this framework, we examine BBB damage as a pathophysiological entry point that allows diagnostic strategies for brain disorders. We examine human and experimental focal epilepsy as a prototype disease to study NVU dysfunctions.
Bio: Nicola Marchi, PhD: Graduate Pharmacy and Pharmaceutical Chemistry, Pisa, Italy; a total of 12 years as fellow, post-doc, and PI Assistant Prof at Cleveland Clinic, Neurosurgery, and then Molecular Medicine. Then nine years PI and tenured researcher at CNRS, France. Funded as PI since 2009 (NIH R01, ANR France, Cure, Era-Net Europe, etc.). Editorial board activities, reviewers for various journals, funding organizations, etc. In 20 years: 120 publications; h-index 46. +10 book chapters. 1 Book as Editor.
