Makin Weldon School of Biomedical Engineering Abstract: Advancements in machine learning are narrowing the gap in visual capabilities between machines and healthcare professionals, resulting in a transformation of the way we understand and address health challenges. Despite these advances, underlying limitations persist in addressing real-world problems, particularly in the precise capture of biological and physiological information. This is primarily because traditional trichromatic cameras fall short of representing reflectance spectra due to their limited spectral information. To overcome these limitations, hyperspectral imaging has emerged as a powerful tool for biomedical applications. By collecting a wealth of information at different wavelengths, hyperspectral imaging provides a comprehensive view of electromagnetic spectra, allowing non-invasive clinical analysis for accurate diagnostics. Snapshot hyperspectral imaging, in particular, is a competitive alternative to traditional cameras as it can capture a hyperspectral image in a single shot without the need for scanning individual wavelengths. Here, we introduce a computational snapshot hyperspectral imaging method, achieved through the integration of a machine learning approach with a streamlined optical system. We design an explainable machine learning algorithm by incorporating optical and biological knowledge into the algorithm. Therefore, the algorithm can reconstruct hyperspectral images with high spectralspatial resolution comparable to those of scientific spectrometers, despite the use of sparse information captured from the optical system. To demonstrate its versatility in biomedical applications, we extract hemodynamic parameters of peripheral microcirculation from embryonic model systems, tissue phantom samples, and human conjunctivas. Furthermore, we validate high accuracy of the results using conventional hyperspectral imaging and functional near-infrared spectroscopy. This learning-powered imaging method, characterized by high resolution and simplified hardware requirements, has the potential to offer solutions for various biomedical challenges by surpassing the constraints of conventional cameras and hyperspectral imaging.

I saw 50 eager-to-learn faces in the classroom. I challenged them with a teaser of Python coding to assess their coding competency. I felt many of us were ready to hit the ground and run. A couple of changes that may affect you: (1) Moving to utilize 10-digit dialing for all outgoing calls: This dialing method requires users to dial the area code of the person being called as part of the dialing process. Important ISS Updates As we enter a new academic year, ISS would like to share the following information with the advising community: Official End of COVID Procedural Adaptations Earlier this summer the Department of Homeland Security announced that COVID-related procedural adaptations for international students would come to an end at the end of the 2023 academic year. All international students must meet minimum full-time enrollment requirements each semester as defined by federal regulation. Minimum enrollment requirements for international students are: * Graduate students must enroll in and complete at least eight credit or research hours each fall and spring semester (unless it is the student's final semester). * Graduate students who hold a graduate staff appointment must enroll in and complete at least six credit or research hours each fall and spring semester (unless it is the student's final semester). Distance Learning No more than the equivalent of one class or a maximum of three credit hours of coursework delivered via distance/online/synch/asynch may be used toward an international student's minimum enrollment requirements. Final Semester Students who are in their final semester MUST be enrolled in all remaining coursework required to complete the degree, they must enroll in at least one class that requires their physical presence on campus for most of their final semester - even when summer is the student's final semester, and the distance learning limitation is still in play. If students have questions about final semester enrollment requirements, please refer them to the ISS office. Abstract: The term neurovascular unit (NVU) indicates a brain-specific multi-cellular assembly controlling brain homeostasis for physiological neuronal transmission in networks. The blood-brain barrier (BBB) within the NVU is the microvascular interface that governs the exchanges between the peripheral blood and the brain parenchyma. It regulates or partakes in inflammatory modifications occurring during disease. Within this framework, we examine BBB damage as a pathophysiological entry point that allows diagnostic strategies for brain disorders. We examine human and experimental focal epilepsy as a prototype disease to study NVU dysfunctions. Bio: Nicola Marchi, PhD: Graduate Pharmacy and Pharmaceutical Chemistry, Pisa, Italy; a total of 12 years as fellow, post-doc, and PI Assistant Prof at Cleveland Clinic, Neurosurgery, and then Molecular Medicine. Leopold Green, PhD, Assistant Professor of Biomedical Engineering at Purdue will present "Designing living therapeutics to control host immunity." (NOTE: Students registered for the seminar are expected to attend in-person.) Abstract: Immune response is critical in septic wound healing. The aberrant and imbalanced signaling dynamics primarily cause a dysfunctional innate immune response, exacerbating pathogen invasion of injured tissue and further stalling the healing process. To design biological controllers that regulate the critical divergence of the immune response during septicemia, we need to understand the intricate differences in immune cell dynamics and coordinated molecular signals of healthy and sepsis injury. Here, we deployed an ordinary differential equation (ODE)-based model to capture the hyper and hypo-inflammatory phases of sepsis wound healing. Our results indicate that impaired macrophage polarization leads to a high abundance of monocytes, M1, and M2 macrophage phenotypes, resulting in immune paralysis. Using a model-based analysis framework, we designed a biological controller which successfully regulates macrophage dysregulation observed in septic wounds. Our model describes a systems biology approach to predict and explore critical parameters as potential therapeutic targets capable of transitioning septic wound inflammation toward a healthy, wound-healing state. Biography Dr. Green is Synthetic Biologist with a diverse academic journey. He earned his bachelor's degree in chemistry from Hampton University in 2011. With accolades, including the UNCF Merck Science Fellowship and Ronald McNair Fellowship, he pursued doctoral studies in Bioengineering at the University of California, Riverside. There, he was awarded the NSF Graduate Research Fellowship and focused on engineered nucleic acid-based nanostructures, coupling mechanical features with synthetic transcriptional oscillators. Completing his doctorate in 2017, he joined Caltech as a Post-doctoral Fellow, concentrating on engineering population controllers within synthetic E. coli-based communities. In the Fall of 2021, Dr. Green joined Purdue University's Weldon School of Biomedical Engineering. Francis, Shirley Rietdyk, Jeffrey Haddad, Sharon Christ, Melissa Newell, Sarah Burgin will present "Updated results from a study relating the effects of hearing loss and listening conditions on postural sway to fall risk in older adults." Abstract: Hearing loss is associated with increased fall risk in older adults, but multiple mechanisms have been proposed to account for this. Hearing loss may reduce spatial awareness and/or increase cognitive load, both of which may increase fall risk but may be ameliorated by hearing aid use. We are currently finishing a study study comparing fall incidence in daily life with postural sway measured under various listening conditions in older adults with and without hearing aids. An earlier version of this talk reported results from the first 17 participants (7 with hearing aids, 10 without). We have now completed enrollment and initial testing of 46 participants, each of whom stood with feet together and eyes closed, listening to noise vocoded speech (4, 8, 16 channels) and spatially distributed environmental sounds (silence, 1, 3 sources) for 1 minute per condition while postural sway was recorded in the lab. Participants subsequently reported daily near-falls and falls for 4 months (this phase will be completed in December). Initial results from the first 17 participants suggested that hearing aid users were more stable (or more rigid) than non-users and this pattern appears similar in the larger sample. Alves, PhD, within the Department of Emergency Medicine at the Indiana University School of Medicine (IUSM), is seeking a highly motivated and independent post-doctoral research fellow. Dr. Alves is a Chemical and Biomolecular Engineer, Associate Professor and the Director of Translational Research for the Department of Emergency Medicine in addition to having an affiliate faculty appointment at Purdue University in Biomedical Engineering. The Alves Lab explores highly interdisciplinary research interests applying engineering and biophysical principles and designs to create translational technologies and treatments in the area of blood coagulation and fibrinolysis. The primary focus of research in the laboratory is to develop targeted clot digesting therapeutic agents to more safely digest clinically relevant blood clots to treat pulmonary embolism (PE, blood clots in the lungs), deep vein thrombosis (DVT), and ischemic stroke. This interdisciplinary research utilizes diverse enzyme assays, chemical synthesis, blood clot formation and digestion assays under shear, and other novel testing/delivery platforms that include multivalent branched molecules and nanoparticle drug delivery systems. Projects Include: (1) Leveraging multivalency to modulate enzyme activity and improve inhibitor selectivity (nanoparticle drug delivery and targeting). (2) Ex-vivo flowing models of clot digestion under shear using in-vivo like fluorescently labeled blood clots from human whole blood. Research Participants Needed: Understanding Brain Signatures for Approval Using fNIRS We are looking for participants who are willing to participate in an experiment involving preference evaluations. A portion of the experiment will involve preference evaluation of beverages and another portion will involve preference evaluation of some engineering requirements. During the experimental session, participant brain activity will be recorded with the help of a special cap containing sources and detectors (fNIRS cap, similar to EEG but faster and cleaner). This method has been safely used with infants, children, and adults. Participants must be: * At least 18 years of age * Right-handed * No neurological disorders such as autism, seizures, or cerebral palsy * Can be either monolingual or bilingual. However, if bilingual, English must be the primary language * Able to consume water, apple juice, and ginger ale The experiment consists of a session lasting approximately 30-45 minutes. Additionally, participants' heads will be measured with a flexible measuring tape prior to the session (to ensure the appropriately sized cap is prepared). Upon completion, participants will be paid at a rate of $15 per hour. Participation in this study is completely voluntary and volunteers can withdraw from the study at any time. The students are back. There was some traffic on State Street yesterday. The Boiler Gold Rush is back. * In the past week, I sent out personal notes to the members of several committees in AY 23 - 24, and had communications with a few committee members. I appreciate so many of you for the willingness of serving on the committees and continuing to take on the committee chair position. I expect the entire list of committee assignments to be sent to you in a day or two. * With the recent changes, we have also made changes on area chairs/co-chairs. Thank you for those who are willing to step in. I'd very much like to mobilize the chairs as you know your area faculty, students, needs the best. I plan to schedule listening sessions with the chairs/co-chairs with each of the areas in the first weeks of the semester. I challenge you to come up with a list of items to cover the needs/plans (teaching and research), and the strategies of improvement before the meeting. * Following item no. 2, Young has emailed the chairs/co-chairs and asked you to solicit information regarding impactful, recent papers, books and patents on a quarterly basis. This can be the first item that helps you shine as the liaison between individual faculty and the leadership team. We will do this together to make efforts in advocating for BME. * Vitaliy and I will have a meeting this noon the CoE Office of Undergraduate Students to talk about Purdue Indy 2024. The two of us, together with Young will come down to Indy to have in-person meetings with a few members of the IUPUI BME faculty on whom we have gotten some clarity. I will have a meeting with David and a few associate deans to discuss the Purdue Indy Faculty and Teaching needs. We have got to taking care of operational issues, which is exciting. This integration business takes a village. * In recent days, there is one email (8/4/2023) from the Provost's office on Call for Proposals for New Programs in Indianapolis and Call for Faculty Engagement and New Faculty in Indianapolis. Important ISS Updates As we enter a new academic year, ISS would like to share the following information with the advising community: Official End of COVID Procedural Adaptations Earlier this summer the Department of Homeland Security announced that COVID-related procedural adaptations for international students would come to an end at the end of the 2023 academic year. All international students must meet minimum full-time enrollment requirements each semester as defined by federal regulation. Minimum enrollment requirements for international students are: * Graduate students must enroll in and complete at least eight credit or research hours each fall and spring semester (unless it is the student's final semester). * Graduate students who hold a graduate staff appointment must enroll in and complete at least six credit or research hours each fall and spring semester (unless it is the student's final semester). Distance Learning No more than the equivalent of one class or a maximum of three credit hours of coursework delivered via distance/online/synch/asynch may be used toward an international student's minimum enrollment requirements. Final Semester Students who are in their final semester MUST be enrolled in all remaining coursework required to complete the degree, they must enroll in at least one class that requires their physical presence on campus for most of their final semester - even when summer is the student's final semester, and the distance learning limitation is still in play. If students have questions about final semester enrollment requirements, please refer them to the ISS office. If a student is not properly enrolled during their final semester, they will lose valuable post-completion benefits which can severely impact their future goals. On-Campus Employment Restrictions On-campus employment is strictly limited to a maximum of 20:00 hours per week for international students, while school is in session. Students may only exceed 20 hours per week during Spring Break, Winter Break, and the summer (unless the student is required to be enrolled in the summer session). Abstract: Cerebrospinal Fluid (CSF) movement within the brain ventricles and subarachnoid spaces of the cranium and the spine is critical to the health and function of the central nervous system. Recent studies have highlighted the role of CSF in the pathophysiology of neurodegenerative, and neurodevelopmental disorders and the glymphatic system. CSF movement is also shown to be linked with sleep, with enhanced magnitudes of movement during Non-rapid eye movement (NREM) sleep. Regardless of the prominent role played by CSF in maintaining optimal brain health, a comprehensive understanding of the mechanisms driving CSF movement in humans across wake/sleep states has not been reached. To this end, multimodal imaging studies were designed and implemented with simultaneous acquisition of CSF and brain hemodynamics with functional Magnetic Resonance Imaging, neural activity with Electroencephalography, and non-neuronal systemic physiology with peripheral functional Near-Infrared Spectroscopy. From these studies, we show that the CSF system responds to multiple physiological forces at the same time. Both low-frequency vasomotion and respiration simultaneously interact and regulate CSF movement in humans during wakefulness. We also found that non-neuronal systemic physiology significantly influences CSF movement even when autonomic neural contributions come into play during light NREM sleep. Therefore, interventions may increase CSF movement by manipulating systemic physiology. Finally, we attempt to modulate CSF movement through manipulation of these low frequency cerebrovascular oscillations via simple breathing challenges. Preliminary results demonstrate their potential in modulating CSF movement and may be developed into clinical protocols for boosting CSF movement across pathologies where low flow is a concern. Abstract: The term neurovascular unit (NVU) indicates a brain-specific multi-cellular assembly controlling brain homeostasis for physiological neuronal transmission in networks. The blood-brain barrier (BBB) within the NVU is the microvascular interface that governs the exchanges between the peripheral blood and the brain parenchyma. It regulates or partakes in inflammatory modifications occurring during disease. Within this framework, we examine BBB damage as a pathophysiological entry point that allows diagnostic strategies for brain disorders. We examine human and experimental focal epilepsy as a prototype disease to study NVU dysfunctions. Alves, PhD, within the Department of Emergency Medicine at the Indiana University School of Medicine (IUSM), is seeking a highly motivated and independent post-doctoral research fellow. Dr. Alves is a Chemical and Biomolecular Engineer, Associate Professor and the Director of Translational Research for the Department of Emergency Medicine in addition to having an affiliate faculty appointment at Purdue University in Biomedical Engineering. The Alves Lab explores highly interdisciplinary research interests applying engineering and biophysical principles and designs to create translational technologies and treatments in the area of blood coagulation and fibrinolysis. The primary focus of research in the laboratory is to develop targeted clot digesting therapeutic agents to more safely digest clinically relevant blood clots to treat pulmonary embolism (PE, blood clots in the lungs), deep vein thrombosis (DVT), and ischemic stroke. This interdisciplinary research utilizes diverse enzyme assays, chemical synthesis, blood clot formation and digestion assays under shear, and other novel testing/delivery platforms that include multivalent branched molecules and nanoparticle drug delivery systems. Projects Include: (1) Leveraging multivalency to modulate enzyme activity and improve inhibitor selectivity (nanoparticle drug delivery and targeting). (2) Ex-vivo flowing models of clot digestion under shear using in-vivo like fluorescently labeled blood clots from human whole blood.
