Opportunity to Mentor Undergrads through GradTrack
Below is an opportunity for graduate students to mentor undergraduates interested in pursuing graduate school. More information is available at: https://engineering.purdue.edu/Engr/Academics/Graduate/gradtrack/index_html.
GradTrack Program Dates: Meet virtually on Monday nights in the Fall from 7-8:15pm for 1.5 hour from August 2022 - Dec 2022 (8 meetings total, dates to be shared soon; please see website for more information). There will also be a mandatory mentor training in August 2022 and a debrief dinner in early Dec 2022.
Selected Graduate Student GradTrack Mentors will be given $1,000 for their time.
What you will do as a mentor:
* Receive mentorship training prior to program start.
* Lead a group of 5-7 undergraduate students in monthly discussions about various topics related to graduate school (The Office of Graduate Education will share discussion topics).
* Actively engage in mentoring. There will be an estimated 2-4 hours a month outside of the monthly meetings to answer student questions, meet one-on-one, and provide feedback on documents when appropriate.
Eligibility criteria:
* Be enrolled in a Master's or PhD program within Purdue's College of Engineering
* Be in good academic standing with advisor support of your participation
* Have an interest in mentoring underrepresented students

PATHFINDER: A program dedicated to career development of Purdue life scientists
Pathfinder is a new program dedicated to help with career development. Pathfinder aims to link with local companies and build bridges for improving the retention of talented life scientists in Indiana. The program exposes our talented graduate students and postdocs to scientists that are currently working in life sciences companies in Indiana and can act as models for successful careers at our doorsteps. The plan for the Pathfinder program is to conduct virtual interviews sessions with industry scientists and capture their story to identify factors for their success as industry scientists.
Our first interview is with Jonathan Snyder, PhD, Principal Research Scientist at Elanco in Greenfield, IN:

ME 584 deals with building models from data or the conditions that experiments must satisfy to extract the parameters of physical systems. Course description: The course this time will be suitable to students all around science and engineering and the data sciences. Students will be able to work on a variety of course projects identifying generalized least squares models, or dynamic system models, or various machine learning models. It will provide a fundamental grasp on the underlying theory (least squares, maximum likelihood estimation and connection to experiment design), models for estimation (ARMAX type or difference equation models, neural nets--supervised/unsupervised, physics-based), estimation algorithms (subspace, linear dimensionality reduction, convergence issues) and their implementation (testing for optimality, robustness, and statistical validity). Extensions to inverse problems and pattern recognition will also be covered. The course will possibly have an online section later on for those looking for schedule flexibility.
The course has typically had students from all over engineering, including those from thermal and fluid sciences, food science, and agriculture. See the attached syllabus from last year for the details.
Executive Functioning for Biomedical Researchers- May 2, 3 PM part of Mental Health Awareness Month
May is Mental Health Awareness Month, and the NIH Office of Intramural Training and Education (OITE) is excited to be starting the month off with the fifth installment of the Mental Health and Well-being for Biomedical Researchers Series.
Please join us May 2nd, 3:00-4:10pm ET for "Executive Functioning for Biomedical Researchers". The webinar will define executive functioning (EF) skills, with a particular focus on the role of EF for those working in biomedical research fields. The webinar will provide information on the types of daily stressors that impact one's executive functioning, as well as concrete ways to manage EF needs and build EF tools. Immediately following the webinar (from 4:10 - 5:10), we will host small group discussions facilitated by an OITE wellness advisor to discuss the material in greater depth. Information on the follow-up sessions will be sent to those who register for the webinar. Additionally, a recording of the webinar will be made available 24-48 hours after the webinar. This series is open to everyone at NIH and beyond, so feel free to forward to colleagues at other institutions.

15TH ANNUAL NIH CAREER SYMPOSIUM- AN ONLINE EVENT

The NIH Career Symposium highlights the diversity of career choices available to biomedical researchers today. Whether you are a new graduate student, postdoc, or clinical fellow just beginning to consider career options or a senior student/fellow ready to look for a job, the NIH Career Symposium is for you. Wondering what the Career Symposium is all about? Check out past events here<https://www.training.nih.gov/nih_career_symposium>.
We have already recruited over 250 speakers for you to network with. We will start each day with an intro on each sector (career options, being a competitive applicant, resources for success. This is a great way for you to jumpstart each day and plan for what events you would like to attend. Then you will have the ability to network with professionals in a variety of jobs, participating in informational interviews, which we hope will help you identify your career of choice and the steps required to get your foot in the door. We will host a series of zoom rooms for career conversations with one or two speakers. Multiple rooms with speakers in similar career paths will run concurrently. For example, research positions in industry would have multiple rooms going at once. Our goal is that each room will have no more than 15 participants, giving every attendee ample time to connect with a professional. Participants will have the ability to move through the "rooms" as they like, meeting a speaker, asking questions, or even just listening in to see if a particular career path looks enticing. NOTE: What is said in these rooms will NOT be recorded.
We will also have a workshop on Why Career Choices are so Hard: a practical guide for navigating the career on Wednesday afternoon, and a workshop on Thursday afternoon on Current Hiring Trends.
Closer to the event we will send a guide to those registered on how to navigate these rooms for informational interviews (including seed questions).

Abstract: Over 150 years since Claude Bernard coined the evocative phrase, "milieu intérieur", to draw attention to a deep mystery in the natural world, fundamental questions remain open about how the simplest living system, a bacterial cell, maintains homeostasis of its best studied attribute, cell size. In this preliminary work towards dissertation, I will address how life shapes time in a bacterial cell, and discuss the interplay between homeostasis and adaptation in this context. First, I will first establish that stochastic intergenerational bacterial cell size homeostasis is maintained under constant growth conditions for non interacting cells, using our high-precision data on growth and division of individual C. crescentus cells using our SChemostat technology. Next, the emergent simplicities or scaling laws revealed by these data, and their mechanistic implications. Following this, the questions, approaches and challenges in the extension to time-varying growth conditions, other microorganisms, and spatially patterned arrangement of cells, and possible new emergent simplicities.

As we approach the end of semester- we have a lot of exciting opportunities to connect, look at future research opportunities, and celebrate accomplishments. Specifically, this week Wednesday we will have an Engineering Solutions in Pediatric Healthcare Symposium from 1:00-4:30 PM. Thursday, we will have a Faculty Mentor Award Celebration from 12pm to 1pm sponsored by BMEGSA for faculty, staff, and grad students. And Friday, an end-of-semester informal celebration for the Weldon School students, faculty and staff will be held at the MJIS South Lawn, from 3 - 5 pm. I look forward to seeing you all at these events this week!

Her check-in time is 11:05 AM; run time is 11:20 AM. 13 Boilermakers (student leaders, faculty, staff, and alumni) will run a relay (some participants will run concurrently) totaling 20.22 miles. Update on Weldon Grad-Program Update Process:
* Based on all the input and feedback we have received and our discussions throughout the year, we have developed thorough DRAFT outline documents for the new Qualifying Exam process and the revised Prelim Exam process.
* Each document describes the Rationale, Expectations, Logistics/Process, and Resources associated with each process, and are intended to inform everyone so that both students and faculty understand the process and rationale.
* Each new process was developed based on the themes that emerged from input and feedback we received, e.g.,
* consistent and broad mentoring from start to finish,
* flexibility in milestone timing given the diversity of entering students' experience and backgrounds,
* a need for more formal writing courses,
* a desire to have milestone processes be useful for the student and not just busy work for students to complete and faculty to evaluate
* consistent and well-communicated rigorous expectations for advancement to each stage of the PhD Training Program
* We have spent one entire Grad-Committee meeting discussing the qual document and another meeting discussing the prelim document to fine tune our plans.
* We are now voting to make sure all committee members are comfortable with the details of the new frameworks we have developed.
* Once formally approved by the Grad Committee (next two weeks), we will be distributing these documents as well as sharing a report on the feedback we received and how it was incorporated into our plans. We will also be fine-tuning the processes (e.g., developing rubrics and registration forms to ensure smooth implementation of these new processes.
* Note: these new processes will be implemented this Fall (2022) for all incoming students. Current students will have the option (individually) to use the old or new system depending on what is best for them.

With the goal of fostering an inclusive BME culture and enhancing our undergraduate curriculum, BMES aims to bring our students and faculty closer together by hosting more Undergraduate Open Forums in future semesters. Trailblazers in Engineering (TBE) are workshops to prepare future trailblazing faculty in engineering with a focus on preparing outstanding scholars who are also committed to increasing the success of underrepresented communities of engineers. Fellows will network with leading faculty in engineering, thought leaders, program managers at key federal funding agencies, Purdue Engineering heads, and of course, each other. Participants will also meet current Trailblazers and benefit from their inspiring keynote talks. TBE 2022 will be held in person on Purdue's beautiful West Lafayette, IN campus July 26 - 28, 2022.

BME PhD Preliminary Exam Announcement for Jordanna Payne (L. Solorio advisor), Tues., April 26, 3:00 p.m., BRK 1001. Title: Development and characterization of a subcutaneous porcine ex-vivo tissue culture model. Please note this prelim is open only to those who have signed the Lilly NDA.

Abstract: The subcutaneous route of drug delivery is desirable due to patient comfort and ease of administration. One of the biggest challenges in studying drug delivery in the subcutaneous space is the lack of in-depth knowledge about the composition of the space. Because of this, several drugs encounter difficulty being delivered via this route presenting challenges during clinical trials. Currently, drugs are tested in vitro and in vivo. In vitro experiments are more cost effective but lack the ability to provide information about drug uptake by tissue and other interactions or toxicities. In vivo experiments can provide this information but are costly. There is a need for a platform that can be used to study subcutaneous injection of drugs ex-vivo. This work details the development and validation of a model of ex-vivo subcutaneous porcine tissue culture for up to one week utilizing three distinct anatomical locations from Yucatan mini pigs. Tissue specimens of 12mm in diameter from behind the ear, the breast, and the belly were cultured for seven days with average viability greater than 85%. Next, Liquid Chromatography-Mass Spectrometry (LC-MS) was utilized to track changes in the subcutaneous proteome as a result of time in culture. Next, LC-MS will be utilized to track changes in the subcutaneous proteome of a commercially available ex-vivo human tissue model to compare to the porcine model. Finally, parameters for scaling of the model for high throughput will be determined.
Raising A Resilient Scientist Workshop Series
(Message from Sharon Milgram, Director, NIH Office of Intramural Training & Education): Over the last year I have interacted with many trainees across the U.S. as part of the NIH Becoming a Resilient Scientist series<https://www.training.nih.gov/nih_becoming_a_resilient_scientist_series>. After carefully considering input from students, postdocs, faculty and program administrators who participated in the program, I have decided to offer a series, RAISING A RESILIENT SCIENTIST<https://www.training.nih.gov/workshops_and_trainings_for_extramural_princip…>, for faculty and administrators who mentor students and postdoctoral fellows in the biomedical, behavioral, and social sciences. The goal of the Raising a Resilient Scientist series is to promote the mental health and well-being of the academic research community by supporting faculty and administrators to develop self-management, relationship-management, and mentoring skills. A variety of topics will be covered with a specific focus on communication and shared problem-solving to help trainees deal with the inevitable setbacks experienced in high knowledge environments. There will be five units in the series; each unit will consist of a 75-minute lecture, which can be watched in real-time or asynchronously, followed by a 1-hour facilitated discussion with peers. There will be two opportunities for participation in the small groups following each webinar. There is no charge for participation, but advanced registration is required. NOTE: Although this seminar will be presented virtually, BME 690 students are still expected to attend in person in MJIS 1001.
Abstract: Metastatic disease is the leading cause of cancer related mortality across a variety of tumors. Disseminated tumor cells (DTCs) are considered the source of metastasis and they remain in a reversible non-proliferative state of dormancy resisting anti-proliferative therapies during the extended and asymptomatic periods of the minimal residual disease. Expanding our knowledge on how DTCs survive in a dormant state in their new microenvironments, how DTCs interact with the immune system, what are the signals that trigger their reactivation programs, and what are the signals responsible for long-term dormancy, may likely improve predicting patient survival rates and increase the opportunities to design therapies that will extend the remission phase.
Bio: Dr. Sosa did her career in Molecular Biology at the University of San Luis, Argentina and her undergraduate training at Leloir Institute in Argentina under the supervision of Drs. Llera and Podhajcer, where she studied the secretome of melanoma cancer. In 2005, Dr. Sosa started her Ph.D. training in Dr. Marcelo Kazanietz's laboratory at the University of Pennsylvania (UPENN). During her training she focused on the mechanisms of action of Rac-GEFs and -GAPs in breast cancer. In 2010, Dr. Sosa joined Dr. Aguirre-Ghiso's lab as a postdoctoral fellow at Mount Sinai Hospital. Since then, Dr. Sosa has focused on the study of minimal residual disease in an attempt to understand the mechanistic pathways in disseminated tumor cells (DTCs). In 2017, Dr. Sosa started an Assistant Professor appointment in the Department of Pharmacological Sciences at Mount Sinai Hospital. Dr. Sosa has published more than 25 articles and has secured several grants including: Schneider-Lesser Foundation Fellow Award, CSBC Pilot Project-Sage Bionetworks, Melanoma Research Alliance Team Science Award (Co-PI), NCI-K22, CCR Basic/Translational and Clinical Susan Komen, Melanoma Research Foundation, Breast Cancer Foundation, and The Mark Foundation (Co-PI) grants. Her lab research focuses on providing new biomarkers and treatments derived from the novel biology of dormant disease to manage minimal residual disease and prevent metastasis.

BME PhD Preliminary Exam Announcement for Angel G. Enriquez (H. Lee, advisor), Wed., April 27, 3:00 p.m., MRGN 129. Title of presentation: Magnetic actuators for smart functionality in biomedical devices.

Abstract: Magnetism provides numerous advantages to actuating systems, specifically for biomedical applications. The untethered transfer of energy and scalability of magnetic actuators enables functionality to an otherwise passive system. For example, the wireless magnetic actuation can turn static 2D and 3D cell cultures into a more physiologically-relevant dynamic environment while limiting contamination. Moreover, indwelling catheters and implantable sensors are typically stationary devices that are notorious for their short lifespan when implanting into the body due to immune responses. Magnetic microactuators may be used for wireless actuation for in situ removal of biological materials accumulated on chronically implanted devices. In this dissertation, I will demonstrate examples of novel biomedical microdevices enabled by magnetic actuation for added functional benefits. First, I will describe a soft polymer magnetic actuator that can facilitate the study of a physiologically relevant cell culturing system. By cyclically stretching an extracellular matrix protein in a 3D cell culture, this system can elucidate the process by which breast cancer cells respond to a dynamic environment in the lungs. The fibrillar fibronectin suspended across the body of the magnetic actuator provides a matrix representative of early metastasis for 3D cell culture that has not yet been recapitulated in vitro until now. Our findings suggest that this dynamic environment leads to significant matrix degradation in breast cancer cell lines with metastatic potential. Our results demonstrate a clear suppressive cellular response due to cyclic stretching that has implications for a mechanical role in the dormancy and reactivation of disseminated breast cancer cells to macrometastases. As a second application, I will demonstrate the use of magnetic microactuators to remove biofouling on an implantable biosensor in order to prolong its functionality. The results of our work suggest that the motion of the actuator on the sensor surface can maintain biosensor signal integrity and prevents the downstream effects of the foreign body response. In the future, I plan to evaluate the performance of the self-clearing biosensor in vivo to demonstrate chronic and reliable sensor functionality.

Bio: Jasmine Kwasa, PhD, is an NIH-funded post-doctoral fellow in the Neuroscience Institute at Carnegie Mellon University (CMU). Originally from the South Side of Chicago, Jasmine earned her B.S. from Washington University in St. Louis, her M.S. from Boston University (both in Biomedical Engineering), and her Ph.D. in Electrical and Computer Engineering from CMU. Her ongoing post-doctoral research seeks to develop neurotechnologies, such as EEG and fNIRS, optimized for coarse, curly hair and dark skin pigmentation with collaborators at CMU. She is also a neuro-ethicist and writes about the future of inclusive neurotech and the history of racial bias in neuroscience, medicine, and technology. Jasmine has received several honors throughout her training, including being named a Ford Foundation Fellow, an NSF Graduate Research Fellow, a Society for Neuroscience fellow, and a "Rising Star in Biomedical Sciences" by MIT. In her free time, Jasmine is a dance fitness instructor and enjoys travel and time with her enormous family.

Bio: Dr. Sosa did her career in Molecular Biology at the University of San Luis, Argentina and her undergraduate training at Leloir Institute in Argentina under the supervision of Drs. Llera and Podhajcer, where she studied the secretome of melanoma cancer. In 2005, Dr. Sosa started her Ph.D. training in Dr. Marcelo Kazanietz's laboratory at the University of Pennsylvania (UPENN). During her training she focused on the mechanisms of action of Rac-GEFs and -GAPs in breast cancer. In 2010, Dr. Sosa joined Dr. Aguirre-Ghiso's lab as a postdoctoral fellow at Mount Sinai Hospital. Since then, Dr. Sosa has focused on the study of minimal residual disease in an attempt to understand the mechanistic pathways in disseminated tumor cells (DTCs). In 2017, Dr. Sosa started an Assistant Professor appointment in the Department of Pharmacological Sciences at Mount Sinai Hospital. Dr. Sosa has published more than 25 articles and has secured several grants including: Schneider-Lesser Foundation Fellow Award, CSBC Pilot Project-Sage Bionetworks, Melanoma Research Alliance Team Science Award (Co-PI), NCI-K22, CCR Basic/Translational and Clinical Susan Komen, Melanoma Research Foundation, Breast Cancer Foundation, and The Mark Foundation (Co-PI) grants. Her lab research focuses on providing new biomarkers and treatments derived from the novel biology of dormant disease to manage minimal residual disease and prevent metastasis.

Intensive Writing Experience for Thesis & Dissertation Writers
The Writing Lab, in collaboration with the Graduate School, is once again offering summer sessions of the Intensive Writing Experience. The purpose of the Intensive Writing Experience is to give masters and doctoral students in good standing with their programs time to write or to revise a thesis or dissertation with support from Writing Lab staff. Participants will receive writing instruction and support from Writing Lab staff, will spend significant time writing or revising their thesis or dissertation, and will network with other graduate writers on campus.

Abstract:
The subcutaneous route of drug delivery is desirable due to patient comfort and ease of administration. One of the biggest challenges in studying drug delivery in the subcutaneous space is the lack of in-depth knowledge about the composition of the space. Because of this, several drugs encounter difficulty being delivered via this route presenting challenges during clinical trials. Currently, drugs are tested in vitro and in vivo. In vitro experiments are more cost effective but lack the ability to provide information about drug uptake by tissue and other interactions or toxicities. In vivo experiments can provide this information but are costly. There is a need for a platform that can be used to study subcutaneous injection of drugs ex-vivo. This work details the development and validation of a model of ex-vivo subcutaneous porcine tissue culture for up to one week utilizing three distinct anatomical locations from Yucatan mini pigs. Tissue specimens of 12mm in diameter from behind the ear, the breast, and the belly were cultured for seven days with average viability greater than 85%. Next, Liquid Chromatography-Mass Spectrometry (LC-MS) was utilized to track changes in the subcutaneous proteome as a result of time in culture. Next, LC-MS will be utilized to track changes in the subcutaneous proteome of a commercially available ex-vivo human tissue model to compare to the porcine model. Finally, parameters for scaling of the model for high throughput will be determined.

Abstract: Type 1 Diabetes Mellitus (T1D), a chronic disease affecting over 1.5 million Americans, is characterized by the autoimmune destruction of insulin-producing β-cells within pancreatic islets. Islet/β-cell replacement therapies, where replenishable β-cell sources are implanted within protective microenvironments, have the potential to provide a long-term solution for individuals with T1D by restoring glucose-sensitive, insulin release and overall glycemic control. However, most conventional encapsulation materials elicit an immune reaction, known as a foreign body response (FBR), which compromises β-cell health and function. In this dissertation, we designed and evaluated various formulations of a polymerizable collagen, namely type I oligomeric collagen (Oligomer), as encapsulation materials for minimally invasive, subcutaneous delivery of replacement β-cells. Preclinical validation in chemically-induced diabetic mice demonstrated rapid (within 24 hours) reversal of diabetes for beyond 90 days with no signs of rejection or FBR after subcutaneous delivery of both allogeneic and xenogeneic (rat) islets. To further define this uncommon mechanism of protection, the tissue response to Oligomer, in comparison to commercial synthetic and collagen-based materials, was evaluated following subcutaneous implantation within rats, a well-established biocompatibility model. Histological and transcriptomics analyses were used to define the immune response at both cellular and molecular levels. Interestingly, Oligomer showed minimal and transient activation of innate immune cells similar to the sham surgical control, with no evidence of foreign body giant cell formation, inflammatory-mediated bioresorption, or fibrosis. Overall, this work evaluates preclinical efficacy and demonstrates mechanistic understanding of immune tolerance for Oligomer materials for β-cell replacement therapy and other regenerative medicine applications.

Abstract: Pancreatic cancer is predicted to become the 2nd leading cause of cancer deaths by 2030. KRAS is the most frequently mutated gene in pancreatic ductal adenocarcinoma (PDAC) patients (~90%) and has been shown to contribute to a variety of tumor characteristics. In this presentation we will present data implicating Protein phosphatase 2A (PP2A) as a key regulator of KRAS phenotypes. We demonstrate that suppression of PP2A accelerates PDAC initiation and progression, and that the therapeutic activation of PP2A can prevent nutrient scavenging to drive cancer cell death.
Bio: Dr. Brittany Allen-Petersen is an Assistant Professor in the Biological Sciences Department at Purdue University. She obtained her Ph.D. from the University of Colorado and completed an NCI F31 funded postdoctoral fellowship at Oregon Health & Science University. The Allen-Petersen lab broadly focuses on the deregulation of phosphatases in cancer using a variety of models, including 3D culture, in vivo mouse models, and treatment with novel small molecule activators of PP2A. Dr. Allen-Petersen has funding through an NCI K22 Transition Career Development Award and a Pancreatic Cancer Action Network Career Award.

"Assessing Patient-specific Cardiac-Drive Brain Motion: MRI-based Biomarkers of White Matter Lesion Formation." Open to all undergraduate and graduate researchers. Lunch will be provided.
Abstract: In recent years, identifying risks factors associated with cognitive decline (occurring at 46-60 years) and defining mechanisms behind clinically visible damage to the brain (occurring at 60-90 years) has garnered attention from researchers and physicians. Due to the inability of clinical treatment to reverse the deleterious effects of damage to the brain, specifically in the form of white matter lesions, emphasis is heavily placed on disease prevention. We expand upon the vascular dysfunction hypothesis that places cardiovascular risk factors at the precipice of neurodegeneration. Specifically, this work leverages state-of-the-art time-resolved MRI and brain modeling techniques to quantify the contribution of biomechanical, morphological, and clinical risk factors to the manifestation of cognitive decline. Successful identification of biomarkers of neurodegeneration observable in mid-life could provide researchers with a window of opportunity to develop treatments and inform clinical management of risk factors before the damage manifests in irreversible cognitive deficit.

Abstract: Magnetic resonance imaging (MRI) has undergone an enormous advancement in diagnostic accuracy through improved comprehensive evaluation of early soft tissue transformation while avoiding the exposure to potentially dangerous ionizing radiation. With decreased cost and enhanced availability, the use of MRI is becoming ever more persuasive throughout clinical practice. Complementary to the MRI, magnetic resonance spectroscopy (MRS) offers unique information about the micromolecular composition of the human body by capturing the chemical signature of metabolites and neurotransmitters via frequency changes and displaying this information in a spectrum. The applications of MRS to investigate the metabolic window on a wide range of biochemical processes are incredibly diverse. For example, we have utilized MRS to investigate the composition and function of the human brain in post-traumatic stress disorder, traumatic brain injury, addiction, and hyperbaric oxygen therapy assessment. In addition to MRS biological specificity, diffusion tensor imaging (DTI) is yet another advanced MRI methodology used to map and characterize the three-dimensional diffusion of water as a function of spatial location. Analogous to the MRS, DTI applications are diverse and not limited to in vivo studies. To address the significant challenges of strict drug user adherence regimens for numerous treatments, we have evaluated the drug release profile from a newly developed in situ forming implants (ISFIs) for sustained and constant drug delivery. Taken together, advanced MRI methods that probe the morphology, microstructure, function, and chemical changes are clinically applicable in various diseases and conditions.

Abstract: Once a medical device is implanted, a series of interactions occur between the surface of the device and the body fluids where proteins start to be adsorbed on the surface which initiates cell and bacterial adhesion, and this can eventually lead to thrombus formation and bacterial contamination. Both of the two complications can significantly reduce the quality and the efficiency of the implanted device and may ultimately reduce the reliability of the implant. The polyvinylchloride (PVC) surface as a common medical material is hydrophobic and highly prone to such complications, urging researchers to develop an enhanced surface coating. Unlike other approaches of inadequate surface coating or limited antifouling polymer density and thickness, we propose to form a hydrogel that is covalently bonded on the PVC surface to significantly reduce protein adsorption, cell adhesion, and biofilm formation by a dense layer of antifouling polymers via surface-initiated polymerization. Hydrogels are prepared based on polymers having the capacity of swelling large amounts of water, which can then effectively repel proteins, cells, and bacteria. The covalently attached hydrogel will be fabricated via a conventional free-radical polymerization initiated through a biocompatible initiator or chain-transfer agent. Conventional free-radical polymerization is efficient, simple, cost-effective, and more friendly to the environment when compared to advanced polymerization methods. We believe that the successful achievement of the project will greatly impact the field of blood, cardiovascular and urological research, and the quality of health care for those requiring PVC-based medical devices and supplies in blood transfusion, cardiovascular and urological applications.

Abstract: This thesis aims to investigate how one fundamental component of the inner-ear (cochlear) response to all sounds, the temporal fine structure (TFS), is used by the auditory system in everyday hearing. Although it is well known that neurons in the cochlea encode the TFS through exquisite phase locking, how this initial/peripheral temporal code contributes to everyday hearing and how its degradation contributes to perceptual deficits are foundational questions in auditory neuroscience and clinical audiology that remain unresolved despite extensive prior research. This is largely because the conventional approach to studying the role of TFS involves performing perceptual experiments with acoustic manipulations of stimuli (such as sub-band vocoding), rather than direct physiological or behavioral measurements of TFS coding, and hence is intrinsically limited. The present thesis addresses these gaps in three parts: 1) developing assays that can quantify TFS coding at the individual level 2) comparing individual differences in TFS coding to differences in speech-in-noise perception across a range of real-world listening conditions, and 3) developing deep neural network (DNN) models of speech separation/enhancement to complement the individual-difference approach. By comparing behavioral and electroencephalogram (EEG)-based measures, Part 1 of this work identified a robust test battery that measures TFS processing in individual humans. Using this battery, Part 2 subdivided a large sample of listeners (N=200) into groups with "good" and "poor" TFS sensitivity. A comparison of speech-in-noise scores under a range of listening conditions between the groups revealed that good TFS coding reduces the negative impact of reverberation on speech intelligibility, and leads to reduced reaction times suggesting lessened listening effort. These results raise the possibility that cochlear implant (CI) sound coding strategies could be improved by attempting to provide usable TFS information, and that these individualized TFS assays can also help predict listening outcomes in reverberant, real-world listening environments. Finally, the DNN models (Part 3) introduced significant improvements in speech quality and intelligibility, as evidenced by all acoustic evaluation metrics and test results from CI listeners (N=6). These models can be incorporated as "front-end" noise-reduction algorithms in hearing assistive devices, as well as complement other approaches by serving as a research tool to help generate and rapidly sub-select the most viable hypotheses about the role of TFS coding in complex listening scenarios.

Raising A Resilient Scientist Workshop Series

After carefully considering input from students, postdocs, faculty and program administrators who participated in the program, I have decided to offer a series, RAISING A RESILIENT SCIENTIST<https://www.training.nih.gov/workshops_and_trainings_for_extramural_princip…>, for faculty and administrators who mentor students and postdoctoral fellows in the biomedical, behavioral, and social sciences. The goal of the Raising a Resilient Scientist series is to promote the mental health and well-being of the academic research community by supporting faculty and administrators to develop self-management, relationship-management, and mentoring skills. A variety of topics will be covered with a specific focus on communication and shared problem-solving to help trainees deal with the inevitable setbacks experienced in high knowledge environments. There will be five units in the series; each unit will consist of a 75-minute lecture, which can be watched in real-time or asynchronously, followed by a 1-hour facilitated discussion with peers. There will be two opportunities for participation in the small groups following each webinar. There is no charge for participation, but advanced registration is required. Abstract: Magnetism provides numerous advantages to actuating systems, specifically for biomedical applications. The untethered transfer of energy and scalability of magnetic actuators enables functionality to an otherwise passive system. For example, the wireless magnetic actuation can turn static 2D and 3D cell cultures into a more physiologically-relevant dynamic environment while limiting contamination. Moreover, indwelling catheters and implantable sensors are typically stationary devices that are notorious for their short lifespan when implanting into the body due to immune responses. Magnetic microactuators may be used for wireless actuation for in situ removal of biological materials accumulated on chronically implanted devices. In this dissertation, I will demonstrate examples of novel biomedical microdevices enabled by magnetic actuation for added functional benefits. First, I will describe a soft polymer magnetic actuator that can facilitate the study of a physiologically relevant cell culturing system. By cyclically stretching an extracellular matrix protein in a 3D cell culture, this system can elucidate the process by which breast cancer cells respond to a dynamic environment in the lungs. The fibrillar fibronectin suspended across the body of the magnetic actuator provides a matrix representative of early metastasis for 3D cell culture that has not yet been recapitulated in vitro until now. Our findings suggest that this dynamic environment leads to significant matrix degradation in breast cancer cell lines with metastatic potential. Our results demonstrate a clear suppressive cellular response due to cyclic stretching that has implications for a mechanical role in the dormancy and reactivation of disseminated breast cancer cells to macrometastases. As a second application, I will demonstrate the use of magnetic microactuators to remove biofouling on an implantable biosensor in order to prolong its functionality. The results of our work suggest that the motion of the actuator on the sensor surface can maintain biosensor signal integrity and prevents the downstream effects of the foreign body response. In the future, I plan to evaluate the performance of the self-clearing biosensor in vivo to demonstrate chronic and reliable sensor functionality.

The workshop will provide Fellows the ability to network with leading faculty in engineering, thought leaders, program managers at key federal funding agencies, Purdue Engineering heads, and of course, each other. Participants will also meet current Trailblazers and benefit from their inspiring keynote talks.
TBE 2022 will be held in person on Purdue's beautiful West Lafayette, IN campus July 26 - 28, 2022.

The Role of PP2A in the Regulation of Pancreatic Cancer

Abstract: Pancreatic cancer is predicted to become the 2nd leading cause of cancer deaths by 2030. KRAS is the most frequently mutated gene in pancreatic ductal adenocarcinoma (PDAC) patients (~90%) and has been shown to contribute to a variety of tumor characteristics. In this presentation we will present data implicating Protein phosphatase 2A (PP2A) as a key regulator of KRAS phenotypes. We demonstrate that suppression of PP2A accelerates PDAC initiation and progression, and that the therapeutic activation of PP2A can prevent nutrient scavenging to drive cancer cell death.
Bio: Dr. Brittany Allen-Petersen is an Assistant Professor in the Biological Sciences Department at Purdue University. She obtained her Ph.D. from the University of Colorado and completed an NCI F31 funded postdoctoral fellowship at Oregon Health & Science University. The Allen-Petersen lab broadly focuses on the deregulation of phosphatases in cancer using a variety of models, including 3D culture, in vivo mouse models, and treatment with novel small molecule activators of PP2A. Dr. Allen-Petersen has funding through an NCI K22 Transition Career Development Award and a Pancreatic Cancer Action Network Career Award.

Summer Science Program in Genomics: please help us find one more TA
An opportunity for undergrads or graduate students to be involved with a highly regarded STEM program for High School students. Students should not have other commitments during the Summer Science Program as it is definitely a full-time commitment.
The Summer Science Program in Genomics at Purdue University
Teaching Assistant and Residential Mentors are tutors and instructors, assisting participants in using equipment and software, and with problem sets, under the direction of the Academic Director. Equally important is their simultaneous role as residential advisors and personal mentors, supervising participants and organizing recreational and social activities, under the direction of the Site Director. Room & board and travel are provided, plus a salary of about $1,000 per week for 6 1/2 weeks.
Working in teams of three, participants will use a programmable chemostat to expose a marine microbe to an antibiotic, then analyze the mutations observed in the evolved genome.
Applicants should be rising college seniors, graduating seniors, or graduate students with wet lab or genomics/bioinformatics knowledge and experience, who enjoy teaching.
