Research Title: Elucidating Pathological Correlations between Traumatic Brain Injury and Alzheimer's Disease

Abstract: One of the greatest consequences of Traumatic Brain Injuries (TBI) are the resulting long-term effects, especially in milder cases where the damage is initially subclinical and thus lacking acutely observable manifestations that can compound significantly over time. Among these chronic issues, Alzheimer's Disease (AD) is one of the most serious. While a link between TBI and AD has been established, it has not yet been mechanistically elucidated. Multiple hypotheses have been postulated based on various animal/clinical models, which have contributed a great deal to our knowledge base and implicated several key pathways. While extremely valuable, these in vivo procedures and analyses are physiologically and ethically complex: there is currently no model capable of visualizing TBI-induced sub-cellular damage in the seconds immediately following injury, and the subsequent associated long-term changes. It is clear that additional investigative tools are needed, and while in vitro methodologies generally offer the type of resolution required, no such model replicates this phenomenon. Therefore, we introduce the "TBI-on-a-chip" concussive model prototype, with a series of concomitant targeted-experiments to address this currently unmet need.
We hypothesize that both the primary, mechanical injury and long-term associated damages (secondary injury) can be successfully recapitulated in vitro, and utilized for mechanistic studies into trauma induced neurodegeneration. Further, we hypothesize that aggregation (β-amyloid) can be initiated by force-solo and demonstrated in our proposed system, further linking TBI with AD. Our ultimate goal is to facilitate translational discoveries by exploring the mechanisms underpinning TBI, acute and chronic, and the link to neurodegeneration.

Graduate Programs – update
The Graduate School will be hosting two Graduate Faculty Mentoring Workshops in West Lafayette this spring. The first will be on Wednesday, March 9, 2022 from 9:00 a.m. to 11:30 a.m.; and the second will be on Thursday, March 10, 2022 from 9:00 a.m. to 11:30 a.m. Both workshops will be held at the Stewart Center in Room 202 unless we have to move to virtual.
The workshop provides an overview to the policies, procedures and expectations associated with mentoring graduate students at Purdue University. Participation in the workshop is required for any faculty member who wishes to seek appointment to the Graduate Faculty at Purdue University.
Historically it has been the practice of the Graduate School to require faculty new to Purdue to attend a Graduate Faculty Mentoring Workshop before they could obtain graduate faculty status. While a practice, it is not “policy” so we will not hold up faculty appointments due to logistics of when the workshops are offered. This does not negate the request for faculty new to Purdue to attend one of the new faculty workshops, but it does mean the Graduate School will not hold an appointment request based on workshop completion.
In addition to attending one of these two workshops, faculty who wish to hold a position as a regular member of the Graduate Faculty must also be recommended by their unit and be a tenure-track or voting status faculty member. This process must be initiated in the Graduate School Database, and is typically done by the department graduate contact. Appointment to the graduate faculty permits a faculty member to teach graduate-level courses, to serve on graduate student advisory committees, and to chair or co-chair master’s and Ph.D. committees. The level of appointment is dependent upon the faculty member’s position, rank and credentials.
Graduate coordinators and contacts - Please provide faculty members the instructions below to register online. I will also still accept email registrations if there are any complications but my preference would be to direct them to use the electronic system. It is very important that participants arrive on time and do not leave early. Missing part of the presentation could cause their attendance to not count toward fulfilling the requirement needed to advise students or become a regular faculty member. Please make sure that he registering faculty members are aware of this.

* Individual Development Plans (IDPs). Please find attached IDP forms for BME. The graduate committee wants all of our trainees/students to use IDPs throughout their career with us. These forms are intended to be student initiated and then reviewed AND DISCUSSED with faculty mentors and ideally mentoring/thesis committees. Again, our forms are just helpful templates – any forms you find that you like are OK to use as an IDP.
* Launching Coffee Chats THIS SEMESTER. To facilitate more interactions between students and faculty outside their own lab, we are launching a new initiative. 1-2 faculty members will host coffee chats each month to discuss anything that is on the students’ minds (some starter topics are suggested for each meeting, but really anything is on the table.). There is a lot of excitement about this initiative from our students, as well as from the Graduate Committee – we look forward to the first coffee chat in early February.
* January is Mentoring Month!! The MOST CONSISTENT THEME that has come out of our systematic Grad-Programs review has been the need for better and more consistent mentoring throughout our student’s graduate programs. Links are provided in the DEI section to several excellent programs (including the NIH webinar series on RAISING A RESILIENT SCIENTIST<https://www.training.nih.gov/workshops_and_trainings_for_extramural_princip…> that was mentioned in faculty meeting on Friday).
* Systematic Grad Programs Review and Update: We continue this important work for our students. The goal is to have a broad framework for changes/updates to milestones and mentoring structure by February for implementation starting in Fall 2022. We started with strategic planning to define the desired outcomes for the design of our Training Program. We have reviewed data from current students, graduating students (exit interviews), faculty, head-search survey, DEI survey, Ology Marketing report, and committee-gathered data from other BME programs’ milestone structures. These discussions have led to many great ideas, which we are just now starting to map to our desired learning outcomes across the length of the PhD program to develop a new plan for the most effective milestones/requirements and support systems to achieve our vision for the training program. Stay tuned for updates, and solicitations for feedback. As always, please talk with any member of the Graduate Committee at any time if you have ideas/suggestions/concerns. Later this spring we will begin discussions on improvements to the graduate curriculum. Thanks for your support.
Undergraduate Curriculum Committee Update
In Spring 2022, the UCC will continue working on introducing BME pathways. The UCC will request each Research Area to present the corresponding Pathway to sophomore students in the next month. This will help the students to decide on their area of specialization. The presentation should clarify the knowledge and skills the students would gain as well as the future career opportunities for each BME Pathway. The UCC will also work on developing the Engineering Design courses for the re-structured undergraduate program, including the Junior and Senior Design and professional development courses. The committee will create a task force for collecting benchmark data on curriculum redesign for the future ABET accreditation. Also, the UCC will work on increasing our offering of honors courses or honors sections within courses.

2022 National Theme is Black Health and Wellness. According to the Association for the Study of African American Life and History which sets the annual focus, "This theme acknowledges the legacy of not only Black scholars and medical practitioners in Western medicine, but also other ways of knowing (e.g., birthworkers, doulas, midwives, naturopaths, herbalists, etc.) throughout the African Diaspora. January is Mentoring Month
The Graduate Student Peer Mentoring groups are running again this semester and new first years have now been paired up with senior students. BMES Undergraduate Peer mentoring is also getting underway and sophomores have the opportunity to participate in a mentoring group led by 1-2 upper classmen or in individual mentoring pair with a more senior student. Mentors and mentees participate in one large group and a small-group or meeting as a pair each month.

Business Services Updates
Going forward, Purdue Travel has been restructured in order to assist with the Concur expense report process for employees who travel. Please see the below update on the new process and contact information for questions:
Purdue Travel will provide all employees with a central, convenient way to submit travel receipts and business mileage, and then compile the travel expense report on the traveler’s behalf. With this change, there is now a team dedicated to the creation of expense reports. Based on a completed pilot program, the centralization of travel expense reports is expected to significantly reduce the time to create an expense report and reimburse the traveler to just four to seven days. By transitioning this work to dedicated Purdue Travel staff, employees will have more time to focus on their core responsibilities.
All employees or their delegates will need to submit expenses for reimbursement using one of the following methods upon returning from business travel:
* Add business expense receipts to the receipt store in the Concur mobile app. If travel was booked through Concur or Anthony Travel, Purdue Travel will reach out to the traveler/delegate to initiate the expense report process.
* Attach business expense receipts in an email to purduetravel(a)purdue.edu<mailto:purduetravel@purdue.edu>. Travelers or their delegates must initiate this process on their own by emailing the receipts.
A list of required information and documentation<https://www.purdue.edu/procurement/travel/expenses/index.php> is available on the Travel website under the “Expense Reimbursement” header.
For business mileage reimbursement to Purdue employees, either of the above processes may be used. The traveler will need to provide a copy of their mileage log or screenshots showing the distance of business mileage traveled (e.g., using Google Maps). The traveler also must provide the reason for travel in the comment box in Concur or in the email to purduetravel(a)purdue.edu<mailto:purduetravel@purdue.edu>.

“New NanoBiotechnology for Sensing and Imaging of Single Live Cells: From Fundamental Discoveries to Biomedical Innovations,” presented by X. Nancy Xu, PhD, Professor in Biomedical Engineering, Biomedical Sciences, Biochemistry, and Chemistry, Old Dominion University
Abstract: Single cells are building blocks of all living organisms. Understanding of functions of individual live cells and their communications is essential to rationally design and engineer effective diagnosis and therapies. New tools for detecting and targeting specific individual cells would revolutionize disease diagnosis and treatments. Single live cells also offer unique inspiration for rational design and engineer biocompatible smart materials and devices for bioengineering innovation (e.g., self-powered, self-repaired, self-assembled, self-replicated “live” robots). To achieve these transformative potentials, it is crucial to depict how individual live cells function and communicate to enable specific functions in cell culture, tissues, live animals, and ultimately in human in real time at single-cell and single-molecule resolutions. Interactions of couples of molecules in single live cells can induce a cascade of vital cellular responses and alter cellular and tissue functions. Current tools are unable to real-time study molecular machineries of single live cells with adequate quantitation, spatial and temporal resolutions and over a desired long period of time. We have pioneered the development of a set of powerful new technologies, including photostable single plasmonic nanoparticle imaging probes, single molecule nanoparticle optical biosensors (SMNOBS), and far-field photostable optical nanoscopy (PHOTON). We have demonstrated that these new tools can overcome the drawbacks of fluorescence-based imaging platforms for dynamic, single molecule and multiplexing imaging of single live cells with superior temporal and spatial resolutions and over a long period of time (hours, days, weeks). We have used these new tools to real-time study: (i) molecular cascades of signaling transduction pathways of single live cells; (ii) molecular mechanisms of multidrug resistance of single live cells; (iii) efficacies of single drug nanocarriers; (iv) native environments of developing embryos and differentiation of embryonic stem cells into cardiomyocytes and neurons; and (v) rare subsets of single cancer stem cells (CSCs) in clinical tissues for exploring new paradigm of early cancer detection and targeted therapy. We are currently developing new nanobiophotonic sensing and imaging platforms for real-time large-scale recording and modulation of neuron-neuron communicates, aiming to understand how brain develops and functions and how to effectively detect, diagnose and treat brain diseases. In this seminar, I will describe these new nanobiotechnologies and their innovative biomedical engineering applications. The work was supported by NIH and NSF.

Bio: Dr. X. Nancy Xu is the AAAS Fellow and Professor of Biomedical Engineering, Biomedical Sciences, Biochemistry, and Chemistry at Old Dominion University (ODU). During her tenure at ODU, Dr. Xu has built a state-of-the-art interdisciplinary research laboratory, and successfully developed a nationally and internationally recognized and well-funded interdisciplinary research program at the interface of chemistry, biology, engineering, and medicine. Dr. Xu has successfully directed NSF and NIH funded interdisciplinary research programs and she has demonstrated a track-record of accomplishments in successfully competing for highly prestigious federal research grants, including NSF NIRT, NSF BRAIN Initiative, NIH R01, and DoD MURI awards. Dr. Xu has pioneered the development of a set of powerful new sensing and imaging technologies for biomedical applications. She is especially well recognized for her pioneering work in single nanoparticle optics, single nanoparticle plasmonic spectroscopy, single molecule sensing, and single live cell imaging. Dr. Xu holds a world-wide patent on DNA Biosensors and has consistently published her work in high-impact peer-reviewed journals. Dr. Xu has led and developed several new graduate and undergraduate courses and she has taught 26 different courses on various topics including biosensing, imaging, biomedical sciences, biomedical engineering and nanobiotechnology. Dr. Xu has served as an expert reviewer and panelist for 15 national and international funding agencies, 62 NIH grant review panels and 26 NSF grant review panels. Dr. Xu has led and served as a track director/coordinator for an interdisciplinary joint PhD program in Biomedical Sciences between ODU and Eastern Virginia Medical School. and as the member of advisory board of Frank Reidy Research Center for Bioelectrics. Dr. Xu has received several prestigious national awards (e.g., Nano 50 Innovator Award, Nano 50 Technology Award, NSF BRAIN Initiative EAGER Award, Finalist of NIH Follow That Cell Challenge, ACS Roland F. Hirsch Award, and AAAS Mentor Award) and university awards (e.g., Distinguished Research Award, Faculty Research Achievement Award, University Presidential Diversity Champion Award, student-nominated Shining Star Recognition and Most Inspired Faculty Member Designation). Dr. Xu received her B.S. from Xiamen University and her Ph.D. from the University of Mississippi. “Electro-Fluidics for Health and Environmental Monitoring,” Mehdi Javanmard, Ph.D., Associate Professor of Electrical & Computer Engineering, Rutgers University
Abstract: In this talk, I will discuss my group’s work on fabricating micro- and nanosensing platforms for health and environmental monitoring. In the first part of my talk, I will discuss the fabrication of novel electronic sensing modalities both in vitro and in vivo and the role they can play in electroceutical applications for monitoring chronic inflammatory illnesses, wounds, and infectious diseases. I will also discuss domain specific use of machine learning to enable data-driven biosensors for improvement in accuracy and specificity. Finally, I will talk about my group's work in environmental monitoring.
Bio: Mehdi Javanmard is an Associate Professor in the Electrical and Computer Engineering Department at Rutgers University. He received the BS (2002) from Georgia Institute of Technology and the MS (2004) and PhD (2008) all in electrical engineering from Stanford University. During his time at Stanford, he worked on the development of electronic microfluidic platforms for low-cost genomic and proteomic biomarker detection, and later worked as a Senior Research Engineer at the Stanford Genome Technology Center. In Fall 2014, he joined the Electrical and Computer Engineering Department, Rutgers University as an Assistant Professor. His research interests include developing portable and wearable technologies for continuous health monitoring and understanding the effects of the environment on health. In 2017, he was recipient of the Translational Medicine and Therapeutics Award by the American Society for Clinical Pharmacology and Therapeutics for his group's work in point of care diagnostic tools for assessing patient response to cancer therapies. He has received various awards as a Principal Investigator from the National Science Foundation, National Institutes of Health, DARPA, and the PhRMA foundation to support his research. He was awarded the National Science Foundation CAREER Award for young faculty in 2019 and the DARPA Young Faculty Award in 2020. In 2018, he co-founded RizLab Health, a startup company dedicated to enabling point-of-care hematology analysis.

Wednesday, February 9, 1:30 pm, Location MJIS 1083. Presenter: Sarah Libring. “Using Fibronectin-Based Drug Screening Platforms for Precision Medicine of Metastatic Breast Cancer Patients.” Open to all undergraduate and graduate researchers. Lunch will be provided.
Breast cancer is the most commonly diagnosed cancer in women. Current practice involves staining a biopsy of the primary tumor for hormone receptors (ER, PR, and HER2) in order to segment patients into more homogenous treatment groups. However, these clinical subtypes are often not representative of how the cancer cells will respond to treatment once the disease has metastasized. We have developed a 3D culture platform utilizing fibrillar fibronectin that facilitates breast cancer cell expansion and maintains cell behavior ex vivo. Using cells isolated from pleural effusions in metastatic breast cancer patients, we are using this platform to compile ex vivo drug sensitivity against 11 common chemotherapeutics with the patients’ medical history and clinical treatment response. We are also utilizing the fibrillar fibronectin platform to explore dormancy of breast cancer cells once disseminated to the lungs. We hope to translate these results to a Phase II Clinical Trial at the Netherlands Cancer Institute using the scaffolds as a predictive drug screening platform.

After carefully considering input from students, postdocs, faculty and program administrators who participated in the program, I have decided to offer a series, RAISING A RESILIENT SCIENTIST<https://www.training.nih.gov/workshops_and_trainings_for_extramural_princip…>, for faculty and administrators who mentor students and postdoctoral fellows in the biomedical, behavioral, and social sciences. The goal of the Raising a Resilient Scientist series is to promote the mental health and well-being of the academic research community by supporting faculty and administrators to develop self-management, relationship-management, and mentoring skills. A variety of topics will be covered with a specific focus on communication and shared problem-solving to help trainees deal with the inevitable setbacks experienced in high knowledge environments. There will be five units in the series; each unit will consist of a 75-minute lecture, which can be watched in real-time or asynchronously, followed by a 1-hour facilitated discussion with peers. Abstract: One of the greatest consequences of Traumatic Brain Injuries (TBI) are the resulting long-term effects, especially in milder cases where the damage is initially subclinical and thus lacking acutely observable manifestations that can compound significantly over time. Among these chronic issues, Alzheimer’s Disease (AD) is one of the most serious. While a link between TBI and AD has been established, it has not yet been mechanistically elucidated. Multiple hypotheses have been postulated based on various animal/clinical models, which have contributed a great deal to our knowledge base and implicated several key pathways. While extremely valuable, these in vivo procedures and analyses are physiologically and ethically complex: there is currently no model capable of visualizing TBI-induced sub-cellular damage in the seconds immediately following injury, and the subsequent associated long-term changes. It is clear that additional investigative tools are needed, and while in vitro methodologies generally offer the type of resolution required, no such model replicates this phenomenon. Therefore, we introduce the “TBI-on-a-chip” concussive model prototype, with a series of concomitant targeted-experiments to address this currently unmet need.
We hypothesize that both the primary, mechanical injury and long-term associated damages (secondary injury) can be successfully recapitulated in vitro, and utilized for mechanistic studies into trauma induced neurodegeneration. Further, we hypothesize that aggregation (β-amyloid) can be initiated by force–solo and demonstrated in our proposed system, further linking TBI with AD. Our ultimate goal is to facilitate translational discoveries by exploring the mechanisms underpinning TBI, acute and chronic, and the link to neurodegeneration.

Abstract:
Chronic kidney disease (CKD) affects 15% of Americans and dramatically increases their risk of skeletal fractures and fracture-related mortality. The progression of CKD is marked by abnormal biochemistries including disrupted mineral homeostasis and elevated parathyroid hormone (PTH). High PTH is linked to the bone alterations seen in CKD including cortical porosity and altered turnover. To mitigate the effects of sustained elevations in PTH, dialysis patients are commonly given calcimimetics which act by sensitizing the calcium-sensing receptor on parathyroid chief cells, lowering PTH synthesis and secretion. While calcimimetics are suggested to reduce fracture risk in dialysis patients, little is known about how these drugs impact bone tissue properties or whether they can be implemented in early CKD to prevent CKD-induced bone deterioration. Therefore, understanding how calcimimetics alter bone quality may reveal new strategies to prevent fractures in patients with CKD. While CKD represents a major global health crisis, nearly half of all CKD patients also have diabetes. Diabetes disrupts numerous physiologic systems and independently increases fracture risk by lowering bone mass and quality. Therefore, we have two major objectives in this work. Firstly, we developed a method for analyzing bone composition and material properties in a spatially resolved manner and found that calcimimetics increase mineral crystallinity and stiffness in newly formed periosteal bone while increasing relative mineralization, stiffness, and hardness in perilacunar bone. Further, we found that these properties depended on distance from the osteocyte lacunar wall. Secondly, we developed a novel combined murine model of type 1 diabetes and CKD which revealed a unique combination of detriments in structural and tissue-level bone microarchitecture, mechanical properties, and fracture toughness. Taken together, this work represents a thorough investigation of how diabetes and CKD alter bone quality on multiple scales and how calcimimetics improve bone quality in localized regions in early CKD.

Abstract: Congenital diseases of the kidney are the leading cause of chronic kidney disease in pediatric patients. Tissue engineering models used to investigate these diseases are limited by an immature phenotype. Models cultured in an extracellular matrix (ECM), a network of proteins and glycosaminoglycans surrounding cells and providing structural support that mimic the matrix found in development will be likely more mature. However, developing kidney ECM composition and structural dynamics are unknown. To address this gap, we studied ECM composition using mass spectrometry and organization by visualizing the ECM in 3D.
In this work, we used mass spectrometry to resolve ECM basement membrane and interstitial matrix dynamics between embryonic, perinatal, and adult kidneys. Surprisingly, we observed a transient upregulation of interstitial matrix structures that corresponded to dynamic 3D structures in the cortex (vertical fibers) and at the corticomedullary junction (medullary ray sheath fibers). Notably, in a model of abnormal Foxd1+ stromal cells, the vertical fibers were disorganized, and medullary ray sheath fibers were no longer associated with blood vessels, suggesting the dynamic 3D structures depended on stromal cell modulation.
One of the effects of abnormal kidney development is decreased amniotic fluid, which limits embryonic movement and subsequent limb development. In additional studies, we looked at the implications of the lost motility in the muscle dysgenesis (mdg) mouse on the development of the myotendinous junction (MTJ). The MTJ links contractile muscle with tendon. We found the ECM protein COL22A1 was specific to the developing MTJ as early as embryonic day (E)13.5. The development of the MTJ from a linear structure to a cap-like structure with invaginations in adolescent mice depended on muscle contraction. Furthermore, we used a model to decouple the muscle-tendon-bone complex at an ectopic lateral triceps insertion (Prrx1Cretg/+; Tbx3fl/fl). We observed disorganized tendon and MTJ markers at the termination of the ectopic lateral triceps muscle but negligible cartilage markers. Together, this indicated MTJ maturation depended on motility but not on the enthesis.
The information gleaned from our studies on how stromal cells affect dynamic 3D interstitial ECM structures and composition change during kidney development can be used as a template for 3D kidney culture systems. Combined with forelimb MTJ development, our results indicate the importance of the interstitial matrix in tissue morphogenesis.

Abstract: The aorta has many complex features including valve and vessel wall geometry, blood flow, and wall composition. Diseases such as aortic aneurysms and aortic valve lesions affect vessel function and may even lead to rupture, which can be fatal. However, current clinical diagnoses of aortopathies mainly rely on simple parameters such as diameter and growth rate. To better understand aortopathies and ultimately provide better patient diagnoses and treatments, it is important to investigate disease progression as well as the effect of vessel wall composition changes and hemodynamic forces on aortic biomechanics, such as strain and wall shear stress distribution. Preclinical research using small animals allows for disease progression to be studied while controlling outside factors. The next important step is to apply the methods used in the preclinical studies to human patient data. Both preclinical and clinical studies often focus on noninvasive, patient-specific methods for further characterizing the biomechanics of the aorta using advanced techniques such as 4D flow magnetic resonance imaging, 4D ultrasound, computational fluid dynamics (CFD), and fluid structure interaction (FSI) modeling. Yet the challenge of bridging these research techniques to a clinical setting remains. Factors such as financial costs, acquisition time, and ease of analysis must be considered. Therefore, the following document highlights two specific aims to extend our knowledge about the effects of aneurysms and aortic valve lesions. We will 1) characterize the regional effects of murine abdominal aortic aneurysms on strain over time, and 2) use CFD and FSI to simulate the hemodynamic effects on the thoracic aorta using both murine and human patient imaging data. Conducting research using clinically translatable methods of biomechanical characterization that consider the complexity of the aorta on a patient-specific basis will contribute to our understanding and lead to better patient outcomes.

Fourth Annual Conference for Associate Professors set for Feb. 23-24
The Susan Bulkeley Butler Center for Leadership Excellence in collaboration with the Office of the Provost will hold the Fourth Annual Conference for Associate Professors on Feb. 23-24 in Purdue Memorial Union. The conference is titled "Charting Your Path to Promotion." The conference will include a keynote address and two panel sessions. Over the course of the conference, attendees will have opportunities to network, share information and learn from others in ways that will enable associate professors, particularly women associate, to accelerate their progression to full professor. The panel sessions comprise speakers from within and outside Purdue. Although this conference targets associate professors, the strategies and insights offered by the keynote speaker and the panelists will be useful for all faculty members.
Conference details include:
o Keynote address: Elizabeth Field Hendry, provost and senior vice president for academic affairs, Queens College, CUNY.
o Panel session 1: "Managing Time: Service, Teaching, Mentoring, and Research."
o Panel session 2: “Promotion to Full Professor.”

"A new online submission form is now available to assist those who travel on official University-related business with the travel expense reporting process.
Last year, Purdue Travel announced that travel expense reports would be centralized through its department. The transition from having travelers submit their own expense reports to having the Purdue Travel department compile the travel expense report on a traveler's behalf was completed in December.
At that time, travelers or their delegates were given two methods to submit expenses for reimbursement upon returning from travel:
* Add business expense receipts to the receipt store in the Concur mobile app. If travel was booked through Concur, Purdue Travel will reach out to the traveler or delegate to initiate the expense report process.
* Attach business expense receipts in an email to purduetravel(a)purdue.edu<mailto:purduetravel@purdue.edu>. Travelers or their delegates must initiate this process on their own.
Now, the online form will assist with the expense reporting process and the following changes will be made to the two submission methods:
* Add business expense receipts to the receipt store in the Concur mobile app. If travel was booked through Concur, Purdue Travel will reach out to the traveler or delegate with the form to obtain additional information to complete the expense report.

Good Afternoon,
Going forward, Purdue Travel has been restructured in order to assist with the Concur expense report process for employees who travel. Please see the below update on the new process and contact information for questions:
Purdue Travel will provide all employees with a central, convenient way to submit travel receipts and business mileage, and then compile the travel expense report on the traveler's behalf. With this change, there is now a team dedicated to the creation of expense reports. Based on a completed pilot program, the centralization of travel expense reports is expected to significantly reduce the time to create an expense report and reimburse the traveler to just four to seven days. By transitioning this work to dedicated Purdue Travel staff, employees will have more time to focus on their core responsibilities.
All employees or their delegates will need to submit expenses for reimbursement using one of the following methods upon returning from business travel:
* Add business expense receipts to the receipt store in the Concur mobile app. If travel was booked through Concur or Anthony Travel, Purdue Travel will reach out to the traveler/delegate to initiate the expense report process.
* Attach business expense receipts in an email to purduetravel(a)purdue.edu<mailto:purduetravel@purdue.edu>. Travelers or their delegates must initiate this process on their own by emailing the receipts.
A list of required information and documentation<https://www.purdue.edu/procurement/travel/expenses/index.php> is available on the Travel website under the "Expense Reimbursement" header.
For business mileage reimbursement to Purdue employees, either of the above processes may be used. The traveler will need to provide a copy of their mileage log or screenshots showing the distance of business mileage traveled (e.g., using Google Maps). The traveler also must provide the reason for travel in the comment box in Concur or in the email to purduetravel(a)purdue.edu<mailto:purduetravel@purdue.edu>.

New NanoBiotechnology for Sensing and Imaging of Single Live Cells:

Abstract: Single cells are building blocks of all living organisms. Understanding of functions of individual live cells and their communications is essential to rationally design and engineer effective diagnosis and therapies. New tools for detecting and targeting specific individual cells would revolutionize disease diagnosis and treatments. Single live cells also offer unique inspiration for rational design and engineer biocompatible smart materials and devices for bioengineering innovation (e.g., self-powered, self-repaired, self-assembled, self-replicated “live” robots). To achieve these transformative potentials, it is crucial to depict how individual live cells function and communicate to enable specific functions in cell culture, tissues, live animals, and ultimately in human in real time at single-cell and single-molecule resolutions. Interactions of couples of molecules in single live cells can induce a cascade of vital cellular responses and alter cellular and tissue functions. Current tools are unable to real-time study molecular machineries of single live cells with adequate quantitation, spatial and temporal resolutions and over a desired long period of time. We have pioneered the development of a set of powerful new technologies, including photostable single plasmonic nanoparticle imaging probes, single molecule nanoparticle optical biosensors (SMNOBS), and far-field photostable optical nanoscopy (PHOTON). We have demonstrated that these new tools can overcome the drawbacks of fluorescence-based imaging platforms for dynamic, single molecule and multiplexing imaging of single live cells with superior temporal and spatial resolutions and over a long period of time (hours, days, weeks). We have used these new tools to real-time study: (i) molecular cascades of signaling transduction pathways of single live cells; (ii) molecular mechanisms of multidrug resistance of single live cells; (iii) efficacies of single drug nanocarriers; (iv) native environments of developing embryos and differentiation of embryonic stem cells into cardiomyocytes and neurons; and (v) rare subsets of single cancer stem cells (CSCs) in clinical tissues for exploring new paradigm of early cancer detection and targeted therapy. We are currently developing new nanobiophotonic sensing and imaging platforms for real-time large-scale recording and modulation of neuron-neuron communicates, aiming to understand how brain develops and functions and how to effectively detect, diagnose and treat brain diseases. In this seminar, I will describe these new nanobiotechnologies and their innovative biomedical engineering applications. The work was supported by NIH and NSF.

Bio: Dr. X. Nancy Xu is the AAAS Fellow and Professor of Biomedical Engineering, Biomedical Sciences, Biochemistry, and Chemistry at Old Dominion University (ODU). During her tenure at ODU, Dr. Xu has built a state-of-the-art interdisciplinary research laboratory, and successfully developed a nationally and internationally recognized and well-funded interdisciplinary research program at the interface of chemistry, biology, engineering, and medicine. Dr. Xu has successfully directed NSF and NIH funded interdisciplinary research programs and she has demonstrated a track-record of accomplishments in successfully competing for highly prestigious federal research grants, including NSF NIRT, NSF BRAIN Initiative, NIH R01, and DoD MURI awards. Dr. Xu has pioneered the development of a set of powerful new sensing and imaging technologies for biomedical applications. She is especially well recognized for her pioneering work in single nanoparticle optics, single nanoparticle plasmonic spectroscopy, single molecule sensing, and single live cell imaging. Dr. Xu holds a world-wide patent on DNA Biosensors and has consistently published her work in high-impact peer-reviewed journals. Dr. Xu has led and developed several new graduate and undergraduate courses and she has taught 26 different courses on various topics including biosensing, imaging, biomedical sciences, biomedical engineering and nanobiotechnology. Dr. Xu has served as an expert reviewer and panelist for 15 national and international funding agencies, 62 NIH grant review panels and 26 NSF grant review panels. Dr. Xu has led and served as a track director/coordinator for an interdisciplinary joint PhD program in Biomedical Sciences between ODU and Eastern Virginia Medical School. and as the member of advisory board of Frank Reidy Research Center for Bioelectrics. Dr. Xu has received several prestigious national awards (e.g., Nano 50 Innovator Award, Nano 50 Technology Award, NSF BRAIN Initiative EAGER Award, Finalist of NIH Follow That Cell Challenge, ACS Roland F. Hirsch Award, and AAAS Mentor Award) and university awards (e.g., Distinguished Research Award, Faculty Research Achievement Award, University Presidential Diversity Champion Award, student-nominated Shining Star Recognition and Most Inspired Faculty Member Designation). Dr. Xu received her B.S. from Xiamen University and her Ph.D. from the University of Mississippi. Abstract: The functionality versatility of polymeric and nucleic acid materials provides diverse engineering opportunities for the detection and therapeutic targeting of disease-associated biomolecules. My work has encompassed nanoscale and microscale technologies to address challenges in cancer therapeutics and diagnostics, from targeting the expression of dysregulated genes to probing rare protein biomarkers. I will first describe the design of a highly potent small interfering RNA nanoparticle delivery system that leverages approaches in nucleic acid engineering and polymer chemistry. Towards overcoming analytical barriers in clinical diagnostics, I will then describe the development of ultrasensitive single-molecule detection technologies to measure low abundance proteins. By utilizing powerful molecular amplification methods and simple signal readouts, these digital enzyme-linked immunosorbent assay (ELISA) platforms enable the detection of low attomolar (10-18 M) protein concentrations, with over four orders-of-magnitude improvements in sensitivity over conventional protein detection methods. Importantly, these methods democratize ultrasensitive protein detection, enabling high-throughput, high-multiplex digital measurements to be carried out with common laboratory instrumentation. Finally, I will highlight the application of digital ELISA technology towards the development of an ultrasensitive blood test for a retrotransposon-encoded protein as a highly specific multi-cancer biomarker. Overall, the development of these technologies can pave the way towards the future integration of materials and biomolecular engineering approaches with ultrasensitive bioanalytical tools for accelerating biomarker signature discovery and precision medicine.
Bio: Connie is an NIH F32 Ruth L. Kirschstein postdoctoral research fellow in Dr. David Walt's lab at Brigham and Women's Hospital and the Wyss Institute for Biologically Inspired Engineering at Harvard University. She obtained her B.S. in chemical engineering from Yale University, where she worked with Dr. Paul Van Tassel in designing porous layer-by-layer polymer films for tissue engineering applications and received a Barry M. Goldwater Scholarship. Connie pursued her Ph.D. in chemical engineering at MIT, working in Dr. Paula Hammond's lab on polymer-based RNA interference delivery systems for cancer therapeutics. She was the recipient of multiple fellowships during her Ph.D. training, including an NSF Graduate Research Fellowship, Ludwig Center for Molecular Oncology Graduate Fellowship, and MIT Presidential Fellowship. In her current postdoctoral work, Connie has developed ultrasensitive single-molecule detection platforms for biomarker discovery and diagnostic applications.
