I appreciate that many of us also valued this opportunity to be able to interact with our bright and talented undergraduate students. There was interesting feedback from them and good discussions throughout the forum. I am sure many of us are still reflecting on the evening. * Young talked about the importance of being proactive in arranging special accommodations for exams in our last faculty meeting. Jo included some information in the Academic Programs Alert session. Please review it if relevant to you. We have gotten additional clarity on the staff side of our academic unit on the PU - Indy campus. We are approved to have one academic advisor, one administrative assistant, one research lab operations person, and one instructional lab operations person. Vitaliy and I are collecting information from the teaching faculty there to clarify the job responsibilities of the instructional lab operations person. We hope to have sufficient clarity by our next faculty meeting. We have a number of students who may need accommodations for final exams and faculty need to be proactive in scheduling with the Testing Center if they plan to use that. Below is some important information instructors should know if they choose to have Purdue Testing Services (PTS) administer their accommodated exams. * The steps to submit a Testing Instruction Form (TIF) for finals week are the same as any other exam with one notable difference. In the “select the exam group/s for this test” field, be sure to select your course that starts with Accommodated FINAL Exam. This is required before students will be able to schedule during finals week. *If you had already submitted a TIF for your final prior to receiving this email, PTS staff updated your TIF to the correct exam group * Hours of operation: * Monday - Friday | 8 a.m. - 10 p.m. * Exam start times: 8 a.m., 10:30 a.m., 1 p.m., 3:30 p.m., 6 p.m. and 7 p.m. * Saturday | 8 a.m. - 4:30 p.m. * Exam start times: 8 a.m., 10:30 a.m., Noon and 1 p.m. * Due to the volume of exams we administer, we will not start any exam outside of the start times mentioned above. GRADUATE PROGRAM UPDATE Fellowship Opportunity The Bilsland Dissertation Fellowship provides support for outstanding PhD candidates in their final year or semester of doctoral degree completion. The goal of the fellowship is to allow exceptional candidates to focus exclusively on finishing their dissertation. Salary & Usage: * This fellowship includes $30,000 annual salary, tuition remission, medical insurance premium contribution, and coverage of most fees. Excellence in Teaching Awards Competition Applications are now being accepted for the Midwestern Association of Graduate Schools (MAGS) 2024 Excellence in Teaching Awards Competition. Jane Grande-Allen, PhD, Department of Bioengineering, Rice University, will present “Biophysically Faithful Biomaterial Platforms for Cardiovascular and Intestinal Mechanobiology.” Abstract: The material behavior of many biological tissues is due to their unique microstructural arrangements of fibrous extracellular matrix (ECM) proteins, i.e., collagen and elastin, within the more amorphous matrix. The orientation of these fibers, and their segregation into discrete regions within the tissues, often gives rise to anisotropy and unique biological stress-strain behavior that enables the essential function of the tissues. Layered or segregated structuring allow hierarchical tissue organization in a manner designed to withstand external forces efficiently while protecting more delicate tissues and cells from damage. These structure-function relationships within biological tissues have been studied for decades but have not been widely translated into the creation of biomimetic scaffolds for use in tissue engineering and in vitro analyses of cell and tissue biology. The Grande-Allen research group has focused on integrating these structural and material characteristics into hydrogel and fibrous biomaterials using a range of fabrication techniques including molding, photolithography, electrospinning, and 3D printing. The majority of our investigations have addressed heart valve disease, which is widely prevalent in our society, with valve replacement or repair in almost 100,000 people in the United States and 275,000 people worldwide each year. More recently, we have translated our fabrication strategies to generate biomaterial platforms for investigating intestinal epithelial cell biology and enteric diseases. Biography: Jane Grande-Allen is the Isabel Cameron Professor of Bioengineering at Rice University. Her research group investigates the structure-function-environment relationship of soft connective tissues through bioengineering analyses of the extracellular matrix and cell mechanobiology, with a focus on cardiovascular and intestinal diseases, as described in >180 peer-reviewed publications. Dr. Grande-Allen received a BA in Mathematics and Biology from Transylvania University in 1991 and a PhD in Bioengineering from the University of Washington in 1998. After postdoctoral research in Biomedical Engineering at the Cleveland Clinic, she joined Rice University in 2003 and was promoted to full professor in 2013. Dr. Grande-Allen is a Fellow of AIMBE, IAMBE, BMES, AAAS, AHA, and the Society for Experimental Mechanics. She served on the BMES Board of Directors and Executive Board from 2009-2022 and was BMES Secretary from 2020-2022. Kolbinger; Kyoko Yoshida; Benjamin J. Landis Abstract: Volume and pressure overload induce cardiac remodeling that may lead to heart failure when left untreated. Conditions such as pregnancy, lactation, and aortic stenosis are common etiologies of pressure and volume overload. However, sex- and female-specific variables and recovery are often overlooked in studies of cardiac overload. One of the most common modalities used to study cardiac function, 2D ultrasound, does not fully capture changes that may occur to heart shape and performance given that it is limited to a single imaging plane. To improve our understanding of pressure and volume overload we propose mouse models of lactation, chronic hypertension with pregnancy, and reversible transverse aortic constriction. We hypothesize that four-dimensional ultrasound will reveal the unique ways in which volume and pressure overload in the setting of pregnancy, lactation, and aortic constriction affect cardiac remodeling. Most experts debate the importance of tonotopy versus temporal coding, but it is still unclear how disruptions of these mechanisms are influenced by sensorineural hearing loss (SNHL). SNHL is complex, but as a diagnosis, it groups variable degrees of inner and outer hair cell (IHC/OHC) and cochlear synapse (CS) damage which likely results in different patterns of pitch perception. With careful stimulus design and electrophysiological data harmonization between animal and human subjects, we are developing a framework to more directly link alterations in neural coding to pitch perceptual outcomes. I will present progress related to my NRSA F30 project (Title: Place and Time Processing of Pitch in the Context of Cochlear Dysfunction), which was funded thanks to the fruitful discussions and feedback sparked by this seminar two years ago. Evans, Dr. Fang Huang, Dr. Krishna Jayant Abstract: Alzheimer’s disease (AD) is a neurodegenerative disease often observed in elderly patients (≥65y/o). Symptoms of AD include, but are not limited to, loss of learning and memory function and epileptic seizures. Symptoms of AD are rooted in dysfunctional synapses between neurons, leading to impaired synaptic plasticity. Several genes are associated with AD. For example, mutations in the gene encoding drebrin (DBN) are strongly implicated in AD. Drebrin1A (DBN1A) is an actin-stabilizing protein prominently expressed in dendritic spines and helps maintain the cytoskeletal structure. The role of DBN1A in structure-based synaptic plasticity has not been described. Dendritic spines are protrusions on the postsynaptic neuron and are composed of a spine head and spine neck. Although spine head enlargement is commonly seen in synaptic plasticity, an understanding of how the regions between the spine neck, head-neck interface, and spine head are regulated and its relation to synaptic plasticity is lacking. A recent discovery of DBN1A and CaMKIIβ interactions near this interface has been reported. However, it is still unclear how CaMKIIβ-DBN1A increases F-actin bundling in dendritic spines. Based on the literature, we suspect that CaMKIIβ-DBN1A binding plays a significant role in structure-based synaptic plasticity. We hypothesize that during synaptic plasticity, structural remodeling of dendritic spines requires CaMKIIβ-DBN1A to stabilize the F-actin pool at the head-neck interface. To test this hypothesis, we propose to use super-resolution microscopy (SRM) and molecular dynamic (MD) simulations to provide comprehensive evidence that CaMKIIβ-DBN1A complex is critical F-actin bundling during synaptic plasticity in dendritic spines. The function of the brain relies on its ability to maintain a normal homeostatic balance between its mechanical environment and metabolic demands, which can be greatly altered in the cases of neurodegeneration or traumatic brain injury. It has been a challenge in the field to quantify the dynamics of the macroscale tissue and cerebrospinal fluid flow in human subjects on a patient-specific basis over the many spatial and temporal scales that it relies upon. Non-invasive imaging tools like structural, functional, and dynamic MRI sequences provide modern researchers with an unprecedented view into the human brain. Our work leverages these sequences by developing novel, open-source pipelines to 1) quantify the biomechanical environment of the brain tissue over 133 functional brain regions, and 2) estimate real-time cerebrospinal fluid velocity from the noise on functional MRI by employing breathing regimens to enhance fluid motion. These pipelines provide a comprehensive view of the macroscale tissue and fluid motion in a given patient. Additionally, we sought to understand how the transmission of macroscale forces, in the context of traumatic brain injury, contribute to neuronal damage by 3) developing a digital twin to simulate 30-200 g-force loading of 2D neuronal cultures and observing the morphological and electrophysiological consequences of these impacts in vitro by our collaborators. Taken together, we believe these works are a steppingstone that will enable future researchers to deeply understand the mechanical contributions that underly clinical neurological outcomes and perhaps lead to the development of earlier diagnostics, which is of dire need in the case of neurodegenerative diseases. Dr. Alves is a Chemical and Biomolecular Engineer, Associate Professor and the Director of Translational Research for the Department of Emergency Medicine in addition to having an affiliate faculty appointment at Purdue University in Biomedical Engineering. The Alves Lab explores highly interdisciplinary research interests applying engineering and biophysical principles and designs to create translational technologies and treatments in the area of blood coagulation and fibrinolysis. The primary focus of research in the laboratory is to develop targeted clot digesting therapeutic agents to more safely digest clinically relevant blood clots to treat pulmonary embolism (PE, blood clots in the lungs), deep vein thrombosis (DVT), and ischemic stroke. This interdisciplinary research utilizes diverse enzyme assays, chemical synthesis, blood clot formation and digestion assays under shear, and other novel testing/delivery platforms that include multivalent branched molecules and nanoparticle drug delivery systems. Projects Include: (1) Leveraging multivalency to modulate enzyme activity and improve inhibitor selectivity (nanoparticle drug delivery and targeting). (2) Ex-vivo flowing models of clot digestion under shear using in-vivo like fluorescently labeled blood clots from human whole blood. (3) Development of novel fibrinolytic diagnostics tools to assess coagulation state. (4) Mouse models of clot formation/targeting/digestion utilizing intravital microscopy.
