I flew to another conference on Saturday afternoon. I have just returned to Indianapolis. * Many of us had a wonderful experience at the BMES annual conference. It was so good to see our faculty, staff, and students. It was so good to meet our alumni and old colleagues at the conference, and particularly, at the school reception. The IUPUI integration is going well. * I am pleased to inform you that Dr. Rachel Surowiec has decided to join the Weldon School of Biomedical Engineering as an Assistant Professor with a tenure-track appointment. Abstract: Achieving a Circular and Sustainable Economy is increasingly urgent to minimize and mitigate devastating effects on the planet associated with continuing business as usual. According to the Ellen MacArthur Foundation, which popularized the concept, a Circular Economy is one that designs out waste and pollution, that keeps products and materials in use, and that regenerates natural systems. These principles apply in the context of manufactured products, as well as to the processes and operations behind those products. This presentation will consider various programs, tools and initiatives helping minimize environmental impact and delivering circular and sustainable products and manufacturing within Sun Chemical, the global leader for pigments, inks, coatings, and advanced materials. A particular focus will be on circular packaging applications that are enabled by Sun Chemical's technologies. Bio: Dr. Nikola Juhasz is the Global Technical Director for Sustainability at Sun Chemical, where she engages with internal and external stakeholders to formulate sustainability-driven technical strategies and oversees the corresponding innovation and product development programs across all of Sun Chemical's pigments, inks, coatings, and adhesives product lines and technology platforms. She represents Sun Chemical at key global cross-industry organizations focused on sustainability and the future of packaging, including the Sustainable Packaging Coalition (SPC), and the Association of Plastic Recyclers (APR), among others. Alan Grodzinsky, PhD, Departments of Biological, Electrical and Mechanical Engineering, Center for Biomedical Engineering, MIT, will present "Human Osteochondral-Synovium Co-Culture Model of Cartilage Degradation & Therapeutic Intervention: Relevance to Post-Traumatic OA on Earth and in Space." Abstract: We established a human osteochondral-synovium microphysiological system incorporating a single impact mechanical load that emulates initial pathways and biomarker-release of post-traumatic osteoarthritis-like disease; we tested the effects of anti-catabolic dexamethasone and pro-anabolic IGF-1 to ameliorate PTOA-like processes. In addition, we tested the effects of microgravity and radiation ± drugs on this system on the International Space Station. Overall, 88 human distal femurs (Collins Grade 0-2) from 55 donors (M/F age 23-83) were obtained. We quantified release of inflammatory cytokines, cell viability, release of sGAG and ARGS-aggrecan and CTX-II collagen fragments, metabolomics/proteomics assays, and the response to Dex and IGF-1. Two Grade-0 knee-tissues were launched to the ISS on SpaceX-21 for 3-week experiments (M/31, F/47). Analyses showed release of inflammatory cytokines, mainly from synovium, on Earth and in Space, causing increases in release of sGAG and ARGS-aggrecan fragments, the earliest biomarkers of disease by 2-weeks in culture. Metabolites and peptide markers of inflammation were also increased. While Dex+IGF-1 decreased the release of sGAG and ARGS-aggrecan over-all, there was distinct donor-to-donor variability. Metabolomics showed increased oxidative stress and inflammation under spaceflight conditions; Dex+IGF-1 substantially reduced GAG release from one donor system in Space, but the second donor showed less response. Ongoing proteomic analyses focus on extent and kinetics of biomarker release, to aid in the timing and choice of therapeutics. Despite the increasing number and complexity of ongoing clinical trials for OA/PTOA therapeutics, the discovery and choice of drug(s) is only half the battle: the other half is delivery of drug(s) to the intended target(s). Most trials for knee OA/PTOA now involve intra-articularly injected drugs, but there is little or no evidence that injected drugs have reached their target tissues in the dose and duration needed. Does failure of such trials mean that drugs were not efficacious, or that potentially efficacious drugs never reached their intended target at the proper dose/duration due to drug delivery inadequacies? Biography: Alan Grodzinsky, ScD, is Professor Emeritus of Biological, Electrical, and Mechanical Engineering in the Departments of Biological Engineering (BE), Electrical Engineering and Computer Science, and Mechanical Engineering at MIT. His research interests center on osteoarthritis and degenerative joint disease; complex microphysiological organ-on-a-chip systems for drug discovery and drug delivery for osteoarthritis; tissue engineering for cartilage repair; cellular mechanotransduction; and molecular, cellular and tissue nanomechanics. He has published over 350 refereed journal articles and reviews in these fields. While his undergraduate and graduate training at MIT was originally in Electrical Engineering and Computer Science, he is a founding faculty member of MIT's Biological Engineering Department. At MIT, he developed two required graduate courses in BE Department and published a textbook on these subjects (Fields, Forces and Flows in Biological Systems, Garland Science, 2011) focusing on transport and biomechanical properties of connective tissues such as cartilage, skin, tendons and ligaments. He has received numerous awards including the NIH MERIT Award (for research on Cartilage Mechanobiology), the Melville Medal of the ASME, the Kappa Delta Prize of the AAOS, the Borelli Award of the Amer Soc of Biomechanics, the Outstanding Achievement in Mentoring Award of the Orthopaedic Research Society (ORS), the Distinguished Investigator Award of the ORS/AAOS, and the Lifetime Achievement Award of the Osteoarthritis Research Society International (OARSI). Abstract The human brain can integrate information from different sensory modalities to produce a unified representation of the external world. Temporally congruent auditory and visual stimulation facilitates multisensory perception and integration, allowing us to better process and understand the information presented to us, especially in the presence of background noise and for individuals with hearing difficulties. The influences of visual speech cues on behavioral speech perception are vastly studied. Functional magnetic resonance imaging and electrophysiological studies have mostly evaluated auditory cortical responses. Little is known about the influence of visual speech production cues on the auditory brainstem phase-locked ensemble responses in normal and individuals with sensorineural hearing loss. To address this knowledge gap, we evaluated the effects of congruent- and incongruent-audiovisual (AV) speech on scalp-recorded brainstem frequency following response (FFR) and the cortical acoustic change complex (ACC). For the FFR, both phase locking to envelope periodicity (FFRENV) and to the temporal fine structure (FFRTFS) were evaluated. I will present our initial results showing that visual speech cues do influence evoked responses at both cortical and brainstem levels, albeit differently. These results appear to suggest that neural activity relevant to audio-visual integration may be already emerging at the brainstem level. Thus, these measures have the potential to be developed as clinical metrics for evaluation of audio-visual integration, and as measures to evaluate hearing-aid outcomes and prognosis. Abstract: We are witnessing the dawn of an era of personalized medicine, driven by technological advances in cell therapy, immunotherapy, and regenerative medicine. In this era, bioengineering capabilities that enable precise control and biofunction design, both at the cellular and tissue levels are increasingly becoming important, especially with regards to supporting patient-specific therapeutics and drug development. This talk will highlight our past and ongoing research efforts to develop enabling microengineering technologies for biofunction design at the cellular and tissue levels, and will showcase potential biomedical applications in cancer immunotherapy, regenerative medicine, and drug development. Biography: Dr. Okeyo is currently a technical information scientist (TIS) and a project manager at Jackson Laboratory Japan. He obtained his PhD degrees in mechanical engineering from Kyoto University, Japan, in 2010 and thereafter worked for two years as a researcher at the Central Research Laboratory of Hitachi Ltd, Japan. He then became an assistant professor at the Department of Mechanical Engineering of the university of Tokyo in Japan before moving to Kyoto University as a senior lecturer in charge of teaching and research supervision. Alves, PhD, within the Department of Emergency Medicine at the Indiana University School of Medicine (IUSM), is seeking a highly motivated and independent post-doctoral research fellow. Dr. Alves is a Chemical and Biomolecular Engineer, Associate Professor and the Director of Translational Research for the Department of Emergency Medicine in addition to having an affiliate faculty appointment at Purdue University in Biomedical Engineering. The Alves Lab explores highly interdisciplinary research interests applying engineering and biophysical principles and designs to create translational technologies and treatments in the area of blood coagulation and fibrinolysis. The primary focus of research in the laboratory is to develop targeted clot digesting therapeutic agents to more safely digest clinically relevant blood clots to treat pulmonary embolism (PE, blood clots in the lungs), deep vein thrombosis (DVT), and ischemic stroke. This interdisciplinary research utilizes diverse enzyme assays, chemical synthesis, blood clot formation and digestion assays under shear, and other novel testing/delivery platforms that include multivalent branched molecules and nanoparticle drug delivery systems. Projects Include: (1) Leveraging multivalency to modulate enzyme activity and improve inhibitor selectivity (nanoparticle drug delivery and targeting). (2) Ex-vivo flowing models of clot digestion under shear using in-vivo like fluorescently labeled blood clots from human whole blood. (3) Development of novel fibrinolytic diagnostics tools to assess coagulation state. (4) Mouse models of clot formation/targeting/digestion utilizing intravital microscopy.
