Due to the variety of operating conditions required for HPLC system #2 using the C-18 column we have devised a system to provide fair and efficient operation to all users.  We will alternate the system operating conditions on a weekly basis.  We will set up the system for the following week on Friday, which will allow the system to equilibrate over the weekend and be ready to run on Monday.   Let us know if you need us to modify the schedule and we would be happy to accommodate your needs, but the goal is to avoid changing the column and mobile phase every day.  While the operating conditions were designed for each experiment, you are welcome to run samples whenever you want.  Here is our tentative schedule:

Week of 5/27: C₃Bio program

C18 column (Agilent, XDB-C18, 4.6x50 mm, 1.8 micrometers), UV detector, and 2 mobile phases: 1) 0.1% Formic Acid in DI H20 and 2) 100% methanol

Week of 6/3: Fernanda Marisa da Cunha’s Feruloyl Esterase Assay

C18 column (EC 250/4.6 NUCLEOSIL 100-7 C18), UV detector, and two mobile phases: 1) 50 mL methanol and .25 mL acetic acid per L water and 2) .25 mL acetic acid per L methanol

Week of 6/10: He Shang-Guessmix Compounds for HPLC Analysis

C18 column (EC 250/4.6 NUCLEOSIL 100-7 C18), UV detector, and methanol and H2O mobile phase

At this point we will plan on repeating this cycle, but if there is a better method of sharing the column we will modify our strategy.
