Bio+Nano Journal Club Kick-off from NSAC

Dear all, It's my pleasure to announce that NSAC is going to resume its Journal Club activity starting next week. The theme for this semester would be "Bio+Nano". We believe there is a lot of overlap in terms of research and goal orientation between Bioscience and Nanotechnology, and the talks aim to bridge the two communities from Birck and Bindley so that we can learn what the fundamental problems are and what solution is technologically possible. The first talk will be given by Rohil Jain (from Prof. Cagri Savran's group). He will be presenting a brief review of technical aspects of detecting circulating tumor cells (http://www.sciencedirect.com.ezproxy.lib.purdue.edu/science/article/pii/S03…) The paper is attached to this email. As previously, some key features of this journal club would be: - Each session would be around 30-45 mins long. - The speakers will be those who are currently active in this field. Most of them will be from Bindley Bioscience Center. - A recent paper of substantial impact would be walked through. The metastatic process consists of several steps including primary tumor growth, invasion, intravasation into the blood or lymph vessels, extravasation, invasion into a secondary organ, secondary tumor initiation and finally metastatic outgrowth, all the while avoiding detection by the immune system and elimination by standard-of-care chemotherapies. While in vivo studies using mouse models are required to recapitulate metastasis as a whole, several aspects of drug discovery and compound development are not feasible using only in vivo approaches. On the other end of the spectrum, drug response studies using cells cultured on 2D polystyrene offer critical, but limited information when assessed in a non-biased manner. Moreover, 2D cell culture results can be misleading due to artificial interactions between the cells and the polymer growth surface. Therefore, we continually seek to utilize or develop robust, cost effective, 3D culture approaches that are be capable of yielding high content information for drug discovery and development. One current approach we are taking combines two commonly used methods of 3D culture; 1. The non-adherent round bottom culture dish, used to form a collective spheroid of tumor cells or some mixture of tumor and stromal cells, and 2. A gel based semi-solid support that allows cells to organize into three-dimensional structures. The technique consists of forming a spheroid in the round bottom dish for an optimized period of time and then physically transferring this multicellular structure to a hydrogel semisolid support. Similar approaches have overlaid the spheroid with a hydrogel, but we find that the transfer of the spheroid to a growth surface to establish an “on top” 3D culture is critical for the phenotypic manifestations shown below. The relative simplicity of this approach allows for compound addition during either or both phases of tumor growth, across a 96-well format. Using this approach, we have found that cells that fail to metastasize in in vivo mouse models also fail to reform their spheroid into a branched structure after being transferred to the hydrogel growth surface. The intricate, label-free phenotypes that are manifested by metastatic cells, in combination with recently acquired imaging technologies here at Purdue, allows us to evaluate serval data points in a medium throughput manner. This approach allows us to quickly identify compounds that may not affect cell viability, but could have potential in inhibiting other aspects of the metastatic process. Compounds of interest can be added at any point. (B) Examples of two isogenic cells lines that were sequentially passaged through mice to yield an increasingly more metastatic cell line compared to the non-metastatic parental line. This increase in metastatic potential correlates with a drastic reorganization of the spheroid after several days of culture on the semisolid matrix. (C) Another metastatic cell line forms highly intricate structures after several days on the semisolid support. This phenotype was potently inhibited by a candidate antimetastatic compound. Wendt, Ph.D. Assistant Professor Department of Medicinal Chemistry and Molecular Pharmacology College of Pharmacy ________________________________ 3D in Focus Hydrogels have structural similarity to natural extracellular matrix and are utilized extensively as cell culture platforms for various biomedical applications, such as tissue engineering. The mechanical strength of the extracellular matrix regulates cellular behavior and physiology in a 3D environment while the effect of diffusional properties has yet to be explored. The conventional approach to modulate the mechanical properties of the hydrogel is to control the crosslinking density that in turn also limits the diffusional properties of the hydrogel. To have more accurate analyses for the matrix effects on cells, there is a great need to uncouple mechanical and diffusional properties in 3D hydrogel systems. In the highlighted article, by varying the number of functional groups on the crosslinker molecules, the authors were able to uncouple the mechanical and diffusional properties in a dextran hydrogel system and study the effects of both properties on the viability of cells. Biosci. 2017, 1700162 Our summary of the article: In the study, the base polymer is dextran and the polymeric crosslinker is methacrylic dextran (MDex) of which the functional groups crosslink with the dextran polymer. The extent of polymerization and consequently the mechanical properties of the hydrogel is controlled by the concentration of the crosslinker and the polymeric functional groups on the crosslinker. Hydrogel of different mechanical properties were prepared by varying the number of functional groups on the crosslinker (MDex-DS) or the concentration of the crosslinker (MDex-C). MDex-DS hydrogels were prepared by mixing dextran at 1% and MDex at 7% with the number of functional groups on MDex being between one and five. MDex-C hydrogels were made by mixing dextran at 1% with 4 to 8% of MDex (with a constant number of four functional groups). The Young’s moduli of both MDex-DS and MDex-C hydrogels were in the range of 1.8 kPa (MDex-DS1 or MDex-C4 = 4% MDex) to 63 kPa (MDex-DS5 or MDex-C8 = 8% MDex). The diffusional properties in the hydrogel were assessed by equilibrium swelling ratio and porosity. Both confirm that the diffusional property of MDex-DS2 to DS5 hydrogels was similar while it was reduced in MDex-C hydrogels with increasing MDex concentration. Different cell types may display varying phenotypical behaviors in response to different mechanical environments. In the study, the viability of hepatocarcinoma HepG2 cells, 3T3 fibroblasts, mesenchymal stem D1 cells encapsulated for one day in various hydrogels was measured. HepG2 cells encapsulated in MDex-DS hydrogels (DS2 to DS5) displayed a maximum viability at MDex-DS4, indicating optimal hydrogel rigidity for the cells since the diffusional properties for DS2 to DS5 are similar. However, the viability of HepG2 cells encapsulated in MDex-C hydrogels increased with MDex concentration (4% to 8%). As a result, the difference in cell viability between MDex-C and MDex-DS was the largest for MDex-DS5 and MDex-C8, the pair with highest rigidity, but showing the largest difference in diffusion. The viability of 3T3 and mesenchymal stem cells in both MDex-C and MDex-DS hydrogels was biphasic, with a decrease followed by an increase. The viability of 3T3 cells in hydrogel was minimum at DS3 and C6 and for mesenchymal stem cells it was minimum at DS4 and C7. At low rigidity (MDex-C5 and MDex-DS2 having similar rigidity), the viability of cells in MDex-C5 was higher than that in MDex-DS2. However, at high rigidity (MDex-C8 and MDex-DS5 having similar rigidity), the viability in MDex-C8 hydrogel was lower than that in MDex-DS5 hydrogel. These results suggest that the difference in diffusion between two hydrogel systems might play a role in the viability of encapsulated cells. Comments from 3D3C: Hydrogels are widely used as a cell culture platform for various biomedical applications. The mechanical properties in hydrogel are known to regulate cell function and physiology. It is still a significant challenge to control cellular behavior and to maintain the viability of cells in 3D hydrogel systems. The highlighted article suggests a feasible approach, shown for the first time, to uncouple mechanical property from diffusional property in hydrogels. The results indicate the distinguishable impact of diffusional properties and mechanical properties on cells. However, in the present system, scientists are still unable to change the diffusional properties without changing the mechanical properties of the hydrogel. Nancy Black and Mary Jo Totten will be collecting money for a farewell gift for Mike (please have to them by Monday Sept. 25th). Mark your calendars for Wed. As part of this new initiative, we are holding three data science forums<http://www.purdue.edu/newsroom/purduetoday/releases/2017/Q3/office-of-the-p…> in the coming weeks to discuss current efforts across campus, with the goal of developing a set of strategic approaches that will position the University as a leader in the field. This joint initiative will prepare students to engage in an increasingly data-driven world, empower our faculty to create and shape the data science research ecosystem at Purdue and to identify and engage with strategic partners. Our faculty will be key players in these open discussions, which will shape this new initiative. It is now available for use but because of the sensitive nature of the sensor, all current user access has been removed, pending retraining on the tool. This retraining is necessary due to the nature of the failure mechanism and is offered at no cost. Training will be handled in iLabs on a first come first serve basis. The BNC staff will be performing appropriate preventive maintenance on the process equipment at this time as well. See posted information. This is a very unsafe act for the individual as well as a poor example for other users! Given the myriad of chemicals used potential inhalation exposure may occur. Your work should be done 6 inches from the sash opening and no part of your face or head should ever break the plane of the window sash. Those that continue this may be subject to access restrictions.
