[cid:image005.jpg@01D9C906.3EE5E1E0] BME WEEKLY ROUNDTABLE August 7, 2023 INTERIM HEAD'S NOTE * In the past few weeks, together with Vitaliy and Young, I have been working on the committee assignments. We announced the chairs for the undergraduate and graduate committees in the last weeks. By the end of last week, we have finalized the committees with quite some discussions. As we navigate the ongoing changes and challenges in our School on both sides, the committee members are critical to our School's operations in many fronts. In addition, the committee roles are critical in collaborating with our colleagues in Indianapolis for the development of the joint program. Vitaliy is sending out a personal thank-you note from the two of us. Here, I want to send my sincere appreciation as well. Other committees are also being finalized as we speak. Several chairs have been looped into the discussion. You may hear from us on this matter soon. * On 8/25 (the first Friday of the fall semester), we will have our first faculty meeting. With all the changes (not only administrative and organizational but also incentivization opportunities and performance assessment), I have invited Dr. Wayne Chen, Associate Dean for Research, and Dr. Luna Lu, Associate Dean of Faculty, to touch on several topics in the latter categories. Thanks to the staff and students organizing the Ice Cream Social, which may have been the extra motivation for them to come. They have both told me they want to socialize with our faculty. I have made requests to schedule information sessions in following faculty meetings on various important topics, e.g., COACH survey; faculty recognition. Let me know if there are things you want to hear from various stakeholders in the college and university levels. * BME Summer Seminar Series of this summer is coming to an end this week. I would like to know how it has helped our students. In addition to my priorities (helping identify strategic important areas for the School to grow and assisting in the establishment of the joint program between West Lafayette and Indianapolis), I take special interest on the issue of how to maximize our students' development (both technical and professional competencies). In the past few years, we have made great progress in this regard, and now that new interventions are in place, we have started to get feedback and comments. Important ISS Updates As we enter a new academic year, ISS would like to share the following information with the advising community: Official End of COVID Procedural Adaptations Earlier this summer the Department of Homeland Security announced that COVID-related procedural adaptations for international students would come to an end at the end of the 2023 academic year. All international students must meet minimum full-time enrollment requirements each semester as defined by federal regulation. Minimum enrollment requirements for international students are: * Graduate students must enroll in and complete at least eight credit or research hours each fall and spring semester (unless it is the student's final semester). * Graduate students who hold a graduate staff appointment must enroll in and complete at least six credit or research hours each fall and spring semester (unless it is the student's final semester). Distance Learning No more than the equivalent of one class or a maximum of three credit hours of coursework delivered via distance/online/synch/asynch may be used toward an international student's minimum enrollment requirements. Final Semester Students who are in their final semester MUST be enrolled in all remaining coursework required to complete the degree, they must enroll in at least one class that requires their physical presence on campus for most of their final semester - even when summer is the student's final semester, and the distance learning limitation is still in play. To reduce its heavy burden on the healthcare system, prevention-based public health policies targeting CVD become the focus through promoting healthy behaviors and managing risk factors, as well as reducing health disparities with improved access to care for vulnerable populations. However, with limited data from the long-term large-scale population studies, public health practitioners and policymakers can't assess the cost and effectiveness of policy implementation well. Instead, microsimulation model (MSM) has been employed as a powerful tool to analyze the public health policy. In our project, we adapted the well-established and validated MSM (CVD Policy Model) for tobacco tax policy analysis in New York city with incorporation of 2 key factors: 1) illicit market and 2) secondhand smoke exposure. We simulated each individual's CVD (e.g., stroke and coronary heart disease) trajectory, and estimated the associated health cost reduction and the quality-adjusted life years (QALYs) gained by implementing tax policy to reduce cigarette consumption and secondhand smoke exposure for adults in NYC. In the model, the population socioeconomical and health profiles were sampled from NYC Health and Nutrition Examination Survey dataset and the relative risk of secondhand smoke exposure on CVD were pooled from the recent meta-analyses. Our preliminary results show that the additional tobacco tax can be cost-effective (defined as <$50,000 per QALY) even with substantial illicit market. Benefits from nonsmokers exposed to secondhand smoke shouldn't be ignored in the analysis which contributes over 27% of total incremental QALYs. An estimated 24% of adults in the US have some form of arthritis, including rheumatoid arthritis, osteoarthritis (OA), and posttraumatic osteoarthritis (PTOA), where the joint degenerates more rapidly following an injury. Arthritis has no cure, with pain being the most prominent and persistent symptom. Current management techniques for PTOA rely heavily on pharmaceutical approaches such as anti-inflammatory and painkiller medications, each with their own side effects and complications. Novel effective and durable intervention techniques are necessary. My research explores the structure-function relationship of nerves that supply the murine knee. My primary aim is to identify the morphology and extracellular signaling characteristics of the nerve fibers that transmit pain-related information to the central nervous system (CNS) in a PTOA model. MicroLeads cuff electrodes with 130 µm inner diameters and 0.15 mm spacing between the two platinum plated leads were implanted in the hindlimbs of 8 C57BL/6J mice to record and stimulate the common peroneal nerve, a branch of the sciatic nerve that is responsible for the sensation to portions of the leg and knee joint. Single-ended recordings from multiple cuff electrode leads showed spontaneous and evoked SFAP activity and allowed for the discrimination of afferent (sensory) and efferent (motor) activity. ECAP signals were then collected with bipolar cuff electrodes in response to a variety of stimulus parameters, giving insight into the composition of the nerve and providing a basis for feedback-controlled stimulation targeted to pain-related fibers. Targeted stimulation is key in neural-based interventions as only unmyelinated C fibers and A-delta fibers carry pain signals. These results show the potential for a targeted feedback-controlled system of peripheral nerve activity, which could lead to personalized PTOA management devices that are responsive to individual pain management needs. Abstract: Cerebrospinal Fluid (CSF) movement within the brain ventricles and subarachnoid spaces of the cranium and the spine is critical to the health and function of the central nervous system. Recent studies have highlighted the role of CSF in the pathophysiology of neurodegenerative, and neurodevelopmental disorders and the glymphatic system. CSF movement is also shown to be linked with sleep, with enhanced magnitudes of movement during Non-rapid eye movement (NREM) sleep. Regardless of the prominent role played by CSF in maintaining optimal brain health, a comprehensive understanding of the mechanisms driving CSF movement in humans across wake/sleep states has not been reached. To this end, multimodal imaging studies were designed and implemented with simultaneous acquisition of CSF and brain hemodynamics with functional Magnetic Resonance Imaging, neural activity with Electroencephalography, and non-neuronal systemic physiology with peripheral functional Near-Infrared Spectroscopy. From these studies, we show that the CSF system responds to multiple physiological forces at the same time. Both low-frequency vasomotion and respiration simultaneously interact and regulate CSF movement in humans during wakefulness. We also found that non-neuronal systemic physiology significantly influences CSF movement even when autonomic neural contributions come into play during light NREM sleep. Therefore, interventions may increase CSF movement by manipulating systemic physiology. Finally, we attempt to modulate CSF movement through manipulation of these low frequency cerebrovascular oscillations via simple breathing challenges. Preliminary results demonstrate their potential in modulating CSF movement and may be developed into clinical protocols for boosting CSF movement across pathologies where low flow is a concern. Abstract: The term neurovascular unit (NVU) indicates a brain-specific multi-cellular assembly controlling brain homeostasis for physiological neuronal transmission in networks. The blood-brain barrier (BBB) within the NVU is the microvascular interface that governs the exchanges between the peripheral blood and the brain parenchyma. It regulates or partakes in inflammatory modifications occurring during disease. Within this framework, we examine BBB damage as a pathophysiological entry point that allows diagnostic strategies for brain disorders. We examine human and experimental focal epilepsy as a prototype disease to study NVU dysfunctions. Alves, PhD, within the Department of Emergency Medicine at the Indiana University School of Medicine (IUSM), is seeking a highly motivated and independent post-doctoral research fellow. Dr. Alves is a Chemical and Biomolecular Engineer, Associate Professor and the Director of Translational Research for the Department of Emergency Medicine in addition to having an affiliate faculty appointment at Purdue University in Biomedical Engineering. The Alves Lab explores highly interdisciplinary research interests applying engineering and biophysical principles and designs to create translational technologies and treatments in the area of blood coagulation and fibrinolysis. The primary focus of research in the laboratory is to develop targeted clot digesting therapeutic agents to more safely digest clinically relevant blood clots to treat pulmonary embolism (PE, blood clots in the lungs), deep vein thrombosis (DVT), and ischemic stroke. This interdisciplinary research utilizes diverse enzyme assays, chemical synthesis, blood clot formation and digestion assays under shear, and other novel testing/delivery platforms that include multivalent branched molecules and nanoparticle drug delivery systems. Projects Include: (1) Leveraging multivalency to modulate enzyme activity and improve inhibitor selectivity (nanoparticle drug delivery and targeting). (2) Ex-vivo flowing models of clot digestion under shear using in-vivo like fluorescently labeled blood clots from human whole blood. (3) Development of novel fibrinolytic diagnostics tools to assess coagulation state. (4) Mouse models of clot formation/targeting/digestion utilizing intravital microscopy. (5) Study the effects of environmental microplastic exposure on coagulation/fibrinolysis. Projects are funded both externally and internally.
