Title: Bespoke Computational Tools for Gastrointestinal Medicine Date: Wednesday, November 19, 2025 Time: 1:00 PM Location: DLR 131 and Zoom [invitation below] Committee: Dr. Matthew Ward (Chair), Dr. Kevin Otto, Dr. Ken Yoshida Abstract: The gastrointestinal (GI) tract is a network of organs that food passes through, with mechanical impairments and neurological compromises impacting one's quality of life significantly. Here, I present two distinct works, promoting interdisciplinary research in the GI scope. The first chapter involves understanding the mechanical characteristics of the esophagus in eosinophilic esophagitis (EoE), an inflammatory condition leading to fibrotic remodeling and diffuse esophageal stiffening. I present a MATLAB-based graphical user interface (GUI) that processes raw data from a functional luminal imaging probe (FLIP) to capture esophageal peristaltic motion in the presence of a distending balloon and help identify mechanically impaired regions. With implications extending to many conditions impacting the luminal structures of the GI tract, the information presented may contribute towards personalized dilation treatments. In the second chapter, I select gastroparesis, a neurogastroenterological condition characterized by delayed gastric emptying, as a model disease in which vagal nerve stimulation (VNS) may be used to manage symptoms like nausea and vomiting. Currently, our knowledge of the function of vagal fibers is limited, and the adopted VNS paradigm is open-loop and stimulus-based, resulting in variable treatment outcomes among patients. Contributing towards the development of autonomous, closed-loop VNS control, we offer heuristic models (HAPI) of extracellular, differential recordings of single fiber action potentials (SFAPs) and compound action potentials (CNAPs) for myelinated and unmyelinated fibers to supplement CNAP-guided bioelectronic therapy. Providing an accessible wealth of explainable information to the broader neuroscience community, HAPI may help engage diameter-specific groups of fibers in the vagal nerve.
