For more information, please see the message in the Upcoming Important Dates section. * The Weldon End-of-Year Celebration, featuring the Awards & Scholarship Ceremony will be held this Friday afternoon. Dear faculty, please come out to recognize our students' contributions and celebrate their accomplishments in the past year. At least, I consider this as a de-stress event for our students. * Six Purdue University PhD students from Biomedical Engineering and Industrial Engineering will be participating in the revamped Bottorff Fellows program for the 2023-2024 school year. This includes two months of in-depth clinical immersion over the summer, followed by courses on technology translation and work with an industry or clinical mentor that will help fellows consider best approaches for moving their work from bench to bedside. This yearly program is funded by the Purdue College of Engineering in close collaboration with the Indiana University School of Medicine and named after Leslie Bottorff<https://www.linkedin.com/in/lesliebottorff/>. * Finally, dear faculty, starting this week, I will have one-on-one meetings with you. I look forward to having the chance to chat about various things with you. Important MJIS Alert Starting Monday, May 22nd, Martin Jischke Drive will be CLOSED to all traffic. The university will be performing infrastructure repairs in the area. The street will be closed to traffic for most, if not all of the summer. When the street reopens, bike lanes will replace the on-street parking, although we do not currently have details/confirmation on the entire process yet. Stay tuned for additional details. "Quiet Period" is defined as a time during which courses that conduct or collect an assessment during the final exam period shall refrain from assigning or collecting assessments. Here, "assessments" are defined as activities relating to the course's learning objectives that students turn in for class credit that the course instructor intends to use to judge whether students have met the associated learning objectives. Assessments do not include class participation during normally-scheduled class time. Courses that do not offer an assessment (such as a final exam, quiz) during the final examination period are exempt from following the restrictions on Quiet Period. It is the responsibility of the unit head to ensure that the unit's faculty preserves this regulation thusly. Dr. Satyajit Mayor, National Centre for Biological Science, TIFR, Bangalore, India, will present "The eukaryotic cell membrane: An integrator of mechanical and chemical inputs." Abstract: The surface of a eukaryotic cell interfaces with the external milieu constantly, decoding signals in the form of chemical and mechanical inputs. These cues which are interpreted primarily by membrane receptors which are embedded in the plasma membrane, a fluid lipid bilayer templated by an active cortical actin meshwork. One such membrane receptor, the integrin receptor binds to the extracellular matrix, and receives mechanical signals from the external environment. Chemical cues activate Rho A-dependent signaling cascades in the cell, whereas mechanical inputs activate mechano-transducers via the Talin-Vinculin axis. Remarkably, integrin receptor activation downstream of these two pathways leaves an imprint on this fluid membrane in the form of spatially localized mesoscale liquid-ordered (lo) membrane domains resembling active emulsions. These membrane domains encode information about both the chemical and mechanical nature of the substrate. This is necessary for integrin receptor function, namely cell spreading and migration in complex environments. The active actin-membrane composite thus behaves as a mechano-responsive medium, integrating chemical and physical cues for the regulation of cell physiology. Searle Professor of Pharmaceutical Sciences, Biomedical Engineering and Chemical Engineering at University of Michigan will present "Engineering strategies to modulate the gut microbiome and immune system." Abstract: New approaches are needed to improve immunotherapies with minimal immune-related adverse events. The gut microbiome has recently emerged as the next frontier in drug development; however, it remains unclear how to effectively alter gut microbiota for treating various diseases, including cancer. Here, we present new biomaterial-based strategies for altering the gut microbiome and improving the safety and efficacy of cancer immunotherapy. We are developing new dietary fiber-based biomaterials for in situ modulation of the gut microbiome for augmenting local and systemic immune responses. Oral administration of inulin-gel in tumor-bearing mice in combination with anti-PD-1 immune checkpoint blockade led to robust anti-tumor efficacy while mitigating immune-related adverse events. In our second research thrust, we are developing a new nanoparticle platform for systemic delivery of STING (stimulator of IFN genes) agonists. While local STING activation can convert cold tumor into hot tumor, it has been challenging to develop STING agonists that can treat disseminated cancer due to their toxicity. Here, we will present our data showing that addition of manganese to STING agonists results in robust synergy. Lipid-based nanoparticles containing manganese and STING agonists allows for systemic cancer immunotherapy with potent efficacy, favorable pharmaceutical properties, and acceptable safety profiles in various murine and rabbit tumor models. Our biomaterial-based strategies may offer powerful and convenient approaches to regulate the immune system as potential therapies for cancer and other diseases. Biography: Dr. James Moon is John Gideon Searle Professor in the Department of Pharmaceutical Sciences, Biomedical Engineering, and Chemical Engineering at the University of Michigan, Ann Arbor. His interdisciplinary research program aims to develop novel biomaterials-based strategies to advance fundamental understanding of the immune system, with the ultimate goal of improving patients' lives with effective vaccines and immunotherapies. Dr. Moon has published over 110 research articles, including those published in Nature Materials, Nature Medicine, Nature Nanotechnology, Nature Biomedical Engineering, Nature Reviews Materials, and Science Translational Medicine. He has over 20 pending/approved U.S. and international patent applications, and his novel nano-vaccine delivery technologies have led to 3 new start-up companies (EVOQ Therapeutics, Saros Therapeutics, and Elicio Therapeutics). Dr. Moon is a fellow of BMES and AIBME, and his contributions to the field has been recognized with numerous awards, including NSF CAREER Award. Nan Kong presents "Shortening Emergency Medical Response Time with Unmanned Aerial Vehicle-Ambulance Joint Operations." Abstract: Unmanned aerial vehicles (UAVs) can improve emergency medical service (EMS) logistics by quickly delivering medical interventions with the help of bystanders. We formulate a Markov decision process to jointly optimize the dispatching and redeployment of UAVs and ambulances in real-time. To tackle the curse of dimensionality, we adopt an approximate dynamic programming approach with neural network-based value function approximations. Our approach outperforms existing benchmarks and provides guidance for effectively incorporating UAVs into EMS operations. By highlighting the potential advantages of using UAVs, we hope to encourage their wider adoption in EMS practice. Following the above focused technical talk, I will talk about more recent research developments in my lab, including (1) supervised learning based recommendation module for smart dispatch system; and (2) UAV-bystander teaming at the scene of emergency response. Bio: Nan Kong is Professor and Deputy Head of the Weldon School of Biomedical Engineering at Purdue University. He is also Associate Director for Health Systems at the Purdue's Regenstrief Center for Healthcare Engineering. Committee Members: Doug Brubaker, Ph.D. (Chair), Elizabeth Proctor, Ph.D., Deva Chan, Ph.D., Tamara Kinzer-Ursem, Ph.D., Brendan Lucey, M.D. Abstract: Alzheimer's disease (AD) is the most common form of dementia and is often presented with aggregates of amyloid-β and intracellular tau proteins in the brain, which leads to downstream progressive memory loss and cognitive impairment. Multiple risk factors for AD development include age, genetics, and other pre-existing diseases such as type 2 diabetes (T2D). While up to 81% of people with AD may have T2D or impaired glucose levels, the biological mechanisms by which T2D exacerbates AD pathology are not well-understood. Previous studies shown that orexin, a neuropeptide responsible for appetite, arousal, and wakefulness, to be involved in AD and T2D pathology, which may be a potential link between the two diseases. We propose that protein aggregation in the brain, and metabolic processes regulated by the orexin network and T2D exacerbates AD pathology. To test this idea, we will first identify functional and biological associations of gene expressions via protein interaction networks and gene set enrichment analysis. Second, we will incorporate publicly available mouse, post-mortem human brain tissue, and human cell line transcriptomic data into our computational model to characterize pathways by which proteinopathy, T2D and orexin are predictive of human neuropathological status. Finally, we will quantify neuroinflammatory cytokines and gene expression by performing multiplexing assays and RNA sequencing of primary neuron samples after stimulation with disease-associated metabolites and orexin antagonist. Upon completion of this study, we will have improved our understanding on biological mechanisms in which orexin and T2D exacerbates AD pathology, which may introduce avenues for disease-directed therapeutics. Bowman; Chi Hwan Lee; Jacqueline Linnes Abstract: Electrochemical biosensor arrays been integrated into microfluidic cell culture platforms have measured metabolic markers, proteins, and nucleic acids. There is a gap for electrochemical biosensor lab on a chip for simultaneously monitoring multiple neurotransmitters, such as glutamate and dopamine, while compatible with recombinant fluorescent neurotransmitter and epigenetic biosensors. A microfluidic neuronal cell-culture platform with these capabilities would be a valuable tool for quantifying phenotypes of PD-linked epigenetic change resulting from environmental chemical exposure. To this end, we aim to develop arrays of enzymatic electrochemical sensors for multi-analyte biosensing. We have developed sensors using direct ink writing robotic printing and conventional thin film processing that can simultaneously measure neurotransmitters (glutamate) and metabolic biomarkers (glucose and lactate) together simultaneously in cell culture media. We have also shown the biocompatibility of theses sensors for neuronal cell culture. What remains to do is verify their stability during many weeks exposure to cell culture conditions and to demonstrate glutamate measurement from glutaminergic neurons in a microfluidic chip. The Diabetes Data and Hypothesis Hub (D2H2) is a platform that integrates tools and datasets related to diabetes research into one integrative platform. The Playbook Partnership Workflow Builder (PPWB) is a system that enables novice users to created bioinformatics workflows by exploring a network of distributed but connect microservices. These two new tools will be demonstrated through a variety of use cases with applications that prioritize personalized drugs and targets for kidney disease, diabetes, and pan-cancer. Bio: Dr. Ma'ayan is a Mount Sinai Endowed Professor in Bioinformatics, Professor in the Department of Pharmacological Sciences, Director of the Mount Sinai Center for Bioinformatics, and a faculty member of the Department of Artificial Intelligence and Human Health at the Icahn School of Medicine at Mount Sinai in New York City. The Ma'ayan Laboratory applies computational methods to study the complexity of regulatory networks in mammalian cells. His research team develops software systems to help experimental biologists form novel hypotheses from high-throughput data with a focus on drug and target discovery through machine learning and other statistical methods. Participants Needed for Research Study: (Aditya Shanghavi is a graduate student in Biomedical Engineering at Purdue.) I am currently working on a research project under the direction of my advisor, Dr Anne Sereno. We are inviting you to participate in a brief arm movement study. We are specifically looking for participants aged 45-85 with no history of neurological diseases who are fluent in English and have adequate hearing (with or without amplification) to process verbal instruction and adequate vision to see the computer screen. Participants should be able to perform simple upper arm motions without experiencing major pain or discomfort. We will use non-invasive motion sensors on a wristband to collect arm motion signals during normal and simulated abnormal movements (after viewing short videos) as well as an eye tracker to collect eye motion signals during a prosaccade-antisaccade task. This study's goal is to develop algorithms to detect latencies and amplitudes of movements and to quantify the sensitivity, reliability, and accuracy of the sensors and these algorithms to differentiate normal versus simulated abnormal movements. The study should take no more than 60 minutes and it will be conducted in PSYCH 3125. Fall 2023, 3.0 credit hours Wednesdays 11:30-2:20 in MJIS 1083 (in person attendance only) Prerequisite: BME Graduate Standing Course Description: The unique medical needs of pediatric patients in the context of current and emerging clinical technologies are presented in this novel course. Anatomical, physiological, and pathophysiological underpinnings of diseases in infants and children are studied. Unique aspects of pediatric device design, regulation, and translation, including clinical studies and ethical considerations, are explored. Numerous clinical sub specialties such as cardiology, surgery, nephrology, pulmonology, and gastroenterology are examined in terms of technology adoption and remaining clinical needs. Future directions in device development and emerging trends in patient care are considered throughout.
