[Bmeroundtable-list] BME Weekly Roundtable - Oct. 3, 2023
[cid:image001.jpg@01D9F5FC.7074B240] BME WEEKLY ROUNDTABLE WEEK OF October 2, 2023 INTERIM HEAD'S NOTE * Last Friday, many of us submitted proposals to the Purdue Life and Health Science Summit, https://www.purdue.edu/research/oevprp/news/index.php?view=8555. I was part of a few teams. It is exciting to see BME is at the front and center of this. We have been told that Karen Plaut's team will be reviewing the proposals today (Tuesday, October 3rd). Notifications will be sent out within a few days. The summit will take place on 10/19 from 1 to 5 pm. Even if you are not part of a proposal team or your team is not invited to a presentation at the summit, I think it will still be exciting to attend the summit and see if you can plug yourself in any of the winning teams. * BMES conference is one-week away. Cindy has sent out an email to ask for the attendee's availability at the conference. It is good we all can take on shifts at the Weldon School booth to be eligible to receive travel support from the BME business office. * We had a good faculty meeting last Friday. The discussions were good. For the next two faculty meetings of the semester, I'd like to try something else. After all, faculty meetings are the few opportunities we all get together. We can wisely utilize the time and leverage our collective wisdom on important department issues. More details will come. If you have any suggestions on the format of the faculty meetings, please let me know. * We have received quite a few applications from the department's Marketing and Communication position. I will work with Vitaliy, Young, and Jo to do the first round of screening in the coming weeks. * You got to know Krishna received the 2022 NIH Director's NIA award through prior communications. I just got to know not one, actually two of us received this big award. They are: * Natalia Rodriguez, "Addressing Cervical Cancer Disparities Among People Experiencing Homelessness"<https://nam04.safelinks.protection.outlook.com/?url=https%3A%2F%2Freporter.nih.gov%2Fproject-details%2F10687561&data=05%7C01%7Cbmeroundtable-list%40ecn.purdue.edu%7Cd38b6100bd574754abdb08dbc4351b51%7C4130bd397c53419cb1e58758d6d63f21%7C0%7C0%7C638319504605124250%7CUnknown%7CTWFpbGZsb3d8eyJWIjoiMC4wLjAwMDAiLCJQIjoiV2luMzIiLCJBTiI6Ik1haWwiLCJXVCI6Mn0%3D%7C3000%7C%7C%7C&sdata=99pEOfNanic0rPekpuR5FVIJTD5cJe8OQniY8ijg0%2B8%3D&reserved=0> * Krishna Jayant, "Massively scalable 3D electrophysiology and two-photon imaging in freely-moving animals"<https://nam04.safelinks.protection.outlook.com/?url=https%3A%2F%2Freporter.nih.gov%2Fproject-details%2F10687565&data=05%7C01%7Cbmeroundtable-list%40ecn.purdue.edu%7Cd38b6100bd574754abdb08dbc4351b51%7C4130bd397c53419cb1e58758d6d63f21%7C0%7C0%7C638319504605124250%7CUnknown%7CTWFpbGZsb3d8eyJWIjoiMC4wLjAwMDAiLCJQIjoiV2luMzIiLCJBTiI6Ik1haWwiLCJXVCI6Mn0%3D%7C3000%7C%7C%7C&sdata=8LOsTa%2FKxy5vdcd50m3lhUUKIRy3RQ%2FJPds8V0BNHdM%3D&reserved=0> Congratulations to both of them! Please plan ahead - DO NOT use chemicals in the lab areas that you would normally use in the hoods. Graduate Programs Update Twenty-four BME and IBSC PHD students presented on their research in the Summer 2023 virtual seminar series, and everyone did an excellent job. This year's results for the outstanding presentation were so close, we needed to select two winners of the 2023 Fearnot-Laufman-Greatbatch Award. Each recipient received the highest average overall score and highest average of all survey questions. Congratulations go out to Marvin Zhou for his presentation entitled "Assessing the Cost-Effectiveness of Public Health Policy for Smoking Control through Microsimulation Modeling on Cardiovascular Disease and Cameron Villarreal for his presentation entitled "The Gut Microbiome Modulates the Structure-function Relationship in Musculoskeletal Tissue: More than a gut feeling." Dr. Neal Fearnot established this award in 1992 to recognize the value of presentation skills learned as a student presenter. The skills he learned served him well in his professional career, and therefore he wanted to encourage participation and reward excellence. This award was originally named the Fearnot Prize, until 2010 when Dr. Fearnot received the AAMI Foundation/Laufman-Greatbatch Prize in recognition of his creativity and leadership in the medical device industry. He donated the funds he received from this award toward the newly renamed Fearnot-Laufman-Greatbatch Prize. Heather Liddell, PhD, Assistant Professor of Mechanical Engineering and Environmental and Ecological Engineering at Purdue will present "Should We Lightweight Electric Vehicles? An Environmental Life Cycle Perspective with an Outlook to 2050." Abstract: This seminar will explore and quantify the forward-looking opportunity space for lightweight structural materials in battery electric vehicles. Leveraging a dataset of over 250 model year 2018-2023 battery electric models sold in the United States, I apply a top-down statistical approach to assess the potential environmental benefits of lightweighting from a life cycle perspective. This work considers both the vehicle cycle (materials) and the fuel cycle (electric vehicle charging). I will discuss the impacts of materials selection and the sensitivity of results to vehicle characteristics such as size and drive range. Analytical results show that lightweighting of electric vehicles can contribute meaningfully to an overall strategy to meet economy-wide decarbonization goals, even 8nder rapid electric grid transition scenarios. However, net emissions impacts depend on specific material selections and manufacturing methods and benefits are not universally observed in all lightweighting scenarios. I will put these results in context to discuss the broader implications for environmentally conscious automotive materials development. Bio: Heather Liddell joined Purdue in Fall 2023 as an Assistant Professor with a joint appointment in Mechanical Engineering and Environmental & Ecological Engineering. She studies resource use and environmental impacts in manufacturing, focusing on the identification and development of technical strategies for9 improving the sustainability of the U.S. industrial and transportation sectors. Core to this work is an integration of top-down strategies (e.g., supply chain modeling and sector-wide energy analysis) and bottom-up strategies (e.g., impact assessments at the technology, product or process level). Prior to joining Purdue, Dr. Liddell was a senior scientist and group leader at Energetics, where she worked closely with the U.S. Department of Energy to develop strategies for resource-efficient and decarbonized manufacturing in the United States. Liddell holds BS and MS degrees in mechanical engineering and a PhD in materials science from the University of Rochester. She completed her postdoctoral training at the U.S. Naval Research Laboratory. Wed., Oct. 4: BME Research Seminar, 9:30 a.m., MJIS 1001 or via Zoom. Alex Chubykin, Associate Professor of Biological Sciences and Member of Purdue Institute for Integrative Neuroscience, Purdue Autism Research Center, will present "Familiarity-evoked theta oscillations in the mouse visual cortex." Abstract: Recognizing familiarity and novelty in the environment is critical for animal survival. Surprisingly, very little is known about how animals distinguish familiar from novel stimuli. We have recently discovered persistent stimulus-triggered theta oscillations in the visual cortex of mice, which are specific to the familiarity and spatial frequency features of the stimulus. These oscillations are dependent on the cholinergic muscarinic receptors for both induction and expression. Familiarity-evoked oscillations lead to the decrease in suprathreshold responses to the non-preferred directions of the drifting sinusoidal gratings and to the improved direction selectivity in V1 neurons. We then used optogenetics to directly measure the synaptic strength of the light-triggered EPSCs in V1 neurons in head-fixed mice. We discovered the weakening of the thalamocortical projections coinciding with the strengthening of the intracortical projections in V1 following visual experience and emergence of the familiarity-evoked theta oscillations. Finally, we have discovered theta oscillations in the higher visual areas (HVAs) which are synchronized with the theta oscillations in V1 following visual experience. This synchronization varies between different HVAs and depends on the properties of the visual stimuli. Biography: Alex Chubykin is an Associate Professor at the Department of Biological Sciences, Purdue Institute for Integrative Neuroscience, Purdue Autism Research Center. Dr. Chubykin's research aims to understand how experience shapes neural circuits and affects their function in health and disease. His laboratory uses state-of-the-art techniques, such as in vitro circuit mapping, in vivo electrophysiology, optogenetics, and behavior, to investigate the synaptic and network properties of the visual cortex and other brain regions. Dr. Chubykin has made significant contributions to the understanding of synaptic plasticity and its role in sensory processing, learning, memory, and neurological disorders. Thesis Committee members: Fang Huang, Chair; Alexander A. Chubykin; Daniel M. Suter; Kevin J. Webb Abstract: 4Pi single-molecule localization microscopy (4Pi-SMLM) enables the revealing of ultrastructure by combining interferometric coherent fluorescence imaging and single-molecule localization. Current methods model the interferometric point spread function (PSF) by scanning in-vitro beads on coverslips to infer in-situ 3D location of single-molecule fluorophores in specimens. Although a resolution of sub-20-nm throughout the whole cell has been achieved, imaging tissue specimens was impeded by the heterogeneous aberration and scattering in tissue environment. Here, we introduce 4Pi in-situ PSF retrieval method (4Pi-INSPR), which allows 12-nm-resolution visualization and quantitative analysis through brain tissue with high accuracy. 4Pi-INSPR incorporates sample-induced aberrations, previously unattainable by conventional 4Pi-SMLM. 4Pi-INSPR also minimizes reconstruction artifacts from instrumental imperfections such as objective and imaging-quadrant misalignments. We demonstrate 4Pi-INSPR capabilities by revealing the ultrastructure of dendrites and dendritic spines in 50-μm mouse brain sections and quantifying their morphology changes and nanoscale molecular distribution. Together, 4Pi-INSPR provides potential in examining synaptic functions for brain development. Thurs., Oct. 12: BME PhD Preliminary Exam Announcement for Donghyun Yim (T. Kim, advisor). Advisory Committee: Taeyoon Kim, Chair; Adrian Buganza-Tepole; Daniel M. Suter; Qing Deng Abstract: Directional cell migration is the essential ability of many cells and occurs in the guidance of different types of cues, including substrate stiffness, surface topology, and chemoattractant gradient. In particular, when the external chemotactic stimuli are present, chemoattractant successively activates signaling pathways in the cell, followed by the nucleation and polymerization of actin filaments (F-actins). As a result, F-actin establishment and protrusion are biased at the sites with the maximal concentration of chemotactic cues, leading to migration toward chemoattractant. For the migration, formation of focal adhesions between the cell and an extracellular environment is necessary. Chemotaxis has been studied in various computational studies. However, to our knowledge, the computational model of chemotaxis considering the dynamics of F-actins and focal adhesions still lacks. To address this gap, we introduce a two-dimensional cell migration model comprising nucleus, membrane, F-actins, and underlying substrate. In this model, it is assumed that the elongation rate of an individual F-actin is enhanced by the local chemoattractant level in order to capture the dependence of the F-actin polymerization rate on the chemoattractant concentration. Repulsive forces act between F-actin and the membrane to simulate the membrane protrusion driven by elongating F-actins. F-actins are allowed to bind to substrate via focal adhesions. In preliminary results, it was observed that a cell located in the middle of a linear concentration gradient for chemoattractant protrudes and then migrates toward the region with higher concentration. We expect the current concept of the model will provide better understanding of F-actin-mediated chemotaxis.
