Happy
Halloween! Will you do trick-and-treating tonight? My 12-year-old daughter is so looking forward to it with her friends. She participated in the costume contest last Friday at school. She went in with a group of friends for the group costume award category,
dressing herself up like the 70s. She got a turtle-neck for the contest, for which she particularly went to the Goodwill store in town. I saw what she got and I was like I have a lot of those in my closet. Time flies.

We
have completed the interviews of our Marketing & Communications Specialist position. We interviewed four qualified candidates in the past 2 weeks. We selected them from more than 22 applicants. We initially contacted five finalists for interview. One withdrew
at the last minute. We spent one hour with each of the four candidates. Questions include, not exclusively, 1) what are the strategies to improve the image and reputation of the school facing outward; 2) what are some of the events you organized for building
up the image; 3) what do you see your strengths and weaknesses; and 4) how do you work on a team and managing student helpers, especially under deadline pressure; and 5) how are you familiar with the operations of a department and major events taking place
throughout the year. Jo has sent out a Qualtrics survey to the interviewers to solicit evaluation and feedback. We would like to decide the first person we want to offer the job to by the end of this week. We can then move on to the offering phase. Many thanks
to Jo who is taking us through the finish line. Thanks to Tammy, Darshini, Cindy, Young, and Vitaliy for participating the interviews.
The
faculty should all receive an email from Luna Lu calling for nominations for the College of Engineering Faculty Excellence Awards. Anyone is free to nominate anyone. This is an important opportunity to recognize our colleague’s work and express our appreciation
for their contributions and achievements. While application to most of the awards is due on Feb 1, 2024, it is the time to think carefully and strategically given these awards are quite competitive and preparing materials take time. I am aware that Chi Hwan,
our awards committee chair has been active. I use this as the first prompt. The committee wants to come up with a short list soon and work with the nominators and nominees.
Ascension
Health / Purdue BME workshop will take place today. All BME faculty are welcome to join the workshop at around 2:45 pm. We will start breakout group discussions by then. The two main themes are 1) cardiology and 2) neurology. Two additional themes are digital
health and AI/ML in clinical practice, and clinical administration and health systems management. After the group discussions, we will bring all of us together to identify a few common areas/projects of interest.

Breathing Disorders.”
Abstract:
I will give
a brief overview of my lab group’s principal translational projects. These have to do with several disorders of childhood breathing. First, I will discuss apnea and opioid-induced respiratory depression, addressing how and why ATLX-0199 can reverse drug-induced
respiratory depression. Then, I will discuss Primary Ciliary Dyskinesia (PCD): how we establish a PCD diagnosis in genotype-negative patients; how PCD cells be corrected and grown on an implantable matrix; and how we are improving diagnosis and management
of PCD in Africa. Next, I will discuss viral pneumonia and bronchiolitis, specifically including a promising new potential approach for treating COVID, RSV and metapneumovirus. Next, I will discuss bronchopulmonary dysplasia (BPD), reviewing whether we might
be able to treat we treat BPD and tracheomalacia with GSNO. Finally, I will go over our translational work on asthma. Specifically, whether GSNO can be used to treat asthma, with FeNO as a biomarker; whether Cavosonstat may be able to benefit an
ADH5 genotype-targeted severe asthma population; whether DHEA could benefit and
HSD3B1 genotype-targeted population; whether inhaled Optate could decrease asthmatic airway inflammation, and whether ciliary dysfunction predisposes to asthma.
Biography:
Dr. Gaston
is Vice Chair for Translational Research at Riley Hospital for Children. He went to college and to medical school at the University of Virginia. He did his military service (USS Iwo Jima) and Pediatric training (Bethesda) in the United States Navy. He trained
in Pediatric Pulmonology at Boston Children’s Hospital. He has been continuously funded by the NIH for 25 years, and currently is PI on a P01 to study airway epithelial biology. He also is PI or CoI on several other grants, including the PrecISE asthma intervention
trial and an R33 in continuity with an R44/SBIR focused on developing a novel respiratory stimulant. He and his collaborators have several other small grants, including grants on PCD; and the P01 is flexible enough to help support PCD research. He has over
230 publications. He has helped to found several companies focused on translating discoveries for the benefit of children with respiratory diseases. Currently, most of his clinical work and a good deal of his research is focused on improving the diagnosis
and management of PCD. He and his wife have three wonderful children.

Abstract: The
temporal binding window (TBW) is a period of time where two stimuli presented at different times are perceived as one event. This is a way to measure how the brain integrates different stimuli, and to see if individuals can distinguish the temporal delay between
a pair of stimuli. The use of visual and auditory stimuli has been used more in prior multisensory integration studies, and the results from these studies have been used to compare individuals with typical sensory and cognitive functions with individuals who
have impairments in these domains. These conclusions are built on differences that aren’t supportable without some reliability measurements. Therefore, we aim to measure the consistency of temporal binding windows across different visits using audio-visual
stimuli. Temporal binding window measures were collected in 10 participants (ages 19 – 58) with normal hearing and no history of vestibular disease. Each participant was tested twice at separate visits (at baseline, and on average 7 days later) using a fixed
psychophysical procedure that consisted of subjective responses of either “flash” or “beep” in each trial. Preliminary results using both Generalized Linear Model (GLM) and Bayesian parameters suggest that there may not be consistency across visits at the
group level when using linear regression analysis. Further research and analysis will include a larger sample size to account for variance, Karolinska Sleepiness Scale (KSS) data analysis to see if subjective sleepiness can influence performance, and perhaps
increasing the number of trials per time interval. Overall, more work is needed to investigate the reliability of audio-visual behavioral data.

Abstract:
Machine
learning and artificial intelligence are driving forces in medical research, including radiology. These technologies heavily rely on data, often millions of meticulously curated medical images, for deep neural network development and optimization. Our lab
focuses on innovative ways to maximize existing data for deep algorithm performance. In this seminar, we'll discuss recent studies from our lab. The first part explores using breast laterality features to improve breast cancer detection in mammograms through
the Radon Cumulative Distribution Transform (RCDT), which can highlight subtle bilateral tissue differences in breast mammograms. We will then discuss how simulated mammograms via Generative Adversarial Networks (GANs) could improve the performance of cancer
detection algorithms. I will conclude the seminar with cautions on when to use GANs for simulating mammograms, as this can significantly affect the performance of the detection algorithm.

Abstract:
Congenital heart
disease affects 1 in 100 infants and is the leading cause of infant mortality in the US. Among the most severe forms of congenital heart disease is single ventricle physiology, in which the heart develops with only one functional pumping chamber. These patients
typically undergo three open chest surgeries, culminating in the Fontan procedure at three years of age. Prior work has extensively explored the use of multiscale models, combining hemodynamics with lumped parameter models of single ventricle physiology to
assess surgical methods for all three stages of single ventricle palliation. Here, we present our recent work, which extends traditional models of Fontan hemodynamics to include multiple physical processes and cardiac function. We will discuss our recent progress
towards: 1) growth and remodeling of tissue engineered vascular grafts in the Fontan circulation, including a finite element framework for fluid solid growth simulations, and 2) model-driven design of a bioprinted pulsatile conduit to provide a power source
for Fontan physiology. Recent methodological advances for generation of vascular networks using optimization will also be described. Finally, we discuss progress and challenges of developing whole heart models incorporating machine learning for image segmentation,
fluid mechanics, active contraction, electrophysiology and valves. We will describe open source software and data resources available via the SimVascular project and the Vascular Model Repository.
Bio:
Alison Marsden
is the Douglass M. and Nola Leishman Professor of Cardiovascular Disease in the Departments of Pediatrics, Bioengineering, and, by courtesy, Mechanical Engineering at Stanford University. She is a member of the Institute for Mathematical and Computational
Engineering. From 2007-2015 she was a faculty member in Mechanical and Aerospace Engineering at UCSD. She graduated with a BSE degree in Mechanical Engineering from Princeton University in 1998, and a PhD in Mechanical Engineering from Stanford in 2005. She
was a postdoctoral fellow at Stanford University in Bioengineering from 2005-07. She was the recipient of a Burroughs Wellcome Fund Career Award at the Scientific Interface in 2007, an NSF CAREER award in 2011. She was elected fellow of AIMBE and SIAM in 2018,
the APS DFD in 2020, and BMES in 2021. She is the 2023 recipient of the Van C. Mow medal from the ASME Bioengineering Division. She has published over 160 journal articles and holds leadership roles in the ASME and APS scientific societies. Her research focuses
on the development of numerical methods for cardiovascular biomechanics and application of engineering methods to impact patient care in cardiovascular surgery and congenital heart disease.

Abstract:
In our proposed study, we aim to optimize the design of Tetrahedral DNA Nanomaterials (TDN) to effectively deliver drugs through the Blood-Brain Barrier (BBB). Starting with two classical TDN structures, we alter the design
by changing the size, structure (one or two double-stranded DNA helices on the edges), and attached DNA aptamers for receptor-medicated endocytosis pathway to study the influence of these factors on cellular uptake, stability of TDNs, toxicity, and potential
delivery efficiency. We plan on testing our materials in vitro HEK293, SUM159, mice cortical, and human astrocytes cellular lines, as well as in the human BBB
in vitro model. To study the biodistribution and tissue toxicity, we will perform in vivo tests of the most prominent TDNs designs on the C57BL/6-wt mouse model, harvest the tissues, and run histology analysis with H&E stains. The TDNs concentration
will be measured by measuring implemented into the DNA strands cyanine 3 and 5 dyes fluorescence and the stability will be estimated by Fluorescence Resonance Energy Transfer measurements. If we succeed in our work, we will develop an effective, exceptionally
biocompatible, and stable nanomaterial with great potential applications in drug delivery through BBB, brain imaging, and gene therapy. We expect the main application of this nanomaterial to be in treating and mitigating the symptoms of Alzheimer's and other
neurodegenerative diseases, as the low BBB permeability poses the main challenge in the way of studying and treating these illnesses.

Fellow Opportunity – IUSM Emergency Medicine
The
laboratory of Dr. Nathan J. Alves, PhD, within the Department of Emergency Medicine at the Indiana University School of Medicine (IUSM), is seeking a highly motivated and independent post-doctoral research fellow. Dr. Alves is a Chemical
and Biomolecular Engineer, Associate Professor and the Director of Translational Research for the Department of Emergency Medicine in addition to having an affiliate faculty appointment at Purdue University in Biomedical Engineering. The Alves Lab explores
highly interdisciplinary research interests applying engineering and biophysical principles and designs to create translational technologies and treatments in the area of blood coagulation and fibrinolysis. The primary focus of research in the laboratory is
to develop targeted clot digesting therapeutic agents to more safely digest clinically relevant blood clots to treat pulmonary embolism (PE, blood clots in the lungs), deep vein thrombosis (DVT), and ischemic stroke. This interdisciplinary
research utilizes diverse enzyme assays, chemical synthesis, blood clot formation and digestion assays under shear, and other novel testing/delivery platforms that include multivalent branched molecules and nanoparticle drug delivery systems. Projects Include:
(1) Leveraging multivalency to modulate enzyme activity and improve inhibitor selectivity (nanoparticle drug delivery and targeting). (2) Ex-vivo flowing models of clot digestion under shear using in-vivo like
fluorescently labeled blood clots from human whole blood. (3) Development of novel fibrinolytic diagnostics tools to assess coagulation state. (4) Mouse models of clot formation/targeting/digestion utilizing intravital microscopy. (5) Study the effects of
environmental microplastic exposure on coagulation/fibrinolysis. Projects are funded both externally and internally.
