are in full swing of the Semester an after an eventful past couple of weeks with the Industrial Roundtable, Advisory Board meeting, Art in Research Exhibition, and the George Wodicka Atrium dedication to name a few of our activities, I want to bring your attention
back to us- the students, faculty, and staff of our school. First, we would like to remind everybody that
mutual
respect and inclusion are pillars of our community.
Please remember to show everyone mutual respect and know that the we are stronger because of the diverse population and viewpoints that make up the Weldon School.
We

As
we continue to grow in many ways- with our largest incoming Graduate student class, our largest incoming class of undergraduate students, and more changes ahead- staying true to what it means to be a member of the Weldon School of Biomedical Engineering is
critically important. This past spring, the faculty and staff of the school worked hard on establishing our school Core values- for what we expect now and how we plan to grow together. This is even more important now as we are developing new partnerships
with the IU School of Medicine and we are starting the process to integrate the IUPUI BME program. Please find the core-values attached to this roundtable email. We will have continual follow up as these are communicated across the school in the following
months. Overall, I think our values in Community, Mentorship, Research, and Partnerships provide a strong base upon which to move forward together as a school. Boiler up!

Simplified, Improved, and Automated 4D Cardiac Image Analysis

Abstract:
Cardiovascular disease is the leading cause of death worldwide. Cardiac imaging is an increasingly utilized tool to help with early identification and treatment of disease, however as imaging techniques advance in complexity and application, there is a greater
need for accessibility and feasibility for clinical and research use. For example, 3D and 4D imaging methods arguably provide a greater morphological and quantitative assessment of cardiac fitness, however these methods also introduce complexity to analysis
and interpretation of results. Deep learning for biomedical image segmentation and classification is a promising method that could potentially be used to automate analysis and reduce complexity but requires an informed approach to do so. This proposal builds
on previously developed strengths in our lab in using 4D ultrasound to evaluate mouse models of cardiac disease and 4D cardiac magnetic resonance imaging in the characterization of Duchenne muscular dystrophy-associated cardiomyopathy. Using these frameworks,
we plan to simplify, improve, and automate 4D cardiac image analysis in both experimental and clinical cardiac image datasets.
We hypothesize that these improvements will decrease analysis time of 4D image datasets by more than 90% and that automated image analysis will achieve >95% accuracy for both image segmentation and classification. Achieving these goals will address a
growing need in cardiac imaging to provide accessible techniques and technologies to researchers and healthcare providers with the goal of improving cardiac outcomes for patients worldwide.

Tues., Sept. 20, 4:30-5:30 p.m., LYLE 1017. Sarthak Mangla will speak on “Individualized Hearing Aids.” As a high school student, Sarthak developed a smartphone-based individualized hearing aid along with a remote
hearing aid fitting and diagnostics interface. Join us to learn more about his inventions and their potential to further the auditory field. Sarthak is a first-year undergraduate student majoring in Computer Science. Before coming to Purdue, motivated by his
grandmother’s experience with hearing loss and hearing aids, he developed an inexpensive smartphone-based individualized adaptive hearing aid using deep learning. He also developed fully automated and remote versions of traditional fitting and diagnostic procedures
like Pure Tone Audiometry and Speech Audiometry. He will tell us about his project and work toward individualized hearing healthcare!

Abstract:
Many
chronic diseases have a fibroproliferative component that, if untreated, results in replacement of healthy tissue with fibrotic scar leading to organ dysfunction and ultimate failure. Examples include heart failure, chronic kidney diseases like diabetic nephropathy,
chronic liver diseases like NASH, idiopathic pulmonary fibrosis, Crohn’s disease, and scleroderma. Cancers like pancreatic ductal adenocarcinoma also have a fibrotic component. Imaging methods for tissue fibrosis are in general deficient and if available
are typically only sensitive to advanced disease. Our lab has been developing molecular probes with MRI or PET imaging reporters to address unmet needs across these disease areas. We are using molecular imaging to address unmet needs in early detection of
tissue fibrosis, in noninvasive staging of disease, in measuring disease activity to predict disease progression, and in measuring treatment response. We have also been applying molecular imaging to accelerate therapeutic drug development by measuring drug
target expression and target engagement. Since molecular imaging is additive, we have developed multimodal and multiparametric methods for tissue characterization. This lecture will provide examples of these molecular probes and their applications in different
models of pulmonary, liver, kidney, and heart disease and in early stage clinical trials.
Bio:
Peter Caravan, PhD, is co-director of the Institute for Innovation in Imaging (i3) at Massachusetts General Hospital (MGH) and Professor of Radiology at Harvard Medical School. He leads a multidisciplinary and translational molecular imaging lab focused
on the invention of novel molecular probes and their broad applications in cardiovascular, pulmonary, renal and hepatic diseases, as well as in cancers. His research spans novel chemistry technologies to advanced MRI and PET imaging in animal models to applications
in patient populations. A particular area of focus is imaging in fibroproliferative diseases and the development of new imaging tools for early detection of disease and for measuring disease activity in order to guide treatment. He holds Investigational New
Drug (IND) applications for a fibrin-targeted PET tracer and a collagen-targeted PET tracer that are currently being evaluated in 8 clinical trials. Dr. Caravan has invented a general purpose gadolinium-free MRI contrast agent that has been out licensed and
is currently in clinical development. He has also invented molecular probes specific to fibrogenesis, acidosis, inflammation and thrombosis.
Dr. Caravan received
a PhD in Inorganic Chemistry from the University of British Columbia. Following postdoctoral work at the EPFL (Switzerland), he spent 9 years at Epix Pharmaceuticals developing tissue-specific and responsive MRI contrast agents, one of which, gadofosveset,
was approved by the FDA and the EMA. He co-founded two companies that are working on commercialization of technology from laboratory. Since joining the MGH in 2007, he has been continuously funded by the NIH.

Abstract:
Despite the high demand for paper-based point-of-care diagnostics, the limitation in sensitivity is one of the ongoing challenges that can hamper the lab-to-market translation. In this project, I propose to develop 2 detection systems to improve clinical sensitivity
and analytical sensitivity of two pathogens: SARS-CoV-2 in saliva and HIV in whole blood.
With
the surging number of COVID-19 tests, false-negative results have been reported. Integrating internal amplification control (IAC) into an assay can mitigate false-negative due to technical errors. While extensive review papers on paper-based NAATs for SARS-CoV-2
have been published, only a few publications included IAC in their studies. Incorporating IAC on paper-based NAATs is challenging owing to limited one-pot multiplex detection capability on the paper-based platform. Herein, I will develop a one-pot duplex reverse
transcriptase loop-mediated isothermal amplification assay by using SARS-CoV-2 as a target and an 18s rRNA in human saliva as IAC on a fully integrated paper-based device.
Due
to limited sensitivity, existing paper-based rapid diagnostic tests for HIV screening cannot detect a low quantity of p24, viral capsid proteins, in acute HIV infection (AHI). Here, an antibody-initiated loop-mediated isothermal nucleic acid amplification
(ai-LAMP) will be developed to offer a novel sensitive platform for AHI. In the presence of p24, viral capsid presenting in the virion in larger quantities than viral RNA, two antibody-partial DNA probes will be in close proximity allowing the completed DNA
target to form and trigger the LAMP reaction. The LFIA will be integrated into this design to enable simple result visualization.

Abstract:
Human disease and the corresponding immune response occur in three-dimensional space and time. Many diseases are difficult to study experimentally, either
in vivo or in vitro, due to the complexity of the system. Despite computational models that can address complexity, many do not capture the spatial aspects of disease. Agent-based models are mechanistic, spatiotemporal computational models that
can be integrated with other mathematical models to create multiscale models. Here I describe two agent-based models used to examine spatiotemporal progression and possible treatment strategies for two diseases with low treatment success:
Mycobacterium avium complex (MAC) infection and Alzheimer’s Disease. MAC are biofilm-forming environmental microbes capable of residing in human lung nodules, causing MAC pulmonary disease (MAC-PD). Clinical drug susceptibility tests and treatment outcomes
are poorly correlated, and nodules are complex and difficult to monitor, leading to low MAC cure rates (45-65%). I have developed an informative model of the initial bacterial deposition in the lung airway and the resulting host innate immune response in MAC-PD.
After characterizing this model, I use it to probe the effects of biofilms and resulting macrophage signaling or changes in the immune system, such as those during menopause. By building a better understanding of these system dynamics on the progression of
infection, I can quantify the impacts of immunological or environmental interventions such as upregulating macrophage killing, reducing cytokines, or reducing frequency of bacterial exposure. Alzheimer’s Disease (AD) is the leading cause of dementia, with
no disease-altering pharmacological intervention. Microglia are phagocytotic neuroimmune cells, known to form barriers around plaques. There has been increased interest in leveraging microglia to slow the progression of neurodegeneration by manipulating these
barriers. I developed an agent-based model of microglia barriers at the single plaque level and use knock-out experiments to probe the role of microglial mechanisms and quantify their effects on plaque progression. This increased understanding can be used
for immunotherapy interventions to slow or prevent the progression of AD.

9:30 a.m. in MJIS 1001 and via Zoom. Megan Yi-Ping Ho, PhD, Associate Professor of Biomedical Engineering at The Chinese University of Hong Kong will present “Microfluidics for Single Cell Applications.”
Abstract:
Microfluidics has gathered significant interest for biomedical applications, particularly the analysis of targeted biomolecules from minute amount of samples. This presentation will summarize our efforts on exploiting microfluidics for the manipulation
and analysis of individual cells and subcellular organelles. In particular, we have explored the possibility of modulating hydrodynamic shear stress at microscale to selectively disrupt cellular membrane while maintaining the mitochondrial membrane intact.
We
have also investigated a microfluidics-based method to encapsulate protein payloads into red blood cells by controlling membrane deformation either transiently or extendedly in a microfluidic channel.
This presentation will also highlight our recent development of a novel surfactant based on fluorinated plasmonic gold nanoparticles, rendering a previously unavailable feature of photo-responsiveness in droplet microfluidics. Looking forward, we plan to study
the fundamentals of how individual cells alter their phenotypes in response to environmental and therapeutic stimuli.
These
efforts are expected to find clinical applications in diagnostics, prognostics and treatment evaluation of infectious and chronic diseases, as well as to expand our fundamental understanding towards disease development.
Bio:
Megan Yi-Ping Ho is currently an Associate Professor in the Department of Biomedical Engineering at the Chinese University of Hong Kong. She received her B.S. and M.S. in Power Mechanical Engineering from National Tsing-Hua University,
Taiwan. She received her Ph.D. in Mechanical Engineering from the Johns Hopkins University in USA. After her postdoctoral training with Duke University in USA, she received the Young Elite Researcher Award from the Danish Research Council for Independent Research
and started her independent career in the Interdisciplinary Nanoscience Centre and the Department of Molecular Biology & Genetics at Aarhus University in Denmark. She is a co-founder of two start-up companies situated in Demark, Zymonostics and vPCiR, focusing
on enzyme-based diagnostics. She has published 67 peer-reviewed journal articles, 6 book chapters, 77 conference papers, edited 1 book and holds 2 granted patents. The results that she presented have been recognized internationally by the American Society
of Gene Therapy and Controlled Release Society. Her research team is focused on developing nanosensors and microfluidics as diagnostic tools to expand the capacity of disease detection and treatment evaluation.
