Weldon School of Biomedical Engineering,
Thank you to all the staff, faculty, and students who represented BME at the annual BMES meeting in San Antonio, TX. I thought that Purdue and the Weldon School were very well-represented and I heard many positive comments from other heads about the quality
they are seeing at Purdue University. It was an exciting time for many of our students who went to BMES for the first time for talks, posters, and for design competitions. It was also great to work closely with our colleagues at IUPUI on strategic alignment
as well as have an opportunity to work together representing the overall Purdue and Indianapolis ecosystem in biomedical engineering. This week- we have the opportunity to welcome an international speaker to the BME seminar series- where she will present
on microfluidics research and applications in Biomedical Engineering. Megan Yi-Ping Ho represents the first of two speakers this fall from Hong Kong. The HK quarantine restrictions have been lifted just in time to allow for an in-person visit. With this
visit, and the well-attended BMES meeting, it’s great to be returning to a greater sense of “normal.” Covid-19 is still a concern, and it has not gone away, but we have better tools to fight it and be responsive to changes as needed. Have a great week!

for faculty, staff scientists, and administrators who mentor students and postdoctoral fellows in the biomedical, behavioral, and social sciences. A variety of topics will be covered with a specific focus on communication and shared problem-solving to help
trainees deal with the inevitable setbacks experienced in high knowledge environments. The goal of the Raising a Resilient Scientist series is to promote the mental health and well-being of the academic research community by supporting faculty and administrators
to develop self-management, relationship-management, and mentoring skills. There will be five units in the series and start on November 9th, 2022; each unit will consist of a 75-minute lecture followed by a 1-hour facilitated discussion with peers.

Abstract: Dr.
Havton will present recent developments in non-human primate studies of transcutaneous spinal cord stimulation (TSCS) for functional mapping of autonomic and motor functions in pelvic organs. He will also present a new anatomical tool for the correction of
varied dispersion angles and myelination in light and electron microscopy as well as a new non-binary approach for the analysis of clustering of defined nerve fiber subtypes in the peripheral nervous system. Integration of detailed morphological and physiologic
data will be discussed with implications for ongoing efforts to refine neuromodulation strategies to reverse conditions.
Bio: Dr.
Leif A Havton serves as Professor of Neurology and Neuroscience at the Icahn School of Medicine at Mount Sinai in New York, NY. He also serves as Staff Physician in Neurology and Principal Investigator at the Spinal Cord Injury Research Center at the James
J Peters VA Medical Center in the Bronx, NY. The Havton research laboratory studies neural repair and plasticity after injury to the spinal cord, proximal nerve roots, and peripheral nerves, and the lab has a special interest in the recovery and restoration
of motor and autonomic functions. Dr. Havton and his research team develop translational research models in small and large animals for the development of new therapeutics and refinement of available treatment strategies, including neuromodulation and cell-based
approaches. The laboratory includes an anatomical platform for the development of new analytical tools with expertise in ultrastructural studies.

9:30 a.m. in MJIS 1001 and via Zoom. Megan Yi-Ping Ho, PhD, Associate Professor of Biomedical Engineering at The Chinese University of Hong Kong will present “Microfluidics for Single Cell Applications.”
Abstract:
Microfluidics has gathered significant interest for biomedical applications, particularly the analysis of targeted biomolecules from minute amount of samples. This presentation will summarize our efforts on exploiting microfluidics for the manipulation
and analysis of individual cells and subcellular organelles. In particular, we have explored the possibility of modulating hydrodynamic shear stress at microscale to selectively disrupt cellular membrane while maintaining the mitochondrial membrane intact.
We
have also investigated a microfluidics-based method to encapsulate protein payloads into red blood cells by controlling membrane deformation either transiently or extendedly in a microfluidic channel.
This presentation will also highlight our recent development of a novel surfactant based on fluorinated plasmonic gold nanoparticles, rendering a previously unavailable feature of photo-responsiveness in droplet microfluidics. Looking forward, we plan to study
the fundamentals of how individual cells alter their phenotypes in response to environmental and therapeutic stimuli.
These
efforts are expected to find clinical applications in diagnostics, prognostics and treatment evaluation of infectious and chronic diseases, as well as to expand our fundamental understanding towards disease development.
Bio:
Megan Yi-Ping Ho is currently an Associate Professor in the Department of Biomedical Engineering at the Chinese University of Hong Kong. She received her B.S. and M.S. in Power Mechanical Engineering from National Tsing-Hua University,
Taiwan. She received her Ph.D. in Mechanical Engineering from the Johns Hopkins University in USA. After her postdoctoral training with Duke University in USA, she received the Young Elite Researcher Award from the Danish Research Council for Independent Research
and started her independent career in the Interdisciplinary Nanoscience Centre and the Department of Molecular Biology & Genetics at Aarhus University in Denmark. She is a co-founder of two start-up companies situated in Demark, Zymonostics and vPCiR, focusing
on enzyme-based diagnostics. She has published 67 peer-reviewed journal articles, 6 book chapters, 77 conference papers, edited 1 book and holds 2 granted patents. The results that she presented have been recognized internationally by the American Society
of Gene Therapy and Controlled Release Society. Her research team is focused on developing nanosensors and microfluidics as diagnostic tools to expand the capacity of disease detection and treatment evaluation.

Abstract:
Metastases are responsible for approximately 90% of all cancer-related deaths, with metastatic breast cancer (BC) holding a 5-year survival rate of only 27%. Recent research has highlighted a complex dynamic between cancer cells and the tumor microenvironment
as essential for the formation of macrometastases. Within this field, tissue stiffening through matrix accumulation and altered matrix organization at the primary tumor site were recently linked with sustained proliferation and increased migration of tumor
cells. Separately, elevated levels of the glycoprotein, fibronectin, were correlated to poor patient survival in BC and were linked to enhanced seeding of disseminated tumor cells at metastatic sites. Through my thesis work, we have identified several mechanisms
through which accumulated fibronectin impacts the metastatic potential of BC cells. First, we identified a transient increase in extracellular fibronectin in the lungs, which peaked before overt metastasis, coupled with a non-transient increase in total lung
volume. To better recapitulate physiological conditions, we then developed a novel magnetically-actuating platform with the ability to apply tensile strain on cells at various amplitudes and frequencies in a high-throughput multi-well culture plate using suspended
fibrillar fibronectin for 3D cell culture that is not reliant on a synthetic substrate. Using this as a biomimetic lung model, we found that cyclic mechanical force acted as a suppressor of cancer cell growth in a biomimetic lung model, implicating the accumulation
and reorganization of extracellular matrix as an attempt by the cancer cells to alter the mechanical properties of the lung tissue and resist entering dormancy. However, our results showed that BC cells could not organize extracellular fibronectin independently.
Instead, BC cells altered the accumulation and architecture of fibronectin by conditioning fibroblasts through soluble factors and extracellular vesicles. We observed that the fibronectin produced by conditioned fibroblasts varied as an effect of both the
method of conditioning and the phenotype of the BC cell as the conditioning source. Taken together, these results have increased our knowledge of the relationship between disseminated breast cancer cells, fibroblasts, and fibronectin architecture in the early
metastatic lung niche that paves the way for further investigation on targeting disseminated BC cells during early disease intervention in order to inhibit later overt metastatic outgrowth.

Abstract:
Envelope or amplitude modulation (AM) cues in a signal are critically important for the perception of complex signals, e.g., speech. Neural coding of AM can be noninvasively probed using the envelope following response (EFR), which receives contributions from
both cortical (slower fluctuations, <40 Hz) and subcortical (faster fluctuations, >100 Hz) generators. As subcortical versus cortical signatures of AM representations vary across hearing loss etiologies, the EFR has great diagnostic potential. AM representations
are routinely evaluated by the temporal modulation transfer function (tMTF), which is the strength of the EFR as a function of AM frequency. Currently, tMTF is measured serially for discrete sinusoidally amplitude modulated (sAM) tones. This process is time-consuming
and inefficient, impeding clinical translation. Here we present a dynamically varying AM tone (dAM) used to measure a tMTF. I will analyze the tMTF obtained with and without hearing pathologies (aging, inflammation). Methods: EEG responses were obtained using
sub-dermal needle electrodes in rodent models. In rodents, carrier frequencies and amplitude modulation rates varied based on species-specific differences. Tones with dynamically varying AM, the frequency of which increased exponentially from 9 Hz to 1.5
kHz over one second, and identical carrier frequencies were also used to elicit EFRs and the tMTFs were compared. Fast Fourier transforms were used to calculate discrete EFR amplitudes. A spectrally specific frequency-demodulation-based analysis was used to
calculate EFR amplitudes from dAM stimuli. Results: Robust tMTFs were obtained for discrete sAM stimuli from all species tested. Preliminary results show strong tracking of dAM envelopes in rodents, which can be used to estimate the tMTF at a fine frequency
resolution. These tMTFs are comparable to those obtained using discrete sAM tones. In humans, preliminary results suggest that tracking of dAM envelopes was comparable to sAM stimuli, but only at lower modulation frequencies. Ongoing analysis is aimed at refining
the dAM stimuli (trajectories, timescales) to optimize AM tracking while simultaneously reducing recording times in humans. Conclusions: These results suggest that dynamically varying AM tones can be used to efficiently estimate the tMTF. However, further
optimization is needed to obtain robust tMTF estimates in humans at higher AM frequencies. When combined with spectrally specific analysis, the dAM tone can be used to substantially speed up the tMTF measuring time, paving the way for potential clinical translation.

Abstract:
The progress of biomedical sciences depends on the availability of advanced instrumentation and imaging tools capable of attaining the state of biological systems in vivo without using exogenous markers. Mechanical forces and local elasticity play a central
role in understanding physical interactions in all living systems. We demonstrate a novel way to image microscopic viscoelastic properties of biological systems using Brillouin microspectroscopy [1]. In my talk, I will discuss the ways how an old spectroscopic
tool can be used for real time microscopic imaging [2-3] and provide possible solutions to long standing problems in Life Sciences and Medicine [4-6] while advancing instrumentation beyond classical limits [7].

Mentoring
and the Dissertation Process for faculty and postdocs,
Tues., Nov. 1, 2:00-3:30 p.m. Among the most consequential documents that writers produce in graduate school is the dissertation. Strong mentoring at specific critical junctures in the dissertation process can strengthen
both the document itself and graduate students’ abilities to later support their own graduate students. Unfortunately, most faculty receive little to no training about the writing instruction central to the dissertation process, from invention to final deposit.
Instead, they rely upon their own experiences producing a doctoral thesis, which might have been problematic and trauma producing or wonderful yet still mystifying. This workshop incorporates current research on graduate student writing and builds on insights
about graduate student struggles that the Writing Lab gains through working with dissertation writers. Faculty will be offered actionable strategies for helping dissertation writers be more effective. The workshop also will offer faculty mentors guidance on
when and how to direct their students to the Writing Lab for intensive and on-going support. This workshop is for faculty and postdocs only.
